Nicotinamide Riboside vs. Nicotinamide Mononucleotide - dosing and effects on NAD+ | David Sinclair

  • Two NAD+ precursors, nicotinamide riboside and nicotinamide mononucleotide, have been shown to ameliorate age-associated diseases in animals. They are well-tolerated at high doses and effectively raise NAD+ levels. Evidence suggests that these molecules, often referred to as "NAD+ boosters," may increase levels of NAD+ in humans, too; however, the doses used in some of the studies equate to far less than what was provided in the rodent studies. Currently, only nicotinamide riboside has been shown to raise plasma NAD+ levels and may possibly have beneficial health effects in humans. Future research, focused on "super boosters," may have relevance for treating complex diseases such as diabetes, heart disease, cancer, and Alzheimer's.     people, only take what you can afford..if you try and go all out with powders and supplemts and end up broke and homeless i can assure you that stress because of your financial problems will cause rapid aging... so just stick WITHIN your means..besides, consistancy is key.. then when you get more money you can buy more suppliments.

    What about Ovasitol aka inositol in specific ratio (different configurations of inositol molecules)...as effective as Metformin in studies for PCOS. Does it also boost NAD+ if works like Metformin??

    We need more clarity on NR vs NMN and whether the whole slc12a8 allows NMN to be transported into the cell. Update from Sinclair is needed at this time based on new studies which he himself posted soon after this video was shot. Can someone make test kits for NR, Nicotinamide and NMN.

      We need to wait few months probably for more research. The fact that David Sinclair says that nicotinamide inhibits sirtuins and new research shows that most NMN and NR breaks down into nicotinamide -- is kind of worrying. I started to take extra Pterostilbene and Resveratrol in hopes that it will prevent inhibition of sirtuins. They need to do studies on sublingual NMN and NR. Also, I think there are no studies on sublingual NAD and nasal NAD because some people say that NAD itself can get directly into bloodstream through these deliveries. Screw nicotinamide if we don't need it in our bloodstream.

    So, are we still thinking Elysium is the best choice for boosting NAD? I understand resveratrol may, but it hasn't been IBS friendly to me. Nor broccoli sprouts which is more NR than NMN, correct?

      Niacinamide is Vitamin B3 and B-Vitamins is water soluble. meaning it can be easily released from your body via pee and sweat so you‘re not getting most of what you need or it doesn‘t stay much in your body if your not taking it many times a day like other water soluble vitamins like Vitamin C.

    why does it have to be nicotinamide riboside or nicotinamide mononucleotide? Wouldn't nicotinamide have similar effects?

      There's some evidence that suggests, according to Dr. Sinclair, that nicotinamide suppresses sirtuin activation. Whether that translates clinically is unclear. Especially in light of the NAD+ flux study shown above. (It was still sort of a brand new pub when Rhonda and Dr. Sinclair had this conversation.)

      thanks a lot for all your answers. So from the literature you guys have posted there's only this one study where NAM could suppress sirtuin activation in vitro, all the rest seems rather favorable.

      @@bonfynil3238 I am using normal Niacin that gives flushing. Also 11 years on a plant based diet low in fat not oil and I could lower this way my arterial age to ome of a 19 years old man. Works better then any supplement out there in keeping you young especially inside.

      Niacinamide is a NAD booster but it is cheap and therefore not profitable like NR or NMN. Tests with human cells do not show a negative effect on Sirtuin expression.

    Too expensive to take these large doses of NR or NMN for the rest of your life .I doubt serious studies will show life extending properties of either one .Supplement companies will take in big money for now until the next big thing comes along .The only fountain of youth is genetics ,diet,exercise,and luck ,

      For diet, see Dr Richard K Bernstein. A type one diabetic who was told around 40 that he had about 5 years to live. An engineer at the time, he managed to get his hands on an early blood glucose meter and realized the food pyramid was upside down. He began to control his diabetes via diet (leafy greens, fats and protein) and small doses of insulin. Became an endocrinologist, reversed his diabetic complications and is now in his eighties, working and working out daily. His contention has always been that diabetics are entitled to normal lives. He's more than proved that, and by the way, recently had a calcium scan and got the lowest score possible. I.e. - no calcification.
      Don't believe the the vegetable malarkey pushed by vegetarians, it is All about blood sugar.

    For an Amazing Arterial Function and Endurance eat a plant based diet low in fat not oil. You will see magic happening... At 43 I reversed my arterial age to a 19 years old young man just with a diet extremely low in fats and of course without dietary cholesterol. The test used to check my arterial age was the PWV test (Pulse Wave Velocity test).

      @@bruttosporcoecattivo fats are everywhere. All veggis, grains, legumes have fats in them in the perfect amount. I simply cut the foods containing high fats. For omega 3s I use 2 tablespoons full of ground flaxseeds amd it is the only high fatty food we consume in our family. Been doing this for 11 years now. My wife also after an angiogram the cardiologist told me it was hos first time to see new born baby arteries in a 45 years old woman. Eat a plant based diet and cut the high fatty foods and oils.

      @@bruttosporcoecattivo only 2-3 walnuts every few days still for omega 3s. Not other nuts. For supplement we use B12 1000mcg 2-3 times/week and that's it. In winter months if we are in a cold dark place we might use some vitamin D3 and K complex bit seldom as we mostly live in the tropical Australia.       Tried "plant based" for a year. Did nothing for me but weakened my immune system and energy levels. Just in one week of a keto, or low carb high fat carnivore diet I got so much energy I felt better than if I was a child!! No more joint pains, and moving light as a feather!
