Yes, without water the paint dries out and hardens. But beware, if you don't use the paint for too long the water will get moldy, so keep painting often ;)

Depends on the way you want it to move and look. Could be a painted cut-out, could be 2D animation, or a CGI element with a painted texture

Any specific resources (books etc.) would you recommend learning to paint in this style? i have few books that feature background from ghibli movies for reference.

We simply scan them in and combine background and animation in After Effects

i'll answer you if you don't mind, these are goat hair brushes, also called sumi-e brushes. i did find them in abundance in japan but I believe amazon and places like aliexpress has them too. you don't often find them in stores, online is your best bet.

Yeah, either Japanese art stores or online are your best bet. I don't know where you're situated, but there's a European online store now that has struck a partnership with Nicker to sell their stuff. It's called 'Trendliving'

Layering with poster colors is hard, because the paint will blend with the previous layer. If you have an opaque layer underneath you can add little details on top, but a full new layer won’t work. With thinner transparent layers it is possible to work in layer. Good luck!
