    After years of trying methyfolate, finally I feel confident that I just needed the glycine but also Dr Ben lynch recommended liposomal glutathione. I think both are helpful. Niacin also helps with the mood sided effects. High doses of zinc (with some copper and a little manganese) also helps with GABA synthesis and insuring I get rest. I also take hydroxocobalamin for the B12. I'm sick of stress ruining my life and the right nutrients definitely are starting to help.

      @@christianfernandez4285 methyl type might cause to over methylate and make it worse with a lot of symptoms ..so if you have certain gene mutation it is better to have adenosyl cobalamin or hydroxocobalamin to avoid some adverse effects in the body
      YMMV, try using methyl version if that is working with no side effects then stick with it. If that doesn't work try with adeno/hydroxocobalamin
      p.s. methyl version of folate or cobalamin doesn't do good for everyone for which you need to do some gene testing to identify what works best for your gene type

      Hydroxocobolamin is given as an injectable, usually as one monthly dose by a medical professional. Methylcobolamin is usually taken as a supplement. Methylcobolamin is better for undermethylation but may take a little bit longer for blood saturation to occur

      The culprit is Not methylfolate, but the way you have been having it - either in isolation or without its cofactors or in higher doses or without balancing methyl groups or without using other methylators in conjunction.
      Most folks are ill-informed. They think 5MTHF is the sole methylator. Here is a list of methylation support.
      1. SAMe - Universal methylator
      2. 5MTHF - Principal methylator
      3. TMG - Secondary methylator
      4. Choline - Auxiliary methylator
      5. Glycine - Buffer for Methyl groups
      6. Niacin - Antimethylator (methyl group acceptor, used to balance overmethylation)
      7. Cobalamin in non-methylated forms can also accept methyl groups from 5MTHF and func ad demethylator.
      Further, Zinc and cysteine are required in many methylation reactions. For me, SAMe is the most critical methylator.

    I took methyfolate after finding out I had MTHFR and had a horrible reaction a few hours later. Horrible anxiety and a panic attack that had me trembling. I still have anxiety from it two days later. When will this go away?

    Overmethylation... COMT gene mutation has a lot to do with it as well. Look towards hydroxy and adeno b12 instead, and use folinic instead of methylcobalamin.

      this is what i do. cyan B12 and folate and no problems. But if i try methyl b12 or methyl folate i get anxiety THAT day and it lasts for 2 days

      You can use Niacin to balance overmethylation as well which is what I do and it works beautifully !

    MethylFolate is critical for DNA, RNA synthesis (& repair), nucleotide synthesis, RBC synthesis, WBC synthesis, methylation, iron & heme metabolism, as well as for activation of several proteins, neurotransmitters, hormones etc.
    It is also essential for homocysteine regulation and repair of blood vessels & nerve cells.
    Those you do Not understand methylfolate well have it in isolation and/or in megadoses without understanding its cofactors & then cite side effects !
    Following are the synergestic cofactors of methylfolate ::
    1. (Methyl)cobalanin / B12
    2. Active B6 - P5P
    3. Glycine
    4. Active B2 - R5P
    5. Magnesium
    6. Zinc
    7. B3 - Niacinamide
    8. Copper
    9. Vit C
    If you are deficient in one / more of above & supplementing folate in isolation esp doses above 400 ucg / day , you are going to have side effects !
    If you are overmethylating, you must balance your methyl groups by having Niacin which is a methyl group acceptor & vice-versa. There exists a delicate balance between B3, B6, B9, B12, glycine.
    Having synthetic Folic acid also disrupts Folate metabolism.
    I am generally tired of youtubers puting up half baked info & confusing the audience.

      Thanks for sharing. I agree with most of what you say but not all of it. This video is meant to be short and is on common reasons for methylfolate side effects (my personal experience treating people). It is not every single reason. Frankly we do not know every reason people get side effects from methylfolate. Sure deficiency (in anything mention above plus a lot of others but especially B12) will lead to problems. Their very nature suggests that. Some people get side effects in the absence of said deficiencies. This video and your comments are good trouble shooting ideas.
      I have been helping people with methylation issues and MTHFR (among many other ailments and illness) for over 16 years in clinical practice. What that teaches you is what you know and don't know. It can be humbling at time but that is how you learn. That's what I try to share here is the synthesis of that learning.       Worst part about methylfolate is that people and advisers dont know anything about it, they just wrote that mthfr is needed for synthesis of methylfolate and they are teaching everyone that if you have mthfr then you need to suplement this nutrient. I really encourage to look at study that showed that 3mg of riboflavin lowered homocysteine by 40% in people with mthfr and this is really key nutrient for those people not methylfolate. Methlfolate is methylated god knows how many times everyday and dose that you get from suplement will be transformed into thf in like 1-2 hours after taking and then it isnt different than folic acid that is demonized. In the end i want to write that b2 is key nutrient because it speeds up mthfr and if you got polimorfism then your enzyme need more b2 and even more facinating is fact that such low dose as 3mg will be enough. If you dont notice any improvement in blood homocysteine or your mood then mthfr isnt your problem and you need to dig more into your biochemistry.

      @swintegrative After experimenting with each B vitamin in isolation for 10 years !...as well as their several combinations and trying several brands (more than 40 !), the #1 reason for methylfolate side effects is HAVING METHYLFOLATE IN *ISOLATION* LONG TERM INSTEAD OF HAVING METHYLFOLATE AS PART OF A FULLY METHYLATED FULL SPECTRUM COENZYME'D B COMPLEX WITH ALL B VITAMINS. 95% companies do NOT know how to formulate an optimum, fully methylated coenzymed full B complex - that includes the active forms, the optimum ratios, the supporting cofactors, and the CONSISTENT batch after batch purity, potency, and efficacy. The closest I could get to such a brand was Douglas Labs which uses Metafolin (the #1 methylfolate from Merck), uses 100% P5P (no mix of pyridoxine hcl), uses 100% R5P (no mix of riboflavin), uses niacinamide (no mix of inositol hexaniacinate), additionally uses some choline & inositol, uses d-biotin variant, and to top it all uses the absorption cofactor for b12 which is IF (porcine derived).

    I took methylcobalamin and I've got all these side effects it's made me very poorly for a year now since I took it, It has caused me awful health issues before I took it I had no issues and was healthy energetic and happy now I'm house bound and have awful histamine issues

      @@queeny4624 Sorry, but there's no way that the B12 you took more than a YEAR ago is still causing any ill health. There's some other stressors in your life that you probably need to address. Millions of people take methylB12 and it helps them to SLEEP, to RELAX because it helps break down and eliminate histamine. You could be low in methionine (which is needed w/b12 to lower histamine), or low in copper which is needed for the DAO enzyme that breaks down histamine in the gut. Good luck.

    I have many autoimmune problems and now we have discovered this homozygous MTHFR C677T deficiency.I have taken an L methyl folate supplement for six weeks.
    I don't know up from down right now.
    I have had serious infections recently, trouble with newly diagnosed IBD, and many other autoimmune disorders and symptoms.
    Depression has always been a battle.
    As I recover from infection,I should be feeling better. but I am waking with anxiety at an irrational fear level each morning. With so many doctors involved in my overall care, I am embarrassed to say anything. I just feel like I am avail are and causing problems all around.
    I am supposed to be getting BETTER, not having an adverse reaction to the supplement meant to make me well!

    Now I have more to consider. Maybe it's more than my faulty brain.

    Hi Dr., I’ve had years of issues, come to find out I have MTHFR mutation, I tried a methylated b complex/methyl folate and for 2 weeks my body and mind absolutely LOVED it, sharp mind, losing inflammation by the second, energy of a child, calm demeanor. After about 9 days the side effects started, burning head and back of neck, pounding constant headache that lasted 10 days, extreme tinnitus, lethargy, brain fog. I stopped the supplement and was so bummed.
    I saw your video just now and you mentioned GABA. Years ago I had to taper off of benzodiazepines (anxiety medication) after taking them for A year for insane anxiety and was told my GABA receptors would basically always going to be fried.
    I’m now wondering if my reactions to methylfolate/methylated b’s were because of my fried GABA receptors and wondering what I can do to help that to possibly reinstate my supplement 😭😭

    Thank you for your video. I have MTHFR and started taking the methylated B complex a couple of days ago. Shortly after taking it I developed a headache and my stomach felt like I just ate a jalapeno pepper. I thought it was a fluke and a couple days later tried taking it again with the same results. My husband decided to take it (he doesn't have mthfr) and he didn't have a problem. I was considering taking a lower dose of methylated b...but after your video...I'm not sure what to do. Any advice? Thank you

    Thanks for the video. This is great information for those who have refractory anxiety. I'm an NP-student and plan on working in functional medicine. But for myself, I'm currently dealing with my own MTHFR-gene mutation. B9 Methyltetrahydrofolate kicks me in the butt, extreme lethargy about 45min post ingestion. Same thing for my husband. I have out of range elevated B12, B9, and high MCV. If these are high, (serum levels) how do I get them in the cell? I am also taking NAC / Glutathione and that gives me a terrible headache and brain fog... thinking i might be depleting my B12 but what's confusing if its elevated, how is it being depleted? Trying to wrap my head around this for myself and my patients. I feel like the methyl trap and MTHFR mutations are the basis for so many unresolved patient complaints that mainstream medicine just isn't investigating

    I know 2 people with MTHFR mutation and double dose of B6 in the blood. Does it mean that it can’t be turned into active form and the body is in deficit? Do they have to supplement with the activated form? How to get rid of this blood excess? They don’t supplement.

    Glycine gave me serotonine syndrome. This activated the gene i think?
    I feel like im pouring out neurotransmitters and adrenaline constantly.
    Does this mean im an overmethylator??

From the Linus Pauling Institute: " Dietary folates exist predominantly in the polyglutamyl form (containing several glutamate residues), whereas folic acid-the synthetic vitamin form-is a monoglutamate, containing just one glutamate moiety. In addition, natural folates are reduced molecules, whereas folic acid is fully oxidized. These chemical differences have major implications for the bioavailability of the vitamin such that folic acid is considerably more bioavailable than naturally occurring food folates at equivalent intake levels.
      The intestinal absorption of dietary folates is a two-step process that involves the hydrolysis of folate polyglutamates to the corresponding monoglutamyl derivatives, followed by their transport into intestinal cells.     I was depression free for 4 years after supplementing b12 for deficiency and taking antidepressants. I travelled and my nutrition got worse and depression back. I had blood test and folate is deficiet. I started on 5mg folic acid, 2 days and Meds get back to function, they sedate me, and appitite back. But day 4, i had sleep problems and depression back with huge anxiety. Stopped folic acid after getting 8 pills 5mg in a week. So what should i do? Methylfolate also worsen depression before. When I will get better? And what i should do?

      I just recently found out I have homozygous c677t. I cannot tolerate b12. It's to stimulating. I could only imagine what methylfolate would do because I know I'm very deficient in both. But I've been doing a lot of research and you can fix methylation without taking b vitamins. Our bodies use 90% of our methylation to create creatine and phosphatidylcholine. So if you supplement with creatine and phosphatidylcholine/ betaine (which is a usable form of choline) you will reduce the amount of methylation you need.

    i have a thalassemia minor trait, but i'm a very active person, i 'm a runner and cyclist and have been taking b complex (syntetic folic acid).. 2-3 times per week for many years now and i do feel the benefit effects(more energy better recovery after training sessions). But recently i 've read about the active form of folic acid and its benefits. Now the question is :
    i if i do start taking all the vitamins B complex in the active form , Methyl Folate included , it's ok , while if i just take Methyl Folate alone it can be dangerous ? thanks a lot

      Either way it is not dangerous but some people get side effects. This is less common with b complex though

    Hi i have MTHFR A1298C Hetero mutation (found after recurrent miscarriage panel). I took Methylfolate for months but it gave me horrible horrible anxiety. Which supplement should I take?

      I am hetero for C677T, recently diagnosed. The way I understand it is that methyl B-12 is the key to activating the folate cycle. Methyl folate and methyl B-12 work together. That is what I know from literature. I am scheduled to see my doctor in 3 weeks and then I'll know more.

      You have been taking your methyl folate in the WRONG way. You need to take a methylation complex having All of the following.
      1. P5P - active B6
      2. Methylcobalamin - methyl B12
      3. 5MTHF - methyl folate
      4. TMG - anhydrous betaine
      5. R5P - riboflavin '5 - active B2
      You also need to some free form Glycine to support methylation buffers and Niacin / B3 to Balance any overmethylation if required (niacin steals methyl groups).
      Some Universal methylation requirements are only fulfilled by SAMe , for which you also need free form Methionine and zinc.
      Also Note that folate/5MTHF is Not the sole methylator. Other methylators are SAMe, TMG, Choline and Glycine. You need to balance methylation workloads between these. Further SAMe is a universal methylator for methylation of DNA, proteins, nucleuc acids, hormones, neurotransmitters, and the activation of transmethyl- X enzymes. Folate cannot do everything that SAMe can do and vice-versa.

      @matthewbrown1849 Wrong, methyl B12 does Not play any direct role in Folate cycle but bridges the folate and methionine cycles. It however plays a critical role in methionine/homocysteine cycle where the enzyme methionine synthase requires methyl B12 for its activation - its a methyl b12 dependent enzyme. This enzyme then facilitates the transfer of methyl group from folate to homocysteine so it can be converted to methionine/ SAMe.
      For reference : " The interrelationship between these two vitamins is best explained by the methyl trap hypothesis stating that vitamin B12 deficiency can lead to lowered levels of methionine synthetase, which results in a functional folate deficiency by trapping an increased proportion of folate as the 5-methyl derivative. "

    I've heard that someone corrected their daily awakenings after 5 hours of sleep by taking methylfolate. I have this problem but hesitate to try it because in the past methyl B12 made it hard to fall asleep and I don't know my MTHFR status either. Glycine, however, I did try, and it made sleep terrible. Now that you mentioned glycine connection, I'm even more hesitant to experiment with methylfolate. I'm taking B50, D3, B12 cyanocobalamin, choline. Do you have any thoughts about this, doc?       not sure why glycine would make your sleep worse. methylfolate can improve sleep at very small doses as it increase glycine in the body.

      @@gamayun1224 Glycine can also be stimulating for some people. And sorry, but the doc is wrong. Folate doesn't increase glycine. Eating glycine rich foods do. You may need B12 to help 'untrap' your folate.

      Hey @paulg6852 Gary is the real deal. We just finished the 10x genetic screen and got the results TODAY. I have THREE methylation gene breaks and without the supplements, I’m screwed. I learned none of this in my nutrition degree about a decade ago.

I guess I just would like to hear someone with medical knowledge have a good healthy debate with Gary, as he puts pretty much everything that can possibly go wrong with us down to these 5 genes.       Not SAMe!... but free form methionine. Methionine is the principal substrate for not only SAMe but also for endogenos cysteine, provided you pair it with a methylated B complex in the right ratio. A methylated B complex will have P5P, methylcobalamin and 5MTHF amoung other Bs.

    Hi, I have a question. Would taking Folinic acid avoid some of these issues? I took a DNA test and due to the SHTM1 gene (?) they recommend i take folinic acid and not methylfolate.

      Yes but if you have MTHFR sometimes it is not sufficient and will depend on your specific genetics and other environmental things.

    (I have anxiety, however it is mild and well managed). I've taken 15MG of L-Methylfolate for about a year now. Prescribed because I had "reduced folic Acid Conversion", per a Genesight test. A couple months ago I started having a spike in anxiety. First in my body, out of nowhere it'd just hit me.. then overall just high anxiety, unlike what I have had before. It occurred to me that maybe it was too much L-Methylfolate.. I looked around and found out that too much can cause these symptoms.. I stopped 5 days ago, and have literally felt better little by little each day. I feel like I could have possibly been deficient before (even though I didnt have any symptoms) but over time the supplement built up and gave me too much.

      This is likely the case and what happens too is that other b vitamins get depleted bc you are taking so much of the methylfolate.

      Just throwing out an update. My primary care Dr looked over my gene site test and recent blood work panel, and told me that I should never have been prescribed the L-methyl folate and to stop it immediately. He also said that he didn’t know why that previous dr (a psych / DO) would have Rx’d me that.
      It’s been 4 weeks now since I’ve stopped taking it. I feel so much better. Definitely going in the right direction. I’m so glad i figured this out and didn’t continue taking it, not knowing what was going on.

      What are you using now? Because I have mutation in mtfhr, and the doctor told me tô use methylfolate tô help the antipressants tô help working just for anxiety and social foby.

      Nothing. I wasn’t Rx’d it to help an antidepressant work. It was Rx’d by a psychiatrist simply because I have a minor mutation of MTFHR. My primary care doctor told me i should have never been prescribed it and that even if I have the mutation that doesn’t mean I don’t process folate.
      Methyfolate shouldn’t be Rx’d to anyone with anxiety or depression when not partnering it with an antidepressant to help it work. That’s right on the Deplin website. That’s what happened to me.

    Worst part about methylfolate is that people and advisers dont know anything about it, they just wrote that mthfr is needed for synthesis of methylfolate and they are teaching everyone that if you have mthfr then you need to suplement this nutrient. I really encourage to look at study that showed that 3mg of riboflavin lowered homocysteine by 40% in people with mthfr and this is really key nutrient for those people not methylfolate. Methlfolate is methylated god knows how many times everyday and dose that you get from suplement will be transformed into thf in like 1-2 hours after taking and then it isnt different than folic acid that is demonized. In the end i want to write that b2 is key nutrient because it speeds up mthfr and if you got polimorfism then your enzyme need more b2 and even more facinating is fact that such low dose as 3mg will be enough. If you dont notice any improvement in blood homocysteine or your mood then mthfr isnt your problem and you need to dig more into your biochemistry.

      Hi thanks for sharing. I agree riboflavin is very important. It seems for some more and others less. It makes sense that it would or could lower homocysteine but clinically I do not see it work that way (even at much much larger doses than what you noted).       @swintegrative riboflavin is Not critical for homocysteine regulation at all ! It is however important to take it in the right form - R5P to support folate cycle and GSH synthesis and recycling. The most critical elements of folate cycle are 5MTHF, P5P, R5P, and Glycine. The most critical elements of Methionine cycle are Methionine (free form), P5P, methyl-B12, 5MTHF, and Zinc. B2 should never be consumed in mega doses or in isolation.

      I have mthfr c667t gentically tested and I tried B2 in all doses and forms and did NOT experience any therapeutic benefits at all ! What immensely benefited me was the RIGHT FORM OF FOLATE - 5MTHF. I tried 5 brands of folate. METAFOLIN from Merck was the best and the only one that suited me. Quatrefolic or generic forms of folate were NOT good enough. And the synthetic Folic acid was a disaster !

      ​@@sgtaneja if you didnt get any benefits from b2 then there are 3 options, you could be taking b2 with others vitamins in b complex which wouldnt lead to significant results as they deplete each other, second option is that you have huge impairement in folate cycle which methylfolate wouldnt do much about(and you should be taking more choline to boost other pathway along with creatine to reduce demand for remathylation) and last option is that you didnt have problem with mthfr but just folate deficency

      @@swintegrative so if i'm getting a headache from taking 400mgc sublingual Methyl-Folate, you're saying it might be some bacterial overgrowth inside the body that the MF is acting as a growth factor for and that is what is inducing the headache? like internal organ microbiome? and if so, what might remedy that? could it be that I just need more glycine? or a lower dose? or taking it through the stomach might actually be better for me? because I'm taking b complex and I don't think I wanna be taking folic acid instead, and taking nothing seems wrong as I'm apparently deficient from taking the b complex without extra folates.

    How do you raise homocysteine? Also if you are b12 deficient and have low homocysteine, could the homocysteine of been depleted by high levels of methylfolate?

      If your not eating enough protein usually. It comes from the amino acid methionine. Sometimes it’s low from inflammation/ infections too

    Thats weird it decreases glycine. Because TMG and NAC are what make up glutathione, which is one of the reasons to take MTHF. is the glycine decreasing b/c its being converted into glutathione?

      These r good questions but you have a few things mixed up. Glutathione is made from glutamine, nac and glycine. Tmg is not needed. Folate is not necessary to make glycine but hold on to it.

      @@123private3 thats very hard to answer to be honest as everyone's body is different. you could either see results right away or it might take a few weeks of consistency. with homocysteine that high, definitely supplement with at least 400 mcg methyl folate per day.

    yes, my labs and whole blood levels came out like they’re fine but I know I cannot be fine because I don’t feel fine and I never eat meat, so how can I not to mention two decades ago I was very very deficient in vitamin B 12 that the doctor at the time 24 years ago told me I needed vitamin B 12 shots which I thought I was just young and stupid and never went and did it so something has got to have gone wrong because I am not well and no one can figure me out… I got a doctor to listen to me because I told them I listen to a lecture and they come to find out my homocysteine is elevated and but the MMA was fine when I take vitamin B complex or just the thiamine I feel awful my head pounds like there’s no tomorrow it’s not worth it it’s just not worth it but yet I can’t live like this either something is not right I am deficient somewhere. They are blaming my elevated homocysteine levels on heart disease and I say I got heart disease because I’m deficient somewhere and I know it’s a quick fix whatever’s wrong with me and I believe I can turn this around if I can just figure it out. I am so fatigued I can’t even work anymore. I am on disability I know I abuse this body all my life by working two and three jobs never sleeping just eating you know I candy bar and Starbucks for 25 years whatever and now I can barely get dressed. I’m so tired my skin is like thin it rips. I have horrible digestive problems with I’m always bloated with gas. I’m still postmenopausal for some reason going on 20 years with hot flashes and night sweats. I am so tired may God forgive me for abusing my body by not eating right I was a single mom and I always worked no excuse however, for the last three years, I ate very clean and bought a bike and try to exercise every day but I just don’t have it in me to even make something to eat. My kids bring me bring me over a plate of food. i’m not very smart when it comes to talking to my medical doctors, but I know somethings not absorbing somewhere. It’s like all the magnesium or all the potassium, or all the nutrients or the sunflower seeds or the blueberries that I eat it seems like the nutrients are in absorbing somewhere but yet my love say I’m fine. In fact, I even was overly find out a few vitamins it’s crazy it’s just absolutely crazy I just don’t trust doctors I don’t know why, but I just don’t trust them. Can you please guide me tell me what to do in kindergarten language. Thank you so much I appreciate your time and your efforts.

    Do you ever stop talking to breathe? Your talking incessantly without pause makes it hard to understand your meaning.
    I've taken it for 14 years and I feel great. It has helped me to wean myself off Zoloft. I would not be without it. The best -for me - is Solgar. Initially I took 800mcg, then increased my dose after stopping Zoloft 18 months ago. I now take about 1200 mcg per day. Without it I get tired.
