- Isoniazid (INH) is a narrow spectrum antibiotic used in the treatment of tuberculosis. This drug is a nicotinic acid derivative with hydrazide as functional group.
Isoniazid is a prodrug that is activated within the mycobacterial cell by one of the important enzyme catalase-peroxidase in presence of hydrogen peroxide. This enzyme is coded by the gene Kat G which can be mutated by the organism to produce resistance.
Finally this drug forms an adduct with NADH molecules which inhibits synthesis of mycolic acid within the mycobacteria.
