How to make: Homemade agar plates

  • Make your own basic agar plate using easy to find ingredients and see what bacteria and fungi you can end up growing!
    I'll show you how I make my own homemade agar style microbiology plates out of ingredients and appliances that you will probably already own in your own kitchen! Failing that, here is a way to make use of that old jar of Marmite that no-one seems to like

    Great info and explained in a nice straight simple way! Although I feel like a little disclaimer of being sterile and using more sterile practices when prepping these should of been said. Contamination is the #1 enemy when working with microorganisms especially in the field of mycology (Fungi/Mushrooms). All the same threw out a 👍and a sub. Interetsed in seeing your future vids!

    If i had seen this video before, our teacher said that we had to grow bacterias just using unflavoured gelatin, but that didn't work correctly.

      Right? But every channel has a slight variation of the Agar Plate recipe. Im wondering if different growths require different recipes of agar and why? No one explains the WHY of each ingredient. ie salt, yeast, bullion, sugar etc.

    Thank you for making this. I literally Just mix all the ingredients (I only had regular yeast, but I read in the comment section that you said it will work just fine) in a mason jar and now it's in my pressure cooker to sterilize. I'm going to test my lion's mane mushroom liquid culture on it. Jam on!

    Some sources suggest to use agar agar powder alone and boil it in sterile water - I tried this, but it turned out a clear but cloudy colour and caused my sister’s science fair project to fail. Other sources are similar to your method, which turn out yellow. I’m confused on the difference and why salt and yeast extract are used for some but not others.

    I don't understand how you can have uncontaminated dishes without sterilizing with heat and working in a sterile environment like a SAB.

      You are right, a sterile environment is best to prevent contamination. Microwaving the gelatine mix keeps the temperature close to boiling, which is the heat treatment you are talking about. You can create a sterile environment by using a camping stove if needs be, it works in the same way as a bunsen burner and should keep a sterile environment around the burner. Wiping the surface with a bleach or high percentage alcohol will keep the surface sterile too in most cases.
      Limiting the amount of time the plate is open will also help prevent contamination.
      Like with anything, having the right controls are essential, that's why I did the "bud control plate" which will help estimate the background cells counts and types from the air and the bud.
      This isn't meant to be a fully scientific analytical technique video, more something you can do as an amateur at home, or to introduce kids to all of the bugs that you can't see.
      Again, you are correct about the contamination issue, but these plates aren't to be used in a clinical environment but a home environment, so sterile technique isn't always possible.

    Pouring of the agar should be done inside a SAB or in front of a flow hood. Otherwise you will contaminate your dishes.

    Nicely done Dr.Chobbsy. Can you tell me if it is possible to use ordinary tap water, instead of bottled water, and if I would need to add anything to compensate for the added chlorine. I know that there is stuff available for fish tank water. Great vid, as usual.

      Glad you liked the video!
      It should be possible to use tap water, but it depends on the water regulations for your supply area. Chlorine is used as a disinfectant so it would be harmful for many of the micro-organisms, but it doesn't mean that in the large quantities that you would to potentially want to grow on a plate that nothing will grow. I believe that I live in an area that has a low level of chlorination, so I might not be able to tell is there is a difference here. The additives that you can get for fish tank water could also have some adverse effect for microbial life, so it might be worth staying away from those.
      Bottled water would keep remove this issue, as well as keep the mineral content as consistent as possible. Not to mention that for people living in hard water areas, it will cut down on the amount of limescale build up.
      The only thing i can say is to try it, and if it works for you, great!

    This video is certainly helpful for my research project! But may I ask? I have noticed that closing the lid prematurely causes my plate to evaporate water, which I only noticed AFTER swabbing the bacteria-containing cotton swab on the plate. Is it normal for the plates, after being contaminated, to be stored upside down? or do scientists normally store them upward without evaporation? Thank you so much

    I want to make agar plates at home and see that how many bacterias exist on objects around us. For example, i wanna see how much bacteria will grow on agar plate from a door handle, how can i do that? Should i use a cotton ball or something like that?will it even work?

    hello, your recipe looks great HOWEVER, i cannot buy yeast extract, it is just not available anywhere in my country, i can buy yeast, which looks like a bar of butter, it is completely solid. can i replace the yeast extract with something else like honey for example if yes should add the same amount like u adding yeast that is using your proportions but with honey?

      Vegemite should be OK. It's not something that we typically have in the UK, but it looks like it is yeast extract based.

      Hi, the easiest way would be to gently heat it in a microwave. Each microwave will be different, so you will need to use trial and error. I would start with a defrost or 50% power setting for a minute or two and see if it starts to melt.
      Remember to top it up with water back to your original volume (before microwaving) as it will lose some as steam as it heats up

      Marmite is dead, it only contains nutrients while bakers yeast contains dormant or alive cells.

    Hi Dr chobbsy . I enjoyed this video ,I would like to do an experiment with this what can I use other then yeast, that’s safe at home .and can we see a yeast colony under a microscope. I’m not sure what a colony is .does this mean that the yeast has grown into separate family groups.

      Hi, Glad you enjoyed the video!
      Yeast is the easiest thing to grow, and works well for beginners. What you could do is use the cotton bud to swab your phone... either on the screen or the buttons, as there is a chance that there will be something there. As for for it being safe? Well if it's there and hasn't caused you to fall ill, then it's likely to be safe.
      As for a colony, on the close up of the plate, you can see some small circular patches of cell growth. These individual circles are known as colonies. They are the result of on single cell being separated from the rest and dividing in that one spot. These can't really be seen under a microscope as they are rather large and the individual cells that make them normally have to be stained to be visible.

      Hey at first thougth i think this will work fine, gelatin melt at 35 C. but the added ingrediense will increase the boilding point. Such as salt. also works the other way around and make snow melt at lower temp.

      I think agar is the better solution as it is vegetarian made from seaweed or something like that where as gelatin is animal made product, orchid seeds best on agar I think with the right nutrient balances for growth and roots , search you tube on tissue culture , there are amazing videos out there for seed and tissue growing with orchids and other plants and fungi that might interest you , good luck .

      Hi, sorry for the long time to reply, I didn't get a notification of your comment.
      Anything that sets in the same way as gelatine should work OK. There are vegetarian gelatine options available now (such as the one I used). Agar powder itself is also an option which you should be able to find on an online retailer.

      You can use it to grow bacteria and fungi that is on your phone, and you can use yeast extract powder instead of paste. We use it to make plates and growth media in the lab, adding roughly half as much yeast extract as salt.
      The extract shouldn't contain live yeast and shouldn't grow on the plate. If you use dried yeast (such as baking yeast) you may get cells surviving in the gelatine mix and growing on your plate instead of the things you want to grow.

    How peculiar! There is no autoclaving nor pressure cooking required, and there is no contaminant without still air box, glove box, or flow hood! How is that even possible?! Probably the culture medium is devoid of sugar. It is amazing Saccharomyces cerevisiae could grow without fermentable carbon source!
    About weighing liquid, I have not the slightest idea why it is 'good luck'. I weigh liquids frequently and it is easily done. As for sticky stuff, first pre-weigh spatula, tare the weighing scale, pick up sticky stuff such as honey or that yeast extract, and Bob's your uncle! It is science. Not magic. Not luck.
