How to Transform DNA into Competent Cells

- In this video, Dan shows you how to transform DNA into competent cells.
Notes:
-the DNA being transformed can be either purified plasmid, or your ligation product
-the amount of plasmid you add will depend on its concentration
-the media that you use may be different (here Dan uses LB)
Remember to keep the competent cells on ice, and don't vortex or pipette up and down when you add the DNA. Good luck!!

Protocols can vary. In some situations it is good to spin down after incubating (between steps 6 and 7 at 2:13 ), however in some cases it isn't necessary. And yes as we've written in the description box, it is important to know how much DNA you need for your transformation, so follow your protocols accordingly.

Hi , I think you need to spin the cells down after incubation inorder to get that LB+calcium cloride +glycerol and then resuspend that pellet in fresh LB medium. It's also good to know what concentration of plasmids you really need for the experiment.
