Toranaga was thinking that he would have to kill Buntaro very quickly if he found proof that Blackthorne and Mariko were having an affair because they are more important to him than Buntaro is

Mariko wants Buntaro to kill her, but she doesn't want to be in any type of union with him.
She doesn't want to unite with him in death.
She wants to die and be free of him.

She's not lying. She would gladly let him kill her if it means she doesn't have to be married to him anymore. She says it numerous times that her father chose her husband poorly and that Bontaru is not ambitious enough to give her the life she wants. And to be clear, Bontaru ASKS her to die with him he DOES NOT COMMAND HER. He gave her the CHOICE because despite how cold and distant she is to him, he genuinely does love her and wants their marriage to work. Toranaga later states that yes, it was a bad marriage but a necessary one because Mariko's father chose Bontaru not out of status or personal merit, but because he was a soldier and could keep her safe but was also far enough from their home where she would not be prosecuted/killed for her family's eventual betrayal.
