How to Filter Buffers (Vacuum Filtration)

  • In this video, Dan shows you how to properly assemble and use the vacuum filtration apparatus.
    Sometimes you need to filter your buffers or other solutions before you use them. If you are using any automated liquid chromatography systems (such as AKTA or FPLC systems), then you need to filter your buffers before loading them on the columns.
    Remember:
    TO START: Attach tubing THEN turn on tap.
    TO END: Remove tubing THEN turn off tap.
    *If you have detergent in your buffer, bubbles will form during filtration -- make sure to continuously monitor and break the vacuum before the bubbles rise up to the level of the tubing.

    The end of the tube of the funnel should be below the port for the vacuum.

    Hi. Can we use any side of the filter when placed on the base as I noticed that filter both sides differ.

    I had tried to filter the buffer through the same glass assembly, but I was facing a leakage problem from the filter base and funnel. I had tried so much to fix it but couldn't do so. Is there any technique to assemble the funnel and base with a clamp? Can you please help me out?

    Good video. I did this but the filtering was very very slow. it took hours to filter just 10ml solution. Any help?

      May I know if what's the pore size of your filter paper? I encountered similar issue during my vacuum filtration process and pore size affects the speed of filtration and the amount of filtrate collected.

      Hi! I am wondering the same thing and was curious if you figured it out- I am wondering how exactly to clean the white part on the base/stopper? Seems like that could lead to contamination unless using the same buffer

    You have a very good video. 2 things that would set you apart from others...1. list the glassware and tell where we can purchase. 2. Answer peoples questions. Those who ignore simple questions are only in it for themselves and I will not give any likes or watch any more of a persons work if they are so selfish.
