I like your idea for finding the bio samples at the spawn points. Makes a lot more sense.

Regarding what you said about the biosamples Zee, isn't it pretty simple to houserule ? I would do something like what you proposed, meaning i would begin with characters empty handed and then when they've found a lair/spawn point or something like that (i don't know exactly, haven't played the game) then either they immediately acquire a sample or the players roll to see if they can extract a sample intact or if they've all been destroyed and they have to search for the next spawn point. Also, i suppose the reason you go into the cave already loaded with the biosample cards is because you carry the empty cases to store them into, so you are encumbered because they are heavy and take up at least one arm to carry, hence the minus in the die roll.

I love the idea of picking up samples in areas going into the cave getting the samples then getting out. However that is the annihilation mechanic.
I think the story narrative behind this one is that like the original game you start trapped in the cave. Except this time instead of merely trying to get out you have to get out with the samples collected. Like your weyland yutani operatives trying to get out to bring alien samples to the corporation. You just join the mission half way through. On the extraction half. Let's say the first part was a cake walk they moved into the area got the sample then boom cave collapse mission begin.
It might be fun to play annihilation and extraction back to back. Saying the bomb tiles are where you collect the samples. As soon as you have all three re rack the board and play extraction with your guys

What did you do with Sam? I always new you and Tom would make him "disappear".

Can one combine two expansions in a game? For example, adding Investigation to the normal game mode and Anihilation/Extraction as the main quest itself...?

The leapers were the best part of this expansion for me by far. The Mercenary is okay but not the most imaginative or unique - basically someone else to do Bodyguard's job so he's not needed all the time. The sanctuary tile is kind of cool, but it's just 1 tile and once you move past it, it's no longer really relevant for the rest of the game.
The Biosamples are just poorly done. We miss out on the mechanics of actually harvesting them, because that was the bomb mechanic for Annihilation... at least let us drop the samples as a token on a tile to pick up later so we aren't stuck with -1 all game.
The balance of this mode shifts things in favor of Geologist, as he can hold the Sanctuary and make huge plays with it when horror/leaper despawns are desparately needed. Then basically it just shifts balance in favor of any caver who's ability allows them to avoid some form of die roll, like Climber. I also think Mercenary is stronger than Bodyguard specifically for this expansion, or whenever Leapers are in play, as the increased numbers favor setting up a Fortify as opposed to individually Repelling them.
