    Beautifully explained thank you! I've been struggling with sample preparation for months now - I have VERY expensive triple-labelled samples for NMR so, naturally, sample stability has been a great concern to ensure we can get the most out of each prep.
    Have you got any rough kind of buffer optimisation "workflow" that you go through when determining protein solubility? I've thus far tried crude hanging drop precipitate screens using a crystallographers microscope, as well as a thermal denaturation assay utilising a hydrophobic residue-binding dye which infers protein unfolding using qPCR (ThermoFluor I believe is the technique name).
    Have you got any other suggestions? I'm still struggling somewhat with long term stability.

    Ammonium sulphate precipitation follows the procedure explained by you. It is the good and simple method instead of ultracentrifugation and other methods. Nicely explained. Continue.

    Great video! Just have a litte remark:
    At 1:52 you mark the carboxylate ion which is part of the protein peptide backbone. Instead you should have marked the carboxylate at the amino acid side chain which is exposed to the enviroment.

From your video, I understand that adding salt ions can make the structure of the protein stable along with reducing of the intermolecular replusion of the protein. Then, what is the mechanism of the water molecular can form the hydrate shell around by the protein?
      Is hydrogen bonding on the hydrophilic regions of the protein?
