Gonna guess that they were acting in the sense that they rehearsed it a few times and had probably at least a couple of directors telling them how to walk/carry themselves and what kind of feeling to emote.
What role did he play that was a villain? He was Sirius Black, Winston Churchill, Commissioner Gordon... The only one that comes to mind is a corrupt cop in Léon.

That's why they find a role that fits with their acting style. For example, I don't really think Dafoe is anything like the green goblin in real life but his style of acting works well with certain villainous characters. There are plenty of actors who do just play themselves but I wouldn't disrespect Dafoe who has put in the effort to be a good actor imo.
