I am not Deaf, but I enjoy using captions. I would have fixed the captions on this, but I am not able to change the automatic ones. Instead I am just going to leave my version as a comment.
Moshe Kasher: I was on the television show on showtime's television show Shameless. Wonderful show
    Pete Holmes: Yes
Moshe Kasher: and I got um...
Audience cheers and Moshe Kasher acknowledges them.
Moshe Kasher: I mean this was a while ago, but it was exciting to get the audition, because like my parents are Deaf. As you know, both my parents are Deaf. They're super into hip hop.
    Audience: laughs Moshe continues talking
Moshe Kasher: And like I was like... but that's true. My parents are both Deaf, so I'm fluent in sign language. I grew up like and um... and I got the role for this this character he was... The character's name was Ruben. He was a Deaf, crack addicted, uh paraplegic, former gay prostitute with AIDS, and I thought, "Perfect!"
    Everyone else: laughs
Moshe Kasher: I thought, "If I don't get this role, there's something wrong with the Hollywood system. I was like I got this role for sure, because I know sign language. There's no way anyone's gonna beat me out for this role. I go to the audition room. I sit down. Its like ALLLLL Deaf actors. Everybody else is a real Deafmen.
    Audience: laughs
Moshe Kasher: Like their all the real thing. You know what I mean?
Pete Holmes: Yes (Trying not to laugh)
Moshe Kasher: It's like if you got a call to be in like Amistad, and you were like, "Yeah!" as a white boy. "I'm just really down with hip hop! I'll get this role!" and then you get there and everybody's like: "Not here."
    Audience: laughs
Moshe Kasher: So I go in and I just I was like, "Oh no!" cause I thought about it. I went in did the audition. Killed it. No, but I got the role. I just felt like...guilty, because just think about it you know. Like here you are. You're a Deaf actor. You make the move to Hollywood to really commit to acting. Already like a pretty difficult, questionable life choice. You know what I mean? Like already tough, and all you ever get is these auditions for things like Sesame Street and Touched by an Angel, and Highway to Heaven and its been off the air for 25 years, and finally you get a role for a showtime show, and you're like, "Something meaningful!" And then this hearing asshole walks out of the room like, "Know this sign everybody?" (1:56 Kasher gives everyone the bird as he speaks). I felt I felt bad, but I did my time in the trenches. I mean my parents are Deaf. I was a sign language interpreter. I...
Pete Holmes: You're an honorary Deaf.
Moshe Kasher: I am! I mean... People are getting weird right now. I don't know why. You this audience literally has no idea if this is true, but I am. I'm like an official. Hearing people with Deaf parents are like sorta like kinda like like Deaf. You know what I mean? Like? (Holds hands up and wiggles fingers while talking 2:17-2:19).
Pete Holmes: Is that the sign for justifying why you got the role? (Holds hands up and wiggles fingers 2:22. It makes me think of the sign for flirt.)
Moshe Kasher: laughs (Copies Pete Holmes' made up sign 2:25). But I was an interpreter for a long time I mean... I wanted to... Actually there's a funny... I thought I would... I wanted to show you some of my favorite signs.
    Pete Holmes: Please
Moshe Kasher: Just to prove to the audience. It feels weird right now.
    Pete Holmes: Yeah
Moshe Kasher: but I'm the real thing.
    Pete Holmes: Yes, I wanna...
Moshe Kasher: So my name sign. Alright sorry everybody has a name sign. I thought maybe I'd give you one at one point.
    Pete Holmes: Uh huh
Moshe Kasher: But its "M". This is "M" (Makes "M" with right hand and points to it with his left hand 2:45). "M" for Moshe.
Pete Holmes: Looks like you're starting a puppet show.
    Moshe Kasher: Again very disrespectful.
    Pete Holmes: Laughs
Moshe Kasher: That's M for Moshe, so my mom of course gave me my name sign, so she knew that I would have a big ol dick. Like she knew. (Puts the M on his leg near the knee 3:02).
    Audience: Laughs
Moshe Kasher: No, it's not. Its M for smart. (Signs his name sign and smart 3:05).
    Pete Holmes: Its up here.
    Moshe Kasher: Yeah
Pete Holmes: Ah so she goes like that? (Brings the M down from his head almost as if trying to make it become "You're welcome." 3:08).
Moshe Kasher: No, no so she's not a marine.
    Pete Holmes: Laughs
Moshe Kasher: She was in the Royal Navy for the grand gaymen islands, but she's not a marine.
    Pete Holmes: You're comeback city.
Moshe Kasher: Welcome to town. I just want to show you some cool ones. This is my favorite. This is reflection. (spells reflection with both hands right hand on top of left 3:25). R-E-F-L-E-C-T-I-O-N reflection right
Pete Holmes: Really couldn't it just be anything as long as you're doing it the same? (makes random movements with hands 3:32).
    Audience: laughs
Moshe Kasher: Thanks dude. (picks up coffee cup and starts to walk away 3:34. Walks back a few minutes later.) I'm just kidding. (Hugs Pete and gives small wave to the audience 3:43).
Pete Holmes: Isn't this hungry? (signs a mix of horny and hungry going up and down 3:45).
Moshe Kasher: Well that's that's actually horny. This is hungry on the stomach. (Signs basically the same thing much lower 3:52).
Pete Holmes: Horny? ( Signs with C hand on the chest moving up and down 3:53).
Moshe Kasher: Horny, but you've gotta do the face like (Signs horny with facial expression at 3:55 with the C hand on the upper chest. Pete Holmes also makes an exaggerated facial expression while signing the same thing.)
(4:00/0:51 Pete Holmes opens his mouth)
Moshe Kasher: No, no, no, no, no... no, no, no That's I had a stroke. That's completely different.
Pete Holmes: We have time for one question. (Looks down at phone until people start to laugh 4:16) You're a ladies man. What is your ratio of breaking up to being broken up with?
Moshe Kasher: You want to try that question again?
    Pete Holmes: No
    Moshe Kasher: What um
Pete Holmes: I communicated it well.
Moshe Kasher: My ratio of being broken up with. How many women have I broken up with?
Pete Holmes: As opposed to it being dumped.
Moshe Kasher: Ohhh being... oh, I don't know. I don't know what is that?
Pete Holmes: You've never been dumped?!
Moshe Kasher: taps his leg. where he first said his name sign was earlier in the interview. 4:41
    Pete Holmes: Laughs

I've seen him perform a bunch of times and bring up the "honorary deaf thing" as a side note kind of deal. What makes the delivery of the joke so good is he literally can speak sign language, and you wouldn't think otherwise, because he doesn't actually sign out full words in his performances, he's way more humble about it. He does what most of us would do and makes some nonsense gesture, then, months later after hearing the joke, you stumble upon this video or another like it, and find out he isn't bullshitting about it lol.
