  • Environmental monitoring involves sampling from various surfaces for microbiological quality. For example, laboratory surfaces, product contact surfaces, floors, walls, and equipment should be tested on a regular basis. Swab Technique has been commonly used for such tests. This technique is very important for microbiological laboratories and food & drug production units. This video explained the detailed procedure of swab techniques.

    Thanks for the video! May I know is there ant reason for using 2 cotton stick to swab 2 directions ? Can I use the same stick for both direction. And, do you know what type of bacterial is on the plate? I often saw similar bacterial colony on my agar plate but I am not sure what type of bacterial it is.

      Yes. You can use same stick.
      Type of bacteria depends what we used for the broth culture preparation. We must know the bacteria name and identity. If you work with unknown bacteria then you could know by the bacterial identification tests (biochemical, immunological and molecular tests).

    What is procedure if swabbed area is not possible to measure...in food industry many areas where is not possible to measure by using template like tubes, bends etc....at this situation what we can do...

      Swab the whole surface and calculate using the surface name, as CFU/tube, CFU/bends

      Culture suspensions are prepared in saline solution because 0.85 % sodium chloride solution is isotonic in nature. Normal saline solution is routinely used as a diluent to adjust the turbidity of bacterial cell suspensions to help maintain cell integrity and viability.
