    Thank you for posting this. It seems like an open flame that torches the containers' brims was unnecessary and it's okay to hover over the explants. I was taught with these cautions in school but I want to improve my efficiency by simplifying the process. This video is informative.

    Thank you so much for sharing this info! Slowly making the transition from growing culinary mushroom culture (reishi, lions mane, oysters) to plant tissue culture. Rare to find these helpful videos for cacti plant tissue culture techniques! Great job and very informative! Hope to find some MS cacti media (and woody plant culture) recipe videos someday :-)

      Regular base media works for almost everything. Some special medias work better or just marketing

    Dude you are awesome, this is a really great and helpful set of informative and detailed videos for such a difficult to expand class of cacti. I really appreciate you doing this. Since I own most of this equipment already, I couldn't be more excited to start trying this.

    I would suggest if you use a sterile 240ml wide mouth bottle with a cap so you can rinse your explant with dist H2O 3x by shaking then decant the water thoroughly. You have too many paper on top of the bench. Cut it into smaller pieces bond paper size (sterile) to carry out your disection of the explant if you do not want to use a petri dish. Bec us we are used to petri dish. There is an effect on the growth of the explant with the size of the tissue vs container to volume ratio of culture medium. You need to maximize the growth in short period of time.

    Loved the video, did you trim the explant material that was exposed to bleach after rinsing, before cutting down into smaller pieces?

    Do you use PGRs in your media? I’m about to start a large tissue culture operation of palo santo but definitely want to branch out into xerophiles and carnivores, and for the palo santo I will be trying somatic embryogenesis and from there using a bunch of pgrs to pump shoots, etc

    @cactiFanatic do you have a list of hormones and where to get them. Such as a package deal where I can buy them all? I’m not tech savy so I don’t really know much out online stuff.     great job, when using the MS medium do you happen to add any other growing hormones for specific growth? would you suggest starting out at half strength ms powder? good looking out.

      Honestly for cactus they don’t require any hormones once they get going but I do add to speed up things

      You could but it’s very long and difficult path. Best to use nodal / meristematic tissue like
      Spines / aeroles in cactus

    So if I were to TC dragon fruit varieties, would my explants need the actual stem from the middle, or just the spines where new growth occurs? Are bigger or smaller chunks better? Do you know if contaminants can get inside the container if it's sealed? Thanks so much for this video. It has helped me tremendously.

      The smaller the piece contains the meristem the better but on the other hand having more tissue they survives with the meristem encourages faster growth. More tissue increase contamination chances.
      No they contamination does not get through. All containers are wrapped with micropore tape of moving to regular non clean room environment

    Question, lophophoras are painfully slow in their own roots, This accelerates that or just helps with propagation? Also you should try tissue culture with seeds

      Yes I have over 100 started from seed. They are very fast in TC compared to regular in my opinion but still not as fast as a pereskiopsis graft

      I was told you don't need it if you are using the Gamborg B5. The media is for callous plants like most cacti. 👍😁

    What about the media. I want to clone Trichocereus cacti. I get the basic idea. I am unsure what media. Is there a protocal for cloning cactus that is not behind a paywall?

      Yeah!! It’s online free. Just use a MS based media with vitamins, 30g sugar, activated charcoal and agar to set. That’s good enough for basic tissue culture then maybe BAP to induce
