Great podcast, thanks for taking the time to publish this! I toured a cannabis testing lab once and was a bit surprised to see that it seemed staffed by all college students with managing PHD. I was watching them prep samples, and while they were fast, I cannot imagine even the most competent lab tech not making a mistake weighing a sample out of the hundreds they do every day. Imho the most probable source of test result error happens when weighing sample material. Especially since labs have an economic incentive to run the samples with as little solvent as necessary -- and calling for a 150mg sample size looks really good on the spreadsheets but opens the door wide for testing error.
Also, having worked in the cannabis industry as a manufacturer I can confirm all your claims regarding the economic incentive of the testing labs. There is no standard testing procedures for testing beverages, so I got 3 totally different test results across 3 different labs -- and of course I went with the lab that gave me the required numbers. And mysteriously enough, these numbers have never deviate outside of the acceptable range...so I have not had to try any other labs! ;)
After teaching labs to college level students for many years, I can agree that their standards for accuracy and precision are not what you might hope for, especially if they are getting paid minimum wage and the results have no repercussions or significance to them.

I see your go for about 2-3” sub depth? Is the idea that a shoebox requires less? Do you tend to just flush them once and for that reason extra would be a waste?
Also interested in how you prep your coir.. pasteurised at roughly field capacity the adjusted to the right moisture when it’s ready to use? You wrap up the left overs for the next day?

I usually do two flushes with shoeboxes and then toss them. I boil my CV and then squeeze it to field capacity one handful at a time.

Mic sounds really good :) Interesting conversation about the HPLC testing although it’s probably not something I’ll be doing … no real need. However, what got my attention was all the possible variables you mentioned and how they could possibly affect potency. I guess that means there are many possibilities within one single grow.
So you use “incubation” … is that the same as colonization ? You said you incubate for 4 - 5 days. Are you going by a strict (ish) timeline or are you waiting to see certain surface conditions like widespread mycelium or what people call knots ?. Of course assuming good growing conditions. Then when you go to fruit are you just dependent upon the humidity within the tent or do you mist the surface ? Do you remove the lid or just loosen and offset it.
Oh, I almost forgot …. Can you over incubate a tub ? If so, I can I do to remedy it ?
What material are you using for a liner ?

I use incubation to mean the time after mixing spawn and sub. Time and protocols vary a lot. I fruit in tents, so the conditions need to be adjusted from time to time. Really changes all the time. I try not to mist too much, but sometimes it's necessary.
I use cheap garbage bags for liners.
Is it possible to over colonize / incubate a tub ? If so, what would that look like? How do I fix it ? I have a couple of tubs that seem to colonize fine but when I switched to fruiting conditions, they just sit there … actually the surface may even be drying out. That’s why I ask. Thank you for your help and all you do for the community with your videos.
@@StickyDankFingers overcolonized probably isn't possible. There could be many reasons why it stalled.

I learned the hard way with aborts when I was younger, work was weird the next day... And I had plenty of time to get back to straight, there was a huge delay!
Aborts and contam are just part of the hobby. Being and staying small, at least the losses are smaller. I can't imagine if you had several thousands of dollars invested in a single grow and aborts or contam happened. I guess you deal with it the same way. Suck it up, take a deep breath, and move on to the next grow! Mycology people and farmers must be some of the most resilient people in the world :)
Lab shopping
A mycoquart of spawn to 3 liter of coir has become the go to here on "The Farm". We're looking to develop an extraction technique that can concentrate alkaloids. We've read that 2g/liter water is the saturation point. A simple evaporation left a thick tar with the lipids that came along without any alcohol extraction involved. Boo...
Concentrated extracts are going to oxidize very quickly.
Ed, you should add the $thanks button. I, for one, feel like I want to send you some $ for your efforts. I know you don't call yourself an expert, but you are a classically trained researcher and a tenured mycology professor (regardless of current status). You have placed yourself in another teaching situation, and there are those of us who would like to give back to you. I get that youtube might censor you and you probably feel like a patreon might be asking too much, but seriously, you are providing value to guys like me, and maybe we want to give back! I got my martha and pee pads with my amazonians(holy shit), and my Malaysians(ole trusty's) and I got my APE's(slow and too blue, but I'm learnin') I've got my overly engineered flow hood in the works and my cheap ass spawn bags ordered from ali-baba, lol.

They are shoeboxes. They come that way. Doesn't have any affect in my tent style of growing.

Lemon/Citrus Tek all the way. Consume immediately or freeze. Keeping stable extracts is problematic because of oxidation. That's why there is no proper extraction tek videos. It's a waste of material.
I’ve been doing citrus method , I figured out a cool method , I used your Drizank video to learn more and bought powdered citric acid and just add it to water and have tinkered with the strength of citric acid, I’ve been making gummies with the extract, will they still oxidize in gummy form , I’ve been vacuum sealing them individually with oxygen absorbing packets ,

@@MycoMoat I'm not familiar with gummies. I would think any kind of extract is gonna oxidize quite fast, but not sure how fast.
What ratios are you playing with, did I hear you say 1:3? What effects have you seen?

I just seen a post of some test results of some TAQ's. It said 13.4mg/g. That seems very high.
Yeah. That is why I've started totally distrusting any HPLC testing. Where was that reported?
@edwardgrand why do they think they are actually that strong? Almost 4x the average potency of both species used to create TAQ.

HPLC is problematic, there are too many constantly changing factors. End of the day it'll probably only help people sell swabs. Then there's the obsession about potency, don't get me started 😅

Yes. That was one main point I think I forgot to make. Controlling all the variables from grow to grow is nearly impossible at home. So, even if we get a high test, it'd probably be extremely hard to replicate the conditions.
