  • NMN, NR, Nicotinamide and NA all offer pathways to increasing NAD levels in human cells. In turn NAD+ is crucial for muscle strength and skin health and for increasing energy levels. But which of these supplements is best to improve NAD levels? In this video, we explore that question as well as dose and safety recommendations.

    Niacin 60mg was equivalent to 1000mg NMN in raising NAD. Don't waste your money.

      @@Terrybear27 thank you and it’s amazingly low dose to equate to 1,000mg of nmn. Most NA supplements are 250mg pills. Can cut up however

      Substantial part of orally administered NR and NAM is metabolized to NA by the gut microbiome

    NMN is still available outside of America. Do Not Age ship third party tested NMN from England and our standards for food and medications are higher than in the U.S.A, so probably much safer than American brands.

    NMN and NR taken orally are a waste of time, as these are largely converted to Nicotinamide by the gut microbiome, and whatever does get through is metabolized by the liver. In order to get enough through into the bloodstream, you would have to megadose 4-5 grams or more, which would be toxic. Even then, there is reliable research that shows it's not getting into the majority of cells, because it needs a membrane transport protein. One of these has been identified for NR, but not yet for NMN. Also, studies showed that injecting NR directly into the cell is more effective than NMN. Any of these pre-cursors that remain in the blood stream are quickly converted by the liver into Nicotinamide. So any benefit that people believe they have by taking NMN or NR is likely to be Nicotinamide. NAD+ injections are also mostly a waste of time, because the coenzyme is too large to enter the majority of cells, with the exception being the blood-brain barrier and the heart.

      Hi, my English is not good. Do you think taking 500mg NAM (Niacinamide) is good for improving NAD level?

    • @LucidAmethyst
      @LucidAmethyst Před 6 měsíci

      @@huseyinmuhtar3095 500mg of Nicotinamide will help maintain NAD levels through the salvage pathway, but if you want to increase NAD, you would be better taking small doses of Niacin, perhaps 15mg - 50mg. In amounts over 50mg you risk the flushing side effects.

      Agreed. NMN is a hype created by David Sinclair with questionable trials In humans. However, vit B3 either as nicotinic acid (which causes skin flushing) or nicotinamide aka niacinamide (no skin flushing) are most effective at raising NAD. Besides b3 in both forms is far cheaper than the marketing around NMN.

      Yes. 500 mg might be a bit high. Start on a smaller dose. It gives your energy very fast. And it’s safe and affordable way for NAD production.

    Sir, how is Nicotine (from cigarette?) related with the "nicotin-" containing compounds mentioned?

    • @PharmacoPhoto
      @PharmacoPhoto  Před 10 měsíci +1

      Hi, it is pretty complicated. Nicotine is a secondary metabolite of the NAD+ biosynthesis and the NAD+ pathway. But all of that happens safely in the cells at a regulated pace, and then it gets recirculated, making NAD again. When you smoke or consume nicotine, it produces a massive amount of nicotine in the blood which then bind to the nicotinic receptors on the outside of the neuron cells. But nicotine has none (0) of the positive benefits that NMN, NR, nicotinamide or NA have and those NAD+ products have none of the negative effects of nicotine.

    • @LucidAmethyst
      @LucidAmethyst Před 9 měsíci +1

      Niacin, or Nicotinic Acid (NA) is the acid form of Nicotine, and is solely derived from Nicotine. The name Niacin was given to stop people believing that smoking is good for you.

      Niacin not niacinamide is best for raising NAD. 60mg Niacin is same as 1000mg NMN in raising NAD levels. Doses of Niacin 300mg twice daily restore NAD to youthful levels. Search for 'Niacin' and 'NAD' CZcams or Google.

    You don't need 1-4 grams of NA to significantly raise NAD+. Just a fraction of that will be the same as much higher concentrations of NR and NMN (NMN has to be broken down into NR to be transported into cells). So if you take under 100 mg of NA, then almost all people will have no flushing and the claimed insulin resistance increase is not significant at that lower level. What's more the existing evidence points to NMN and NR being almost all degraded by the gut microbiome intor nicotinamide. The claims in this video that only 125 mg of NMN is needed to get an effect are just complete nonsense. Sinclair himself takes at least a 1000mg or more to raise NAD+. As to the claimed benefits, there has been no studies proving that at all, just clinical studies providing evidence of safety. Of course NO list of references to back up those big claims for NMN. Experiments inhibiting the conversion to nicotinamide also remove the increase in NAD+. So NMN and NR are a BIG waste of money. Supplementing with Nicotinamide is known to inhibit the synthesis of NAD+ so without references, doubt those claims as well. There has been research showing the benefit of taking D-ribose along with NA, both of these supplements are cheap building blocks of NAD+. But since companies can't patent them and they are so cheap, there is no profit in talking about them. The use of exercise at least is the one thing this video gets right. Suggesting using NMN is just garbage advice.
