It's funny how it's theoretically possible to unlock every single character in your first run via the R Key except for Lilith, Keeper and The Forgotten because the former two are Greed unlocks and the latter would require getting an R Key, beating the Lamb, resetting and beating the boss of Basement 1 in under one minute.
@@SrJrXVIII As described in the video, using a combination of We Need To Go Deeper!, Clicker and lucky secret rooms (with literally every single one containing R Key) you could theoretically go infinite and unlock all non Lilith and Forgotten characters including the tainted ones.
Okay so if you get a sac room right next to your starting room and somehow either have enough health because of chests it can spawn or the character you picked basically means you can get to the dark room. Now if you somehow get great items in the dark room or got let's say a godhead or sacred heart in the sac room you might be set for mega satan? So basically 3 rooms you have to enter/1 room actually completed(two if you count spawning the angels in the sac room).
As long as you have schoolbag and we need to go deeper, you can just continue doing victory laps untill you find an R key since R key removes the victory run modifier and resets it to a normal run once again, and you can just keep on doing it how ever many times you want to.
actually this is a killer idea, just gotta be careful about losing certain items through re running, but remember you also become the Lost after a few which means you can’t go to dark room, meaning it’ll just end at moms/moms heart/ satan
A nice speculation video would be “how many runs do you need to complete everything” (perhaps excluding things like butter and the daily run shenanigans)
From what I know it is exacly 2 with the echo chamber + ancient recall + ? card + R key gamebreak, one run on hardmode and one on greedier mode. This doesn't include unlocking for example echo chamber first and challenges. Just theoryticly.

So to reiterate: -be good and get EVE and SAMSON -be lucky and get CAIN, LAZ, MAGGIE, and AZAZEL -Unlock and find Schoolbag, find WeNeedToGoDeeper before Mom and R Key before Lamb -skip to Mom's Heart to unlock EDEN -skip again to Satan to unlock JUDAS -sacrifice or error room to The Lamb and use R Key after killing him -find the Clicker and switch to Laz before fighting Mom's Heart again to unlock BETHANY -find and use Missing Poster to unlock LOST, and clicker back to someone else before sacrificing to Dark Room a second time. -Defeat Mega Satan to unlock APOLLYON In theory, after switching off of Lost, you could technically toss the Clicker now in favour of potentially infinite R Keys, which would allow the following before ending off with Apollyon: -Defeating Mom's Heart 8 additional times, unlocking BLUE BABY -Unlocking and defeating Hush 3 times then defeating The Witness to unlock JACOB -I don't remember if breaking progression allows you to continue after fighting Mom, but if it does, you can now take the steps to unlock FORGOTTEN as well. -Likewise, after unlocking Jacob, and if Forgotten is possible, you can ditch WeNeedToGoDeeper to find Clicker again and go to Home to eventually unlock all the TAINTED characters of ones you have and R Keying before the Dogma fights, before ending it off with Apollyon, as he's the only character unlock that you can't guarantee a continue off of. So implying breaking progression skips the Mom epilogue, Hush doesn't proc upon unlock (I don't remember), and you are able to continue after unlocking Apollyon, you would need 28 R Keys to unlock 28 characters in a "single" run. Yes. Thanks BB and Jacob for padding out the runtime.
Ideas: ? Card + R Key allow 3 runs, making 3 Hush kills possible which opens the Antibirth floors. If Schoolbag can hold 2 of the same item, R Key could be Diplo’d, potentially allowing a 4th run where you can actually access those floors? Otherwise, finding another ? Card (in the second run) or another R Key (in the second or third) would do the same thing. If you can get a 5th run through ridiculous luck or another exploit, going through Ascent unlocks the Red Key. It isn’t *likely* you’ll get the Red Key, but it is in a fair few pools including the secret room, which I believe means it could be farmed for, using the infinite Steam Sale trick. If you can get the Red Key and a charging item or two, you can open extra rooms on any floor. This can include Sacrifice rooms, which if used in the Dark Room, teleport you to a *new* Dark Room. You could theoretically chain this endlessly, with 50+ chances each floor. Potentially endless rooms mean potentially endless chances for a 6th run, at which point your biggest concern is probably crashing before you can break the game enough to unlock literally every character possible in the first run.
Theoretically, if you get enough r keys and forget me nows you could unlock 30 characters, only missing keeper and Lilith which are from greed mode. Although there are a finite number of seeds so its unlikely that its even possible
Tainted characters are tricky, as you can't acquire polaroid or negative. I am not sure, maybe dad's key work thought. Also, I wonder if alt mom counts as the first mom kill and would cause problems for Jacob and Esau unlock
Could you theoretically do forgotten or would that negate the lost unlock, because after doing boss rush the broken shovel acts live we need to go deeper so you could get to the dark room still, I guess it’s a throw up on those 2
This is insane I’m gonna go for it, I have about 1100+ hours on flash Issac and rebirth+ combined and I think this is actually viable, when I first started playing flash Issac I accidentally skipped the mom fight because I got we need To go deeper and didn’t know what it did, and I actually haven’t unlocked the womb at the time so I actually did one of the things talked about in this video without knowing so it’s definitely possible! (Also in that run I died cause I was a noob at the time and didn’t have the best items for it😅)
Good video. Before Repentance and maybe some updates (6 years ago), i managed to beat all last bosses (including blue baby), and beat Boss Rush. I always using a clean save slot and in different runs, in random seeds an with no mods/console/challenges beated. But to access some place i use some tricks (like closing the game from the windows task manager or exiting to the main menu in the exact milisecond). I have videos of all in youtube in spanish (i no monetize my videos). I don't know how compatible thoses tricks are with Repentance (probably it's outdate now days). There 2 most dificult places to access are Boss Rush and The Chest. For the Boss Rush you have to exit to the main menu in the exact moment after beat mom and under 20 min and before credits appears. Continuing the game you will be in mom's room and you can enter in the Boss Rush and non exit down. For the Chest you need reach The Cathedral having the teleport 2.0. This object in old days you could find it in the secret room or va chaos. ´ I create a route to depend less or randomness. Provably is outdate at so many levels nowdays. My route was: - No retries. Always delete de save slot before start a run. - No make devil deals nor black market trades. It's ruin the angel room appareance. - No take pills randomly in the critical floors. Because telepills can ruin the run if brings you to the wrong place (mom's fight). - Try to obtain The battery, the habit are usefull or any other item that increases the chance of battery drops (for the teleport 2.0 after clear all the floor). - Try to obtain the telepor 2.0 (secret room) or the shovel and bring it to mom's floor. - Try to obtain the telepor 2.0 or the shovel in mom's floor simultaneously (no school bag needed). - Don't beat mom. Don't even enter to the fight (there is no point in it). - Exit the floor with the teleport 2.0 usin the shovel to create an exit (devil room exit is and alternative, but random to reach). - In the Womb II we need and angel room, because it contains an access to The Cathedral. Sacrificial room are welcome to increase the chance of angel room or to access it directly. - In The Cathedral we need to enter to the I am error room. It's contains the acces to The Chest. - Exit to the Chest vía I am error room exit. - Kill the smurf.

Well you can also unlock the Forgotten either by speedrunning Basement 1 immediately or after R Key, which is more easy, getting you one extra character.
no, you have to beat the lamb for Broken shovel, and R key leaves the timer as is. you'd need spindown wheel to reduce an item to broken shovel, and it's unclear if that skips story items.
This is a way I thought up to get as many characters as possible in one run (not your first run) without R keys. You have to choose to either unlock Eden or ??? as both can't be gotten in the same run for obvious reasons. Jacob/Esau will not be unlocked because that would require beating hush three times, which would require beating Mom's heart ten times first, so both Eden and ??? would not be obtainable. plus there are 3 other characters who are only available in the womb path, so it would be a net negative. Keeper and Lillith are not gotten because they are in greed mode. tainted characters are not gotten because they have the same problems as Jacob/Esau. If you use the sacrifice room to get to the dark room and not the black market or error room, you don't need a get-out-of-jail-free card or Dad's key unlocked as you'll be forced to kill Uriel and Gabriel which will net you the key pieces. prerequisites: get the mom ending get the lamb ending without beating satan (and optionally mom's heart if you want Eden instead) using sacrifice room or a black market in Sheol. (optional)at some point after beating the lamb, assemble the golden key to unlock Dad's key (optional)If you are getting ???, then get mom's heart endings 1-9 (you can technically do one mom kill in the same run you get the lamb ending to decrease the number of runs by 1.) (optional)complete challenge #22, SPEED! to unlock the Get-out-of-jail-free card do all of this without getting any of the other characters Actions in run: play as Isaac (or optionally Eden if you went for ??? for a chance to start with the backpack, clicker, or a chance to instantly unlock some of the characters. (preferably without getting hit or picking up hearts as well) do the next floor without taking damage or picking up hearts (and the one after that if you failed the first floor) to unlock Samson and Eve. pick up four soul hearts to unlock Lazarus. Use the clicker at any point after this until you become Lazarus. If you manage this early in the run and find Dad's key, you can swap it out for the clicker if you don't have a get-out-of-jail-free card to open the Mega satan door. acquire seven heart containers to unlock Magdalene. take three deals with the devil to unlock Azazel. hold 55 coins at once to unlock Cain. do the boss rush after mom to acquire the other half of the shovel. Beat mom's heart to unlock Bethany and Eden/???. die with the missing poster in a sacrifice room at any point afterward to unlock the lost Use Mom's shovel to go to Sheol. at some point in Sheol, you must either find a crawlspace with a black market or a sacrifice room and enough health to use it 12 times and live. kill satan to unlock Judas. Exit the boss room and go to the dark room using the methods above or, alternatively, finding some way into an I AM ERROR room. open the mega satan door using the Golden Key, Dad's key or a Get out of jail free card but don't enter it yet. find an unmarked grave and use Mom's shovel to dig it up to unlock the Forgotten. Go back to the mega satan door, and kill mega satan to unlock Apollyon and end the run. If you have done everything correctly, you should have unlocked Lazarus, Azazel, Magdalene, Cain, Samson, Eve, Eden or ???, Bethany, Judas, the Lost, the Forgotten, and Apollyon on between your 3rd and 13th run. and all without the R key. You can then get Lillith and Keeper on the next run using Azazel, deep pockets, an infinite money trick, and some greed machine luck. and then all you'll have left is Jacob and the Tainted characters. make sure to like this stupid post that took like an hour to write.
Very good video but ffs use the video :D I was just watching a run with you talking about the unlockable characters and how to squeeze them in; With visualisation, this would be a masterpiece. Just showing every character you talk about and some challenges about the unlocks in a list. I would have appreciated a systematic approach, at least visualized, take us to an adventure: Talk about all the characters, how to unlock them, which one can you unlock without problems, which need specific unlocks/floors? how do you get there? What compromise do you do? You could casually point out near impossible ones like Blue baby or Apollyon and say why that would be impossible and just construct a meh run and come back and point out why they wouldn't be impossible to unlock in you're very first run. Keep up the work, stil.
Could you even unlock the forgotten in the first run? I'm not sure the broken shovel spawns before you unlock the negative, and R key doesn't reset run timer, so BB and forgotten are not possible in the first run as far as I can see
I think the idea might breakdown about halfway through. Because you need the Schoolbag to hold both We Need To Go Deeper and R Key, but the Schoolbag is only unlocked by visiting 6 shops in a single run. Because Silver Dollar and Forget Me Now need to be unlocked with Cain and ??? you will need a 5 Pip Dice Room to do one of the floors twice so that School Bag could appear in the shop in Depths 2, and only Depths 2. Unless beating Mom during the first half of the run lets you proceed past her fight after using the R Key, you'd need to abandon We Need To Go Deeper to take the R Key and since the R Key doesn't reset item pools you wouldn't be able to re-find it after using R Key.
Jacob and esau would be really hard even with infinite r keys because you need to unlock hush, beat hush 3 times, then finally go alt path to beat mother.
I haven't watched the video so I'm assuming -magdelene -azazel -eden (use shovel to evict mom fight and get straight to womb) -cain -keeper (not sure about this one) Because you need to enter the chest to unlock eden you can't get any further, that means you can't get judas and eden on the same run Edit: this list is without using r key
You can unlock quite a thew using R-key but i know you cant unlock lilith and keeper depending who you play you cant unlock bethany however mega satan might end your run so i would say unlock apollyon last then go defeat mother with more r-keys unlock the tainted characters and then you can try obtain clicker to unlock more tainted characters and then that all i got

I belive you need 100 death to unlock Scissors so 100 + every compliton mark for every charecter(maybe little less if you could manage more then one for run) is optimal number
You don't need we need to go deeper to get to the lamb first you beat mom, after going to the title screen, continue the game, which will unlock the heart then you beat the heart, and you can get a devil deal which will get you to sheol in sheol there is a chance that a room with a trapdoor to go to the dark room spawns, kill satan and go to the trapdoor you found then kill the lamb and do the same trick we did with mom to get unlock mega satan's door.
Can't you also try to go for the forgotten in that run? If you don't have all runes unlocked you can get the runeshard and hope it's ehwaz (or any other method stated in the vid), making your spacebar free for mom's shovel.
Similar sort of challenge: Can you unlock a tainted character before unlocking their non-tainted counterpart? You'd have to be playing as a character you haven't unlocked yet, and reach home. Obviously, you can't start the run with the character. Clicker can only change you to characters you have unlocked. Lazarus's rags are a lazarus unlock, so you can't get them without unlocking lazarus Same with Judas's Shadow Missing Poster can only turn you into the lost after you unlock the lost. The Ankh and Broken Ankh do actually work to turn you into ??? without unlocking them first, and aren't tied to unlocking ???, but to unlock home, you must beat mother by going down the alt paths, and the alt paths are unlocked by beating hush three times, and the blue womb is unlocked at the exact same time ??? is unlocked anyway. I think there's only two possible ways to do it: #1: "Lostify" yourself in whatever method may work (tainted jacob getting hit by tainted esau, the white fires in the downpour, ect., but i doubt this would work) #2: Unlock TMTRAINER, and hope you get a lazarus's rags / judas's shadow effect without having unlocked those items or characters yet
#1 won't work because you don't technically change characters. Notice you still keep whatever abilities your character has (like Azazel's mini Brimstone or your Tainted shtick) after going in the white fires, and Tainted Jacob still gets Tainted Jacob completion marks after touching Esau. #2 might just work, not 100% sure if it can get effects you haven't unlocked though.
if you are patient enough you can farm R keys using spindown dice and the schoolbag, this way, in theory you coud unlock also blue baby,the problem lies in unlocking spindown dice
you only cannot get keeper, the forgotten, lilith, or bethany because they all need you to do a new run (except the forgotten since he needs to go to boss rush but he cant)
I'm pretty sure you could technically unlock almost everything in "One" run with tainted Cain because you can use clicker to change your character and R key to continue the run and mama mega to open hush and boss rush, you can also use School bag and butter to keep the bag of crafting Only problem is that mother, the beast, delirium, and mega Satan all give you the cutscene right after killing the boss
If you're super lucky and unlock the alt floors during the run and manage to get a ? card and R key, you can use the ? card to restart so you can go fight Mother and unlock Jacob and Esau, then use the R key to go fight Mega Satan. 13 characters
The alt floors require you to kill hush 3 times and that requires you to kill mom's heart 10 times so you would need to be super lucky and be able to do all of that in under 30 minutes.
Unlocking every character (reasonably) actually only has 4 conditions. 1. Find the moving box 2. Find the R key 3. Find the school bag 4. Find Diplopia or crooked penny on the first and every R key'd run (or find the R key again) Thought goes like this: Get moving box Find diplopia, crooked penny, or R key Put either in your box Find the other Use diplopia or crooked penny Find (or have) schoolbag Pick up one and put the other in the box Repeat until all characters (- keepy boy) are unlocked There is also a super shortcut that would be ridiculously rare, but sick af. Any run with: - Crooked penny or double diplopia - R Key - Schoolbag - Moving box Is guaranteed to be able to unlock every character (- keepy boy) 25% of the time with crooked. Adding a single Diplopia makes it around 50%. Double diplopia is almost guaranteed. Note: Yes, the previous calc hinges on crooked penny being unlocked to get there. This means that you'd have to replace it with double diplopia OR get one every reset until you can unlock the forgotten, beat boss rush, and find the crooked penny.
until you realize you cant get past the mom fight without a cutscene, meaning you can't ever open flesh door, so no tainted, and moving box is not a base unlock, you need to go to chest for it, which you cant do yet
there is a way to get extra r key uses, using ? card it was changed in rebirth to use single-use items again, and now, as long as you have some way to gain infinite cards, you have a way to reset your run infinitely
it is still impossible to actually unlock any tainted characters, as unlocking them requires that you get the polaroid or the negative, and those require you to defeat mom. however, you can unlock jacob and esau with his.

Is there some way to golden teleporter + backpack (broken shovel) your way into boss rush from the mom fight? If that's the case you can unlock the forgotten in the dark room too, yeah?
@@bd1pLP yeah if you had backpack and golden teleporter/shovel piece one if would work. with piece 2 the shovel would function the same as "we need to go deeper" but if you cant tp in, that's where it ends. ive never explored this, but are there any ways to get into boss rush without killing mom?
@@bd1pLP I'm saying you start off the very beginning of the run by beating the first floor in 1 minute and getting the shovel to drop, as isaac spawns with a bomb. You can then crank through it; however, if you cant go into bossrush it doesn't matter anyway
A more reasonable amount of runs video would be interesting and fun, and the every item from zero guide also sounds interesting in how you'll go about it. I mean obviously the first thing you want is to get every non-unlockable item from the start, then get the unlocks and such. And I guess the best way to get them would be to just get glowing hourglass, a two of diamonds and chaos and break the game in greed or greedier. Sounds hard to pull off but maybe you can think of an even faster way to get them.
Ok but like, add tainted Cain into the mix who can make infinite R keys (i got 11/12 unlocks that way in 1 run) and theoretically you can unlock all other tainted characters with clicker to make it 16

In theory, after switching off of Lost, you could technically toss the Clicker now in favour of potentially infinite R Keys, which would allow the following before ending off with Apollyon:
-Defeating Mom's Heart 8 additional times, unlocking BLUE BABY
-Unlocking and defeating Hush 3 times then defeating The Witness to unlock JACOB
-I don't remember if breaking progression allows you to continue after fighting Mom, but if it does, you can now take the steps to unlock FORGOTTEN as well.
-Likewise, after unlocking Jacob, and if Forgotten is possible, you can ditch WeNeedToGoDeeper to find Clicker again and go to Home to eventually unlock all the TAINTED characters of ones you have and R Keying before the Dogma fights, before ending it off with Apollyon, as he's the only character unlock that you can't guarantee a continue off of.
So implying breaking progression skips the Mom epilogue, Hush doesn't proc upon unlock (I don't remember), and you are able to continue after unlocking Apollyon,
you would need 28 R Keys to unlock 28 characters in a "single" run.

Ideas:
? Card + R Key allow 3 runs, making 3 Hush kills possible which opens the Antibirth floors. If Schoolbag can hold 2 of the same item, R Key could be Diplo’d, potentially allowing a 4th run where you can actually access those floors? Otherwise, finding another ? Card (in the second run) or another R Key (in the second or third) would do the same thing.
If you can get a 5th run through ridiculous luck or another exploit, going through Ascent unlocks the Red Key.
It isn’t *likely* you’ll get the Red Key, but it is in a fair few pools including the secret room, which I believe means it could be farmed for, using the infinite Steam Sale trick.
If you can get the Red Key and a charging item or two, you can open extra rooms on any floor. This can include Sacrifice rooms, which if used in the Dark Room, teleport you to a *new* Dark Room. You could theoretically chain this endlessly, with 50+ chances each floor.
Potentially endless rooms mean potentially endless chances for a 6th run, at which point your biggest concern is probably crashing before you can break the game enough to unlock literally every character possible in the first run.

Theoretically, if you get enough r keys and forget me nows you could unlock 30 characters, only missing keeper and Lilith which are from greed mode. Although there are a finite number of seeds so its unlikely that its even possible

Tainted characters are tricky, as you can't acquire polaroid or negative. I am not sure, maybe dad's key work thought. Also, I wonder if alt mom counts as the first mom kill and would cause problems for Jacob and Esau unlock

This is insane I’m gonna go for it, I have about 1100+ hours on flash Issac and rebirth+ combined and I think this is actually viable, when I first started playing flash Issac I accidentally skipped the mom fight because I got we need To go deeper and didn’t know what it did, and I actually haven’t unlocked the womb at the time so I actually did one of the things talked about in this video without knowing so it’s definitely possible! (Also in that run I died cause I was a noob at the time and didn’t have the best items for it😅)

Before Repentance and maybe some updates (6 years ago), i managed to beat all last bosses (including blue baby), and beat Boss Rush.
I always using a clean save slot and in different runs, in random seeds an with no mods/console/challenges beated.
But to access some place i use some tricks (like closing the game from the windows task manager or exiting to the main menu in the exact milisecond).
I have videos of all in youtube in spanish (i no monetize my videos).
I don't know how compatible thoses tricks are with Repentance (probably it's outdate now days).
There 2 most dificult places to access are Boss Rush and The Chest.
For the Boss Rush you have to exit to the main menu in the exact moment after beat mom and under 20 min and before credits appears.
Continuing the game you will be in mom's room and you can enter in the Boss Rush and non exit down.
For the Chest you need reach The Cathedral having the teleport 2.0. This object in old days you could find it in the secret room or va chaos. ´
I create a route to depend less or randomness. Provably is outdate at so many levels nowdays.
My route was:
- No retries. Always delete de save slot before start a run.
- No make devil deals nor black market trades. It's ruin the angel room appareance.
- No take pills randomly in the critical floors. Because telepills can ruin the run if brings you to the wrong place (mom's fight).
- Try to obtain The battery, the habit are usefull or any other item that increases the chance of battery drops (for the teleport 2.0 after clear all the floor).
- Try to obtain the telepor 2.0 (secret room) or the shovel and bring it to mom's floor.
- Try to obtain the telepor 2.0 or the shovel in mom's floor simultaneously (no school bag needed).
- Don't beat mom. Don't even enter to the fight (there is no point in it).
- Exit the floor with the teleport 2.0 usin the shovel to create an exit (devil room exit is and alternative, but random to reach).
- In the Womb II we need and angel room, because it contains an access to The Cathedral. Sacrificial room are welcome to increase the chance of angel room or to access it directly.
- In The Cathedral we need to enter to the I am error room. It's contains the acces to The Chest.
- Exit to the Chest vía I am error room exit.
- Kill the smurf.

Well you can also unlock the Forgotten either by speedrunning Basement 1 immediately or after R Key, which is more easy, getting you one extra character.
no, you have to beat the lamb for Broken shovel, and R key leaves the timer as is. you'd need spindown wheel to reduce an item to broken shovel, and it's unclear if that skips story items.
This is a way I thought up to get as many characters as possible in one run (not your first run) without R keys.
You have to choose to either unlock Eden or ??? as both can't be gotten in the same run for obvious reasons.
Jacob/Esau will not be unlocked because that would require beating hush three times, which would require beating Mom's heart ten times first, so both Eden and ??? would not be obtainable. plus there are 3 other characters who are only available in the womb path, so it would be a net negative.
Keeper and Lillith are not gotten because they are in greed mode.
tainted characters are not gotten because they have the same problems as Jacob/Esau.
If you use the sacrifice room to get to the dark room and not the black market or error room, you don't need a get-out-of-jail-free card or Dad's key unlocked as you'll be forced to kill Uriel and Gabriel which will net you the key pieces.
prerequisites:
get the mom ending
get the lamb ending without beating satan (and optionally mom's heart if you want Eden instead) using sacrifice room or a black market in Sheol.
(optional)at some point after beating the lamb, assemble the golden key to unlock Dad's key
(optional)If you are getting ???, then get mom's heart endings 1-9 (you can technically do one mom kill in the same run you get the lamb ending to decrease the number of runs by 1.)
(optional)complete challenge #22, SPEED! to unlock the Get-out-of-jail-free card
do all of this without getting any of the other characters
Actions in run:
play as Isaac (or optionally Eden if you went for ??? for a chance to start with the backpack, clicker, or a chance to instantly unlock some of the characters. Theoretically, you could even play as the keeper if you managed to beat greed mode enough times to fill the greed machine, only holding a max of 54 coins at a time, never having more than 6 red hearts or 4 soul hearts at once, never taking more than two devil deals, and never going more than one level without getting hit or picking up health. Don't do that though.)
play on hard difficulty
at some point in the run, you need to find: backpack, clicker, a Get out of jail free card (optional), and the missing poster.
complete the first floor in a minute to get the broken shovel. (preferably without getting hit or picking up hearts as well)
do the next floor without taking damage or picking up hearts (and the one after that if you failed the first floor) to unlock Samson and Eve.
pick up four soul hearts to unlock Lazarus.
Use the clicker at any point after this until you become Lazarus. If you manage this early in the run and find Dad's key, you can swap it out for the clicker if you don't have a get-out-of-jail-free card to open the Mega satan door.
acquire seven heart containers to unlock Magdalene.
take three deals with the devil to unlock Azazel.
hold 55 coins at once to unlock Cain.
do the boss rush after mom to acquire the other half of the shovel.
Beat mom's heart to unlock Bethany and Eden/???.
die with the missing poster in a sacrifice room at any point afterward to unlock the lost
Use Mom's shovel to go to Sheol.
at some point in Sheol, you must either find a crawlspace with a black market or a sacrifice room and enough health to use it 12 times and live.
kill satan to unlock Judas.
Exit the boss room and go to the dark room using the methods above or, alternatively, finding some way into an I AM ERROR room.
open the mega satan door using the Golden Key, Dad's key or a Get out of jail free card but don't enter it yet.
find an unmarked grave and use Mom's shovel to dig it up to unlock the Forgotten.
Go back to the mega satan door, and kill mega satan to unlock Apollyon and end the run.
If you have done everything correctly, you should have unlocked Lazarus, Azazel, Magdalene, Cain, Samson, Eve, Eden or ???, Bethany, Judas, the Lost, the Forgotten, and Apollyon on between your 3rd and 13th run. and all without the R key. You can then get Lillith and Keeper on the next run using Azazel, deep pockets, an infinite money trick, and some greed machine luck. and then all you'll have left is Jacob and the Tainted characters. make sure to like this stupid post that took like an hour to write.
Very good video but ffs use the video :D I was just watching a run with you talking about the unlockable characters and how to squeeze them in; With visualisation, this would be a masterpiece. Just showing every character you talk about and some challenges about the unlocks in a list. I would have appreciated a systematic approach, at least visualized, take us to an adventure:
Talk about all the characters, how to unlock them, which one can you unlock without problems, which need specific unlocks/floors? how do you get there? What compromise do you do? You could casually point out near impossible ones like Blue baby or Apollyon and say why that would be impossible and just construct a meh run and come back and point out why they wouldn't be impossible to unlock in you're very first run.

Could you even unlock the forgotten in the first run? I'm not sure the broken shovel spawns before you unlock the negative, and R key doesn't reset run timer, so BB and forgotten are not possible in the first run as far as I can see
I think the idea might breakdown about halfway through.
Because you need the Schoolbag to hold both We Need To Go Deeper and R Key, but the Schoolbag is only unlocked by visiting 6 shops in a single run. Because Silver Dollar and Forget Me Now need to be unlocked with Cain and ??? you will need a 5 Pip Dice Room to do one of the floors twice so that School Bag could appear in the shop in Depths 2, and only Depths 2.
Unless beating Mom during the first half of the run lets you proceed past her fight after using the R Key, you'd need to abandon We Need To Go Deeper to take the R Key and since the R Key doesn't reset item pools you wouldn't be able to re-find it after using R Key.

Jacob and esau would be really hard even with infinite r keys because you need to unlock hush, beat hush 3 times, then finally go alt path to beat mother.

You can unlock quite a thew using R-key but i know you cant unlock lilith and keeper depending who you play you cant unlock bethany however mega satan might end your run so i would say unlock apollyon last then go defeat mother with more r-keys unlock the tainted characters and then you can try obtain clicker to unlock more tainted characters and then that all i got

I belive you need 100 death to unlock Scissors so 100 + every compliton mark for every charecter(maybe little less if you could manage more then one for run) is optimal number
You don't need we need to go deeper to get to the lamb
first you beat mom, after going to the title screen, continue the game, which will unlock the heart
then you beat the heart, and you can get a devil deal which will get you to sheol
in sheol there is a chance that a room with a trapdoor to go to the dark room spawns, kill satan and go to the trapdoor you found
then kill the lamb and do the same trick we did with mom to get unlock mega satan's door.

Can't you also try to go for the forgotten in that run? If you don't have all runes unlocked you can get the runeshard and hope it's ehwaz (or any other method stated in the vid), making your spacebar free for mom's shovel.

Took me about 200 hours to get every achievement before repentance came out.
I imagine it will be about +400 now thanks to all the tainted characters.
I played a good amount for fun before each of the DLCs came out but I was at 444 hours when I got afterbirth 100%
Similar sort of challenge:
Can you unlock a tainted character before unlocking their non-tainted counterpart?
You'd have to be playing as a character you haven't unlocked yet, and reach home.
Obviously, you can't start the run with the character.
Clicker can only change you to characters you have unlocked.
Lazarus's rags are a lazarus unlock, so you can't get them without unlocking lazarus
Same with Judas's Shadow
Missing Poster can only turn you into the lost after you unlock the lost.
The Ankh and Broken Ankh do actually work to turn you into ??? without unlocking them first, and aren't tied to unlocking ???, but to unlock home, you must beat mother by going down the alt paths, and the alt paths are unlocked by beating hush three times, and the blue womb is unlocked at the exact same time ??? is unlocked anyway.
I think there's only two possible ways to do it:
#1: "Lostify" yourself in whatever method may work (tainted jacob getting hit by tainted esau, the white fires in the downpour, ect., but i doubt this would work)
#2: Unlock TMTRAINER, and hope you get a lazarus's rags / judas's shadow effect without having unlocked those items or characters yet
#1 won't work because you don't technically change characters. Notice you still keep whatever abilities your character has (like Azazel's mini Brimstone or your Tainted shtick) after going in the white fires, and Tainted Jacob still gets Tainted Jacob completion marks after touching Esau. #2 might just work, not 100% sure if it can get effects you haven't unlocked though.
if you are patient enough you can farm R keys using spindown dice and the schoolbag, this way, in theory you coud unlock also blue baby,the problem lies in unlocking spindown dice
you only cannot get keeper, the forgotten, lilith, or bethany because they all need you to do a new run (except the forgotten since he needs to go to boss rush but he cant)
you can actually get every character other than normal/tainted lilith and normal/tainted keeper

I'm pretty sure you could technically unlock almost everything in "One" run with tainted Cain because you can use clicker to change your character and R key to continue the run and mama mega to open hush and boss rush, you can also use School bag and butter to keep the bag of crafting
Only problem is that mother, the beast, delirium, and mega Satan all give you the cutscene right after killing the boss

If you're super lucky and unlock the alt floors during the run and manage to get a ? card and R key, you can use the ? card to restart so you can go fight Mother and unlock Jacob and Esau, then use the R key to go fight Mega Satan. 13 characters
The alt floors require you to kill hush 3 times and that requires you to kill mom's heart 10 times so you would need to be super lucky and be able to do all of that in under 30 minutes.

Unlocking every character (reasonably) actually only has 4 conditions.
1. Find the moving box
2. Find the R key
3. Find the school bag
4. Find Diplopia or crooked penny on the first and every R key'd run (or find the R key again)
Thought goes like this:
Get moving box
Find diplopia, crooked penny, or R key
Put either in your box
Find the other
Use diplopia or crooked penny
Find (or have) schoolbag
Pick up one and put the other in the box
Repeat until all characters (- keepy boy) are unlocked
There is also a super shortcut that would be ridiculously rare, but sick af.
Any run with:
- Crooked penny or double diplopia
- R Key
- Schoolbag
- Moving box
Is guaranteed to be able to unlock every character (- keepy boy) 25% of the time with crooked. Adding a single Diplopia makes it around 50%. Double diplopia is almost guaranteed.
Note: Yes, the previous calc hinges on crooked penny being unlocked to get there. This means that you'd have to replace it with double diplopia OR get one every reset until you can unlock the forgotten, beat boss rush, and find the crooked penny.
until you realize you cant get past the mom fight without a cutscene, meaning you can't ever open flesh door, so no tainted, and moving box is not a base unlock, you need to go to chest for it, which you cant do yet

there is a way to get extra r key uses, using ? card
it was changed in rebirth to use single-use items again, and now, as long as you have some way to gain infinite cards, you have a way to reset your run infinitely
it is still impossible to actually unlock any tainted characters, as unlocking them requires that you get the polaroid or the negative, and those require you to defeat mom.
however, you can unlock jacob and esau with his.

once you beat mom or moms heart you have a choice to restart the last floor and still get unlocks

So I've been playing isaac for a long time (about 500 hours) and I still don't have Bethany and Jacob and esau
A more reasonable amount of runs video would be interesting and fun, and the every item from zero guide also sounds interesting in how you'll go about it.
I mean obviously the first thing you want is to get every non-unlockable item from the start, then get the unlocks and such. And I guess the best way to get them would be to just get glowing hourglass, a two of diamonds and chaos and break the game in greed or greedier. Sounds hard to pull off but maybe you can think of an even faster way to get them.
You can't open only Lilith, Keeper, Tained Lilith and Tained Keeper for first run, just use R button for many times.

Ok but like, add tainted Cain into the mix who can make infinite R keys (i got 11/12 unlocks that way in 1 run) and theoretically you can unlock all other tainted characters with clicker to make it 16
