Is there a potential for adverse effects of 5-10 mg of folate for heterozygous MTHFR?

Sometimes when you have a deficit in a vitamin you get depleted at the cellular level so the rationale is to take high doses for a period of time so that the cells can become repleted. There is a complex relationship between thiamine and folate in that they are exchanged in cells. I think it is TPP is exported in exchange for Methyl folate to be imported. Paper: Folate and Thiamine Transporters mediated by FacilitativeCarriers (SLC19A1-3 and SLC46A1) and Folate Receptors

So to support slow Methylation you can take in combo essentially
    Choline
    Creatine
    Methyfolate

Very informative and helpful. Thank you!
I have adhd and have given up on prescription medicine long ago. I'm starting l methylfolate 400mcg next week. I already take cdp-choline thrice a week, it works wonders for me. Should i also try supplementing with Glycine?
If yes, at what times should i take these three? All together in the mornings or only glycine at night? Please advice 🙏

You can try anything. Best to run labs first if you can.

@@chrismasterjohn thanks for your reply! I think you understand nootropics better than anyone else on yt. People should be listening to you.
I had lab done on a few things. I have low vitamin D3 & low testosterone. I have been on statins for 6 months.. It might be affecting my memory but I'm not sure.

Great video as usual. If I may, I'd like to post a comment I made on your video about alcohol as a beneficial, hormetic stressor:
    ---
Dunno if you still reply to old videos, but what do you think about using this alcohol for hormesis protocol if you have a history of gout?
EDIT: Decided to self-experiment. 1/3 glass of Italian red or New Zealand white wine a day with dinner. Currently a week in. I myself have a history of gout (last flareup 2 months ago) which gets triggered by a "standard" night of drinking (3+ servings of alcohol). I also have a history of alcohol "cravings". Drinking elicits some minor itchiness, but no rashes. Will very shortly start supplementing with 10K IU Vit A every lunch time.
My ultimate goal is to better metabolize Vit A (with all of its health benefits), strengthen histamine tolerance, and just enjoy a small alcoholic beverage at night to unwind without fearing its negative effects. To anyone coming across this comment, wish me luck.
EDIT 2: After over a week of trying this, I have decided to quit. For now. Despite the short timeframe, I was mildly surprised not to have had gout flareups from drinking every single night. It helped that I've become an alcoholic lightweight due to keto & losing weight. Even the first sip of my single tiny wineglass is enough to get me tipsy. I did encounter an all-too familiar craving for alcohol on some days. It's been 4 days since I stopped drinking though, and so did the cravings.
I mainly stopped because a) I lost no weight, b) it was interfering with my intermittent fasting schedule, and c) it was interfering with my Vit A supplementation, which I take at night. Once I reach (or am close to) my goal weight, and gone at least a solid month of daily 10K IU Vit A, I may consider resuming this experiment.

Okay so just to make sure I’m hearing correctly: Do *not* supplement methylfolate at 5-10mg per day. Instead, eat more choline and glycine, add in creating and some riboflavin. Is this correct? What about supplementing methyl b12?

      I took 5:2 mf:mb12 and i became deficient in b12 it’s better to take 1:1

If a pregnant woman suspects she has the worst form of MTHFR after an extensive history of second trimester normal chromosome baby losses, and has no other known blood clotting disorder (that was detected...though highly suspected)....what should she take during pregnancy...?

That was informative, thanks! I would appreciate if one day You could try to explaine [in your talented way] the difference in COMT & MAO-A enzymes. If there is any. The MTHFR thing You help me aaa lot with your videos! but when I see that I have 2x heterozygus MTHFR (rs1801133,rs1801131) + "slow?" COMT (rs4680 AA [same as TT] and rs4633
AA ) + "slow?" MAO-A (rs6323 AA). Do you see in the mathylation pathway a connection MTHFR and the COMT, MAO-A? and would those slow enzymes change my strategy (Riboflavin, choline, B groups ect from foods like liver and fresh leafy greens - I call it for my self the Masterjohn protocol :D ). Thanks for Your time on this!

Super interesting - thanks! I wonder if there are any online calculators where you provide your genetics and any other relevant personal info to see calculations of what your requirements are for 5-MTHF, choline, creatine etc. If there are mathematical formulas in existence, it should be possible to provide such a thing, right?

This is actually confusing, because well I have been reading about recommendation to take such high doses of L-Metylfolate on many occasions already. It seems like people are feeling better and their blood markers are improving, so this is all a lot of information that is not aligned with anything else I have read so far. Also the supplement with L-Metylfolate I take recommends only 400ug a day (single pill) or two pills 800ug. Which is consistent with your stance, but It seems impractical trying to grind the pill to spread the L-Metylfolate several times throughout the day. I know taking the food is the best, but let's be realistic here who is keeping track of micronutrients on a daily basis, I used to weight my food and count calories and macros, but this is like a next level, in a simple worlds it's just very impractical with a high chance of error compared to just take a small pill every day in the morning.

Lower doses can be done with a liquid like this: www.amazon.com/Holistic-Health-International-LLC-Methyl/dp/B07N7FMH3M/ref=sr_1_1_sspa?crid=S8ALW33KYM3S&keywords=liquid+methylfolate&qid=1654982860&sprefix=liquid+methylfolate%2Caps%2C59&sr=8-1-spons&psc=1&spLa=ZW5jcnlwdGVkUXVhbGlmaWVyPUEyQVRTTjRCNVVONUdTJmVuY3J5cHRlZElkPUEwMjA1MTgxMTVHRkNTSzJLUlVINiZlbmNyeXB0ZWRBZElkPUEwMjQ4MzY0MlpXUUZLS0pYT00zJndpZGdldE5hbWU9c3BfYXRmJmFjdGlvbj1jbGlja1JlZGlyZWN0JmRvTm90TG9nQ2xpY2s9dHJ1ZQ==

As i read folate deficiency needs 1:5 ml for 1:4 months b12 deficiency needs 1m injections every other day for two weeks or 5 ml oral for 2 months
