Column Chromatography Basics and Lab Procedure

- In this video I have discussed about what is Column Chromatography, what is its principle and lab procedure and How to perform Column Chromatography in lab manually step by step.
First step is to choose the stationary phase based on sample.
Second step is to pack the column, packing can be done by two method 1) Dry method 2) Wet method.
Third step is to choose Mobile Phase
Forth step is to load sample onto the packed column. Sample gets purified on the basis of polarity.
Later we have also look in brief at isocratic elution and gradient elution.

In the column chromatography, If stationary phase is silica gel, the mobile phase is the mixture (ratio of petroleum ether: ethyl acetate is 9:1 ). The sample include compound A and compound B. IF the polarity of A is great than the polarity of A.
Which compound will be eluted first. Please explain the reason.

mam agar sample polar hai to wo silica ke sath bond form karega to polar sample ka separation karne ke liye kaunse solvent use kar sakte hai.
