Serotonin and Melatonin Synthesis | Tryptophan Metabolism

  • Lesson on Serotonin and Melatonin Synthesis from the amino acid Tryptophan. Tryptophan is an essential amino acid, which is used to produce the monoamine neurotransmitter serotonin and the hormone melatonin. Serotonin is important in regulation of mood, appetite and sleep in the central nervous system, whereas melatonin is produced in the pineal gland and is important in regulating circadian rhythms. In the process of converting tryptophan into serotonin and melatonin, several important cofactors are used including tetrahydrobiopterin, s-adenosyl methionine (SAM), and co-enzyme A. Vitamins B5 and B6 are also required for serotonin and melatonin synthesis.

      @@ExecutiveZone1 mugwort. you're welcome... Make a tea 🍵 enjoy before bed. And smoke it. It's going to be better. Just letting you know. You can also mix it with sleepy time tea and some honey and a bit of lemon which will curb the bitterness. And also if you actually want to know the effects. Set your alarm. Learn your REM cycle. Normally people within the first 90 minutes are going to have a dream. If you go to sleep and you expect to remember and recall your dreams. It's not going to happen if you fall asleep and just sleep. If you really want to dream recall. And you're not one that wakes up. Go ahead and set an alarm. If you have a watch that helps you find your rim cycle that'll help you more. Cheers have fun

    I recommend to improve the quality such video, is to mention the location of all these enzymes, namely Cytosolic, and for added clarity you also can mention the gene symbols of the enzymes, so the viewer can go and search for them better. Enzymes gene symbols are: TPH1 orTPH2 then DDC then AANAT then finally ASMT all are cytosolic, then the product which is either Serotonin or Melatonin will go to Vesicles via special channels like VMAT2

      JJ Medicine can you please explain supplementing L-Tryptophan? I normally take 10mg of Melatonin before bed, but would it be beneficial to also supplement 500mg of L-Tryptophan with the Melatonin before bed?

    Great video ty. What parts of our bodies are used in the making of melatonin and serotonin? I know the pineal gland is one but just curious do we have to have healthy kidneys and liver etc for this to work?

      If you take a liquid melatonin, it would enter your brain in 5-10 mins

    So... This is why I crave chocolate and cant sleep at night, lol. Guess I'll take some B6 and see what happens although when I take B complex, my anxiety goes away mostly and I feel more level headed and calm.

    Do the enzymes belong to mitochondria-dna, or to nucleus-dna?
    Is there a transporter from cytoplasm to mitochondria for serotonin, or just for melatonin?

    Did you know this? - Glyphosate (aka Roundup, herbicide) suppresses a plant enzyme EPSPSthat plants need to synthesize tryptophane and phenylalanine, as well as tyrosine, leading to the death of the plant. But also: These three amino acids, forming the aromatic group of the amino acids are essential for the production of serotonin, dopamine, and melatonine.
    Just saying.
    (Now I would like to know how much decades of use of glyphosate have influenced the physiology of the plants we eat, and what this might mean for the mental health of humanity ).

    Melatonin not produced from pineal gland. It is released from there after dark, where it is stored after production in the retina in sunlight exposure upon the melanopsin there.

    Nice video, but I want to ask if magnesium involved in this reaction? Because magnesium has some positive effect on sleeping ,but it seems not to be relevant to this melatonin synthesis, can you tell me more why magesium is so powerful to help us sleep "deeper"?

      Mg acts as a muscle relaxant. Helps the muscles open, relax, detox and let go. Which is essential for sleep.

    Nice video, meanwhile do you know anything about how sunlights impact these conversions? at which phase of this entire pathway does sunlight takes place? I’m talking about seasonal affective disorder, thanks

      i suspect the conversion from normelatonin to melatonin. the absence from blue light might be involved in the production of the last methylation enzyme. but that is just guessing from my side.
      at least a bit of supplementation of tmg and msm can boost this. now i understand why some people (including me) are reporting from vivid dreams when taking msm - which is a methyl donor.
      taking tmg+msm is something that i do sometimes.

    My daughter has a high level of Melatonin, and low Vitamin D. She also has nodules on her finger joints. Can anyone lead me in the right direction to recommend nutritional support. From this I am thinking a B Complex, and tryptophan?

      I would be cautious taking L-Tryptophan as this can also be converted to Quinolinic Acid. This molecule is eventually converted to NAD+ but Quinolinic Acid is toxic in high amounts and is the limiting step of the chemical reaction. If you notice any ill effects after taking L-Tryptophan I would recommend consulting with a trusted medical professional.

    Do you believe that N, N-Dimethyltryptamine is synthesized by the human brain? The structure is so closely related to tryptophan and serotonin but there doesn't seem to be any clear answer anywhere. A study has already confirmed that rats synthesize DMT endigenously, however nothing has confirmed this in humans to my knowledge.

      DMT is synthesize from L tryptophan. Not 5 htp. I dont remember the pathways, but to generate DMT on the pineal gland, you need L tryptophan as the genesis.

      Theres no evidence for N,N DMT production in the brain but there is evidence of breakdown of melatonin into 5 meo DMT and there's a paper where this was detected in the urine of schizophrenic patients.

    Error was not citing the production of melatonin in the gut for bacteria and produced for mitochondria.... Not just for sleep... Thought the body. To stop at sleep is a narrow statement

    Only total body melatonin produced by the pineal gland is about 5%. You need to learn how important melatonin is 😊
