    I would definitely break up the mushroom material giving more surface area for extraction as well as making it easier to agitate through the process. Def could have saved on the amount of alcohol use too. And in my experience it’s better to have a sealed vessel 1. So u can agitate daily or when u get to it and 2. Making sure the alcohol doesn’t evaporate as I leave mine outside rather than in the fridge. I also have always used the same material that was extracted through ethanol in the decoction step just making sure I get every medicinal compound extracted. I’ve also recently read that dual extraction is not always the way to go for certain fungi like lions mane, cordyceps, turkey tail. But chaga and Reishi will benefit from the ethanol for bc of their alcohol-soluble terpenoids.

      I agree. Smaller pieces = more surface area. I had some large pieces of Ganoderma Applanatum and I cut it into 1" strips and was still afraid it was not in small enough pieces. I also agree with the point of not using cordyceps or lion's mane in the tincture process. I thus far have tried ganoderma and lions mane in the tincture, and a fresh charge of lion's mane and cordyceps in the decoction along with the material from the tincture in a dual extraction process.

      Good additions to this process. So you are saying that you typically used the strained mushroom material that was used prior in the alcohol extract for your water based ‘decoction extract’? Rather than using a diferent batch of unextracted mushrooms for the water decoction and then combining? I’ve never done this before and trying to get that straight before I start. From what you are saying, its not necessary to use a different set of mushrooms for the water, perhaps since the water soluble compounds are still present in the mushrooms that went through the alcohol soak. Thanks for clarifying this for me!

    What you do by putting your alcohol extract in the fridge is reducing the extraction rate by a factor of 3 to 9.( A factor of 2,5 to 3 for every 10°C). At this alcohol level your extract is safe at room temperature and you get a better extraction in the same amount of time.
    Further it might be profitable for the extraction to cut or mill the mushrooms to small pieces, especially the reishi.

    Hi I do A/B extractions to great effect with cubes.. I use cracker dried samples in a dehydrator.. then powder them in a small blender or food processor for the greatest surface area before immersion in either liquid. I do reuse the fungal material as the A/B method of extraction is about getting all the compounds out of your initial sample. You run it through one process, strain (I use a Buchner funnel and vacuum flask), then run the sample thru the other process for a complete extraction..
    For my purposes, I use a weakly acidic water first (distilled water and lemon juice). I also add an anti-oxidant (vitiman c) to preserve potency.. I steep the powdered sample in the very warm slightly acidic water (I only use about an once or so lemon juice per L) and I also put it on a gentle stir as well.. as both the heat and agitation aid in the extraction. After several hours (covered so no evap), I then strain thru the Buchner funnel.. then I take the fungal material (setting aside the liquid extract) and then use the grain alcohol, no heat, little more vitiman c, and stir bar lightly for several hours. I then strain again using the Buchner funnel.. and discard the spent fungal mass.. then combine the two extracts and then boil off the alcohol (can only get rid of 95%, which is why grain alcohol is 190 proof as further distillation takes a special technique. Your final product will come out to be 5% abv but not noticable). You can boil off the alcohol as it's boiling point is below waters boiling point.. (or you can distill it off and recapture it for reuse).
    Depending on the compounds that you are trying to capture sensitivity to heat, you may have to make modifications regarding any heating.. it's fine for cubes. I get over a 95% extraction with no loss in potency for the active ingredient I am extracting. (the antioxidant helps and it's fragility to heat is a bit of a myth). I've directly sampled the spent sample material in large amounts with no note able effect at all (and no, my tolerance is low). I use the extract to make gummies.. and taking in my starting sample size of material weight (usually 2oz) divided by amount of gummies produced, I get a rough dosage of each gummy to equivalent dried material of my sample.. and they are roughly the same potency. If a gummy mathematically comes out to 2g each (dried equivalent) it does roughly match the real world dried sample amount in effect. No significant potency loss noticed.
    You are working with different compounds that you are trying to extract each with their own characteristics and sensitivities.. but this method that I adapted after a lot of research and experimentation works very well and perhaps can somehow be modified for your specific purposes.. tho heat really would be the biggest possible worry.. and oxidation.. some compounds are fragile but heat isn't essential, it just speeds up the extraction (as does the agitation and powdering due to increased surface area). Tho it is needed for removing the alcohol but perhaps a more gentle evap method might work..
    I hope to one day create the Stamets Stack incorporating also lion's mane and niacin into my own process as a personal daily suppliment for neurogenesis..
    Note: while somewhat descriptive in detail of my method, it's not a full detailed recipe/procedure in Tek format. This was conversational and off the top of my head on how I make extract. I might of left small stuff out. And to use it for gummies, there's a recipe for that as well. So, just take as such and not being totally complete. I'm also working with like 1-2 ounces at a time or more dried mush material. I just don't want people to attempt it, get it wrong because I didn't go into every detail and be disappointed.

      Dual extraction also isn't always recommended. It depends on the compounds and what you are trying to extract. It could be a redundant/unnecessary step.. you might do much better extracting each mushroom to it's own tailored process..
      And as stated in another comment, that's a LOT of alcohol, lol.. that's like a half years supply in one go for my usage, heh.

      Also, check out using a Soxhlet condenser for both the alcohol and water extractions. I know it works wonders with dried reshi.. it's a type of distillation process that repeats itself and when the evaporate starts running clear, you have a full extraction.

      I did some further research, Soxhlet might end up being overkill.. tho at first glance, the basically automated, low attention aspect looked very promising.. it just might end up being a bit of overkill.. it all depends on the solubility of the compounds.. Soxhlet is used more for lipids and sugars (it was designed for milk) but it has many uses.. for my specific use, it seems to be overkill due to the specific substance I am extracting being fairly easily soluble already.. but I am not fully ruling it out, as an actual organic chemist made encouraging remarks but that was in regards to creating a solid of the substance I am looking to extract.. I'm not actually looking to do that, tho of course am making a note for further research, hehe.
      there are some great mushroom tincture videos out there, I just watched one that used reshi doing a water and then alcohol extraction.. and saw another using both for Lion's Mane as well.. very informative and concise clear info with the science behind it.
      I took inspiration from those as well as organic chemistry and combined them to come up with my method...
      dried is often used as mushrooms are 91% water.. basically, their internal water content dilutes the final product, so starting with dried is actually better but in some cases, there are maybe some compounds that do not survive the drying process that you may want to extract but so far for lion's mane and reshi, it's common to dehydrate them first.. before an A/B extraction..
      the reason I boil off the alcohol, besides for easy concentration and reduction of volume is that there are many people with an aversion or addiction to alcohol.. many tinctures are fairly high in alcohol content 30% (60 proof!).. and while you cannot distill off all the alcohol completely, you can get 95%.. that last 5% would require you to break the water and alcohol azeotrope and that's complicated to do compared to simple fractional distillation.. (or evaporation/boiling off).. also I use the extract tincture in gummies, it is the actual water that I use to make my gummy base.. an alcohol solution will not allow it to set (5% is negligible in this case, you can't even really taste it in the water). so I remove all (well, 95%) of the alcohol.. by either fractional distillation (thus recovering all the alcohol for reuse) or simply boiling it off at it's lower than water's boiling temp (water is 212, ethanol is 173); the "good stuff" remains in the water, the whole extraction is concentrated and the overall volume is basically halved..
      but if you are looking into making specific supplements commercially, you may want to consult a proper organic chemist for bulk methodology..
      you can also come up with a pure powder of each compound, using a rotary evaporator then make mixes..
      but generally, for where you are at..
      start with the same sample and run it through the two processes.. that's how the a/b extraction process is designed.. also, starting with dried, while not essential, is usually a good idea.. but definitely chop it up finely as possible.. powder it if able. the more surface area the better, the faster and more complete the extraction as this is down to the cellular level.. breaking apart the cell walls..
      also, straining.. getting rid of the spent material.. chitin, etc.. which is indigestible.. you just want the goodness.
      a tincture press, to really squeeze out all the liquid after extraction or in my case, since I powder my dried material.. I use a buchner funnel and vacuum pump.. I get a nice dry little cake.. (use the proper filter type, there are two for quantity (one strain and done) and qualitive (for reusing the material, easier to separate the filter disc from the material).. I think I got that right..
      but either way, extractions are fun as is learning about them.. enjoy!

      I suppose the most simple method of all would be to just dehydrate the mushrooms and then finely powder them.. then mix them together in various ratios.. tho you wouldn't get any concentration.. and it may take a lot of dried mushroom to get to a proper daily dosage of any specific wanted compound.. so, might not be cost effective or really user friendly if you say had to ingest many many grams of a supplement just to get the proper daily dosage... also, quality and dosage control could be a factor as there would be a natural variance of the substances and while you can get a reasonable homogenization via a powder but it may vary greatly by batch depending on genetic and environmental issues.. if you isolated the compounds, you'd have much more specific control of dosage, concentration, serving size, quality control, etc..
      a lot of factors to consider.. at least for a commercial minded project.. for a more limited artesian or personal one.. not so much..

    I dry it out, then Grind it to a fine powder then use it for water or alcohol… works way better (surface area) and you maximize the mushrooms, it does however look cool the way you did it, like some jar in Frankenstein laboratory, but not efficient, cheers!

      Is there a simpler way to make the mushroom bio available. I dont care for a ticture or extract, I just want the easiest way to convert Loins mane and Turkey Tail into a health benefiting product. No extraction or concentration.
      Mince in alcohol and leave to stand, Boil product (heat breaks down Chitin and cooks off alcohol)?
      Am I close?

      I have some Powdered that I am filling into capsules bit want to do a double extraction also can you use powder? Then strain? I was thinking of simmer in water as I do have some not Powdered then strain and sit in alcohol then strain again as water extract some different benefits I was told

    I can confirm that all compounds are present besides the glucans. The molecular weight is too large on them for my mass specs. You will need someone with a ms-TOF

    Well that was an interesting video, seeing as you are usually very meticulous, that was really ... different to see you just YOLO it.
    As others have said, I would have blended the material and then later filtered it through a cheesecloth.
    You just want enough ethanol to cover the mushrooms, so I would have used like a plate on top to keep them from floating.
    Do you intent to evaporate all the alcohol or do you have the mother of all parties coming up ? With the punch to end all punchs ?
    Also mechanical agitation with a magnetic stirrer could not hurt.

    Would tossing the mushrooms in a powerful blender like a vitamix with the water/ethanol help to extract more of the compounds? Way more surface area. Then, would taking the extracted matter, squeezing out the matter and re-extracting in the opposite solvent, that is water to ethanol, ethanol to water, get you even more of those compounds? Could you take those tinctures, and evaporate them into concentrates? Even down to a powder? Questions for science.

    Have you done an update video from this? Are you producing them for market yet?
    I've done herbal dual extractions and in order to get a universal dosage, the herbs should be weighed at harvest, then dried out and extracted from the same amount of 80% alcohol by weight, so thecmethod you're doing here is new to me. 190 proof alcohol with no measured weight or drying out of the mushrooms, I'm curious because I esteem you to be quite intelligent and scientifically minded to search out information and I've learned a lot from you

      Also wanted to add that I've heard many mushroom compounds take 1-3 months to extract and that when they are processed into smaller particles it makes more surface area available, which increases the overall potency of the tincture as well as reduces the amount of time necessary to do so.

    Please consider replacing that spigot with either a wooden, or food grade plastic one. That "silver" paint will eventually flake off and who knows what chemicals it's leaching into your tincture. FYI
    Also less alcohol and chop/blend mushrooms for better/stronger results.

    This may not be the optimal extraction process but it's a really cool mushroom display for a dispensary or health food store. I like smaller pieces but not ground ( for most mushrooms) . I find it difficult to remove the liquid from ground material. Also the material to alcohol ratio is to high. The good stuffs in the liquid so if you're using less you're getting less medicine and it's not nesesarily more concentrated.

      thanks for the feedback! this was definitely for the optics we got it tested for potency and food safety and it came back with great results but our next batch will be better with smaller pieces/more surface area

    Grinding will dramatically improve yield. Reishi is a tough bugger. It will spin with The blade of a blender or Cuisinart and gouge pieces of plastic off. You need a grinder with a metal bowl. Something like this listing on Amazon. “CGOLDENWALL Safety Upgraded Electric Grain Grinder Mill High-speed Spice Herb Mill Commercial powder machine Cereals Grinder CE 110V (700g Hand-held Type)”. But you need to watch the motor amperage to be sure you have an electrical supply that can handle the grinder.

    You may be wasting some medicine by extracting separately. Some compounds can only be extracted with hot water and some only with alcohol. When I do chaga for instance I do a hot water extraction first as it breaks down the chitin walls and allows the alcohol extraction to work more effectively. Also , be warned reishi should be brewed outdoors , it reeks! Maybe lucidum isn't as strong as tsugae but I imagine it's similar.

    It's just labeled for tinctures (marketing)....but there is no difference than any other 190 proof grain alcohol, buy the cheapest you can afford.
    You really could have used way less alcohol for a more potent tincture, and saved money.

    I noticed a vid with doubleblind.. something about mushrooms explained.. where paul stamets says the lions mane fruiting body reduces neurogenesis.. an that you need the mycelium. So.. that got me totally confused.. what are your thoughts on that?

    Dry them. Then grind them. Then soak or homogenize with a sonic extractor. Then low heat (100°)in a vacuum to concentrate and remove most of the alcohol. Cut with the water extraction to 50%. You can add a bit of honey to thicken and take the edge off. Enjoy.

    this is the least effort harvest of a reishi i have ever seen. i was so confused until i realised you just let the thing fruit through the filter, and realised the bag hasn't even been opened.
    id still personally make use of the stipes, but still, that snap harvest was dope.

    Apologies, but you have this process all wrong. You need to maximise the surface area of the mushroom by blending it and finely chopping. I always dry my mushrooms first, then powder them, you get a much better extraction this way. Never refrigerate your alcohol. It just slows everything down. Also you need to leave the mushrooms in the alcohol for at least 4-6 weeks. I leave them for 2 months. You must measure your alcohol content at the end as well, so you know how much water extraction to add. Too little alcohol and your tincture will degrade quickly. Too much and it will overpowering. Hope this helps.

    I would definitely run everything through the blender. It is only common sense to expose the most surface, the most particles to the alcohol. Then let it sit.
    Then do the same to the ones you do with water.
    I simply make mushroom gravy with my LM, and I run it through the blender first in water, before cooking it.
    Then when cooked add cream and flour as required, to make gravy. Then have a tablespoon or two, with my meals.
    Found it very helpful for focus, clarity, and healing old trauma (PTSD).
    Plus as much beef as possible. Only beef. Listen to Mikaila Petersen's story on how she healed with an all beef diet.
    Beef, salt and water. And her dad's story (Jordon) too.
    I can't do all beef, so I have some potatoes, homemade sourdough bread, cheese, and Kefir. Some desserts and fruit juices.
    and lots of water. Half ounce per lb of body weight per day, spread through out day.
    Works for me.
    Vegs, grains and all other foods became toxic to me.

    This way of making tincture is wastу of mushrooms! So big chunks can not be soaked well, so you can't get good extraction. The only way to get a good extraction is to pour with vodka a fine powder. Again, powder!

    Might make a good "screwdriver"....with a little OJ, or 5 Alive.
    Failing that...you might try grinding the DRY materials, and then refluxing with the alcohol. The lab glass you'll need wouldn't cost too much, and it's a very simple process. Many videos, here on YT, will show you how to set up for reflux, and then you could also concentrate your tincture VIA simple distillation, as long as the compounds you're extracting won't degrade with the temperatures involved. Closely related is "soxlet extraction"...basically refluxing the solvent through the materials, rather than having the same materials in the boiling flask (basically like a coffee percolator).
    I just did a rudimentary search, and it seems that Cordycepin and Adenosine can be extracted with 50% ethanol at 65C for 6 hours, and can indeed be concentrated by removing the solvent under vacuum. Good news...this means you won't have to reflux it.
    Seems as though a "dual extract" is recommended for both the Lion's Mane, and Reishi mushrooms. In the Reishi, the Ganoderic acid, is extracted, with other triterpenes, in alcohol for several weeks. A water extraction is used to extract beta-glucans, by simmering for several hours. These are then combined.

      Thanks for all the suggestions - this video was not the best method but we are getting them tested as a baseline for improvement - they are undergoing shelf stability testing over the course of a year to see how stable the compounds are, then we will see how to increase the potency.     Waaay too much alcohol.. and if you put it together with the water-based extraction that is - again - way too much liquid, (enough for many years to come?!) diluting the ingredients unnecessarily. If you are using separate the mushrooms for the alcohol and separate for the water extraction that is just wasting the mushrooms. (meaning after the alcohol-based extraction, the water based materials are still there in the mushrooms). 10 days of alcohol extraction time - in my opinion - I am not sure is enough. Grinding the mushrooms for the "bigger extraction surface area" is also advisable here, way too big portions. Putting the extraction into the fridge is also not a good idea. Btw, your alcohol is going to escape from that jar, that is the worst type of lid that you have for this kind of job.
