    I'm 41 and been taking NMN + Resveratrol intermittently, for almost 4 yrs. Keep in mind, I never took more than 500mg. Thorough out those years, I probably took around 240mg every day for about 4-5 days, break a couple days, then now, I am trying out 500mg NMN + 80mg Resveratrol and 500 TMG every other day. I take every other day because one...I'm fairly small framed - 5'3.5" and 95 lbs. and also, I work out often and don't want the resveratrol to interfere with that, so I always take NMN and resveratrol within an hour or so of each other (Rsver. with mixed nuts for absorption) after my cardio and lift sessions around 11am. Timing of that seems to be going well, and not affecting sleep too much.

    I been taking 250 mg in the morning with coffee for about a month. I do feel very good, high energy, alert, focused, very minimal aches or pains, sleep is good and I do not wake up feeling tired. but this could be from my healthy eating and almost daily 5 or more mile morning run.

    A series of 12 studies on a molecule called NMN (Nicotinamide Mononucleotide).
    The conclusions are the following:
    - NMN supplementation does not increase muscle NAD levels, which contradicts the theory that NMN supplementation can boost NAD levels.
    - older adults who exercise have similar muscle NAD levels to younger adults, suggesting that exercise is essential for maintaining NAD levels.
    - NMN supplementation did not show significant improvements in various measures of health and fitness, including body composition, blood sugar levels, insulin sensitivity, sleep quality, fatigue and exercise performance. Although NMN supplementation appears safe, its benefits are not as robust or compelling as some might expect based on the influencer hype surrounding NMN.
    Studies on NMN supplementation have shown mixed results. Most studies have shown no improvement in muscle performance, body composition, blood pressure, cholesterol, or insulin levels. In fact, an increase in insulin levels was seen in one study, which is not a desired outcome.
    Although NMN appears to be safe and increases blood NAD levels, it does not appear to improve muscle NAD. There is no improvement in VO2 max, strength, walking speed, sleep, fatigue, well-being, hearing, cognition or arterial stiffness.
    In conclusion, human studies of NMN have been largely disappointing. The benefits experienced by social media influencers praising NMN are likely the result of better diet, exercise, and adequate sleep-that is, a placebo effect.
    Additionally, there is evidence that NMN and NR supplements are broken down by our gut microbiome and not directly absorbed. Any small amount that is absorbed and enters our bloodstream is quickly broken down by our liver.
    So while you may choose to buy NMN supplements, you should know that the benefits may be limited.

      Well thank you! The way these influencers and educated people praise and Pretty much promoted as the holy grail for youth definitely could fool me! Actually it did. It’s so easy to just trust what these guys say And not even care to look for the research. It’s pretty sad it makes me wonder how much they’re getting paid people who spend thousands of dollars on these kinds of things just do to the hope alone. We are supposed to be able to trust these educated people. 🧐😑

      Speak for yourself and ofcourse if something is working for people "THEY" are always going to manufacture fake research papers contradicting all the good effects on people's health. I always say try it yourself regardless of what other say. So far PQQ has worked for Me just 10 minutes ago I bought liposomal nmn which is absorbed easier and liposomal resveratol. I'll try these 2 and see what happens. I'm sure it will have some positive effect.

    NMN makes me tired if I take in morning. A close friend told me the same thing. Late afternoon/early evening seems to work best for me. There's a Japanese study that seems to confirm this timing for older people.

      Hi Jay, yes, like you, I'm not too convinced about taking everything in the morning.
      There is the value of personal testing like what you did. I intend to do it too :)
      Have you tried taking 0.5 gram in the morning and 0.5 gram in the afternoon?

      I’m the same. Was getting sleepy during the day taking it in the morning. Way I look at it is, your bodies repair/upkeep/growth/renewal etc happens while you sleep. So providing mitochondria with “energy” prior to sleep has you get better quality “upkeep” going on. I find the same with Pemf treatment. “Energise” the adrenals, makes you sleepy, coz while you have more energy at a cellular level, your body is gonna wanna sleep to get all those things done. Cells having more energy isn’t the same as “feeling more energetic”. Dopamine will leave you feeling like you have more energy. In reality, 99% of people have more than enough “energy” on board (stored fat etc), it doesn’t mean they feel “energetic”. Just my thoughts and experience.

    I started taking RBS sublingual NMN under the tongue about a month and a half ago, it made me extremely groggy the first week and I almost stopped completely but I pushed through and by week two I wasn’t experiencing that anymore. I also take TMG, Resveratrol, Quercetin, and Spermidine. I’m going to add Berberine and NAC to my supp stack soon too.

    Hello Dr. Sinclair, is it true that if we take NMN, we also have to take TMG (trimethylglycine) as otherwise bad effects from methyl donor depletion occurs? Most of your videos do not mention TMG but i heard this by other specialists.

    Interesting again. So D. S. takes 1g NMN in the early morning. There is an interesting talk on the minimum dose of NMN. I hope people designing the next NMN trial will take it into account and and go over 250mg as it appears difficult to have significant results by staying at the minimum of dose and time.

    I wouldn't recommend taking it in the evening unless you intend to pull an all-nighter. Sometimes I have to work a 32 or even a 40 hours shift (intense intelectual work). After 24 hours usually brain fog would set in but this time I took NMN and lo and behold brain fog didn’t set and mental alertness was normal in even at the 40 hours mark. I took my usual dose of 750mg in the morning when I started my shift, and then 500mg after 16 hours, another 500mg after 24 hours, and finally another 500mg after 32 hours.

    Hi Rimon, can you please guide me? I implemented David Sinclair’s supplement 2023 with the only difference using berberine instead of metformin. Week 5 in I had an allergic reaction, a prickly, itchy red rash started on my hands, it spread up my arms, my face became red and swollen, from my eyebrows to the corner of my eyes went black. The redness and swelling was on my neck down onto the top of my chest. I stopped everything for nearly a week and everyday it slowly calmed down. These were purchased through donotage, so a good provider. Did I overload myself my taking everything at high levels straight away and should I have very slowly started these implementing them slowly one at a time. Your guidance would be greatly appreciated because for a time in the middle say within 2 weeks I felt fantastic…

    NR is the more efficient molecule because of its smaller molecular size offering direct cell entry and more Nad+ density per mass. And it is safer as Niagen has more extensive human studys and a more reliable supply chain. But NR is patented, so the NMN are manufactured in China and packaged in the US by numerous small firms without any patent infringement and the industry did massive marketing. Basically, people are buying less efficient and less reliable NMN simply because of its massive marketing budget.

    It seems that NMN raises the level of NAD and it's metabolites in blood.
    But why?
    I thought the model were that NMN would go into the cells, and there be converted into NAD+.
    Furthermore, that NAD+ can't enter the cells. So what use is it in the blood?
    It seems that the level of NMN doesn't increase in the blood. At least, no one has said it does.

    I'm really confused. Iv been taking nmn one capsual of around 250 for a few days, and it's made me feel sluggish and have to go bed by 10 (usually up until 1). Not sure if I should stop. I also have adhd which seems to have increased so may not be good for me.

      i only take nmn occasionally, but when i take it without magnesium glycinate I quickly feel disoriented somehow.

    I bought good quality NMN and took 125 mg daily for two weeks. Omg! My liver or gallbladder got progressively more painful or achey. As soon as I stopped the pain stopped. Is it just me?

      Hi Crystal, great question!!! I'm just making a video about it haha.
      In essence, for now, senescent cells, are basically healthy cells reaching "retirement age", are no longer useful, produce bad chemicals, and have lost their ability to die.
      Besides killing them, there is another strategy to help them/your body. You'll see it in that video. I think it will come in 2 weeks.

      HI Glenn, how are you? I've experimented with NMN and NR back and forth. Right now I'm not taking it , mainly for financial reasons - I can purchase both my parents and my wife's parents health & longevity supplements instead of 1gram NMN for myself.
      I care more about them right now.
      However, I'm about to do NMN experiment myself for 90 days and publish results on this channel.

      David said that you should buy brands that have GMP on the label, which stands for good manufacturing practices, he said that means it should be over 99% pure. There's a lot of fake nmn products out there.
      Reque by Science is the brand recommended by Andrew Huberman, it's where I buy mine from, it has GMP on the label.

      I watched in another video that it shouldn't be refrigerated if the room temperature is normal and stable. If it's too hot , put it in the fridge. In any case, what you shouldn't do is take it out of the fridge and back again. My suggestion is open the bottle, take the pill while keeping the bottle in the fridge, don't take it out and back.

      Tony Robbins and Peter Diamedis are stakeholders in the company LifeForce. They wrote a book about current and emerging science of longevity. The company has this year started selling supplements. One in particular is Pure Healthspan which has a number of ingredients that have been studied to help with age related health decline. Not in my price range at the moment though unfortunately.

      Remains to try Niacin, just to see if flushing is real!
      I guess that you have to be really deficient to feel an effect of a supplement.
      Or very susceptible to the placebo effect.

      There are conflicting studies on this, so probably it's worth testing how you feel with your body (energy, fatigue, sleep) based on different times. Especially morning vs afternoon.

      All medicines are based upon mice and other animal studies besides human trials. They use mice that have been genetically modified to be very close to human DNA in many ways. I'll gamble on the mice. You can avoid NMN and just save your money if you want.

      The secret is creating synergy between different molecules targeting all different longevity genes.
      Metformin and NMN work differently and are likely to have a synergetic effect. In the resveratrol series on this channel, I explain the different synergies between resveratrol, NMN (or other NAD+ boosters, NR, or niacin), and metformin. I believe this is why Sinclar takes all of them. Trying to cover all his bases.

      Metformin does not extend the life span of mice. David Sinclair gave mice NMN and they lived much longer than the typical mouse. I'll go with the mice and the NMN. You can stick with the metformin if you want to, but metformin will ruin your workout and make you weaker and have less stamina.
