  • Determination of Crude Fat content is a common proximate analysis. This parameter is very important for the analysis of food and feed samples. This video represents the detailed procedure of Crude Fat analysis using Soxhlet Apparatus according to the Method of AOAC 2003.05.

    Hi, thanks for the nice video, it is very illustrative. Nevertheless, I have a question: the method actually calls for the Randall modification of the standard Soxhlet extraction to perform the assay, which implies submerging the sample into boiling solvent, (in this case n-hexane). Is it correct to perform the assay with a regular Soxhlet extractor, as shown in the video? Any comment will be helpful. Thanks!!!

    Thanks for the video. Would like to ask isn't the weighing of the sample actually including the weight of a paper-made thimble even though you tared the weight of the thimble prior to weighing?

      No. If you tare the thimble weight, then the thimble weight will not be counted. Only sample weight will show in the balance machine

      Please use better grade of petroleum ether from a reliable source. I hope your problem will be solved.

      Mahmood Hashem, Set the temperature in such a way that facilitate the reflux rate of 5 drops per second. Usually, 100-150 degree Celsius is used for the Fat extraction. Than you for being with us.

      @@MahmoodHashem Thanks for your second question. In case of hot plate, it will heat only flat bottom part of the flask and only Hexane of the bottom layer will be evaporated at a particular time. But in the soxhlet apparatus, it heats on the maximum part of the flask, as a result evaporation of Hexane from all side of the flask will occur which is much effective way for Fat extraction.
      So, if you don't have Soxhlet Apparatus facilities, you can use Hot Plate temporarily but it is not recommended for permanent use.

      Please take a look at the principle of fat extraction of a solid sample. Normally a dried, ground sample is extracted with n-Hexan which dissolves fats and oils of the sample. During extraction, in the Soxhlet Apparatus, n-Hexane dissolves fats and oils of the sample and carries into the Round bottom flask. In this way, the extraction of fat continues for 6 hours by refluxing the n-Hexane (Oil free n-Hexane goes up by evaporating and after condensation it falls on the samples and get down to the flask carrying fats & oils from the sample). After 6 hours, the n-Hexane is then evaporated from the flask to separate it from the oil. The resulting residue (Fats & Oils) is weighed and referred to as ether extract or crude fat.
      So there is no need to use hydrochloric acid (HCl) in this fat extraction procedure.

      Don't use diethyl ether in soxhlet extraction. You can use petroleum ether replacing the n-hexane

      @Raghaban619, You can not evaporate the moisture and remained n-Hexane completely by keeping the flask at room temperature. Because n-Hexane and moisture will remain in the mixture form with the oil content in the flask. You must apply heat to remove misture and n-Hexane from the flask completely.

      - Weigh in the original sample, dry it and then extract it or,

      @@SourodiptoOfficial First, extract the fat from your sample using the Soxhlet Apparatus. Then evaporate the solvent using Rotary Evaporator. Finally you will get pure Fat in the flask. Now take wright and calculate the fat% as done in this video.

      1. Whether the fat is collected in the round bottom flask, or certain portions are in the extractor too, if I have to vacuum evaporate it ,do I need to mix the both the portions, ( the Solvent in the RB as well as in the extractor)
      2. What's the quantity of solvent taken for 250 ml RB ( as shown in the video), can I perform the same analysis if I have 500 ml Soxhlet extractor ,in that case what would be the amount of solvent.

      @@SourodiptoOfficial Fat is only collected in the flask. So you have to dry & consider the flask only.
      Secondly, there is no specific amount of solvent to be added. Amount of solvent should be enough to carry out the continuous reflux through the Soxhlet. You can get idea of amount of solvent from this video. So add solvent in your 500ml Soxhlet accordingly which is enough to recycle/reflux smoothly.

    I think that the weight of sample=weight of sample+ thimble - weight of thimble=4.7152-1.9177=2.7975 Right?

      No Sir. Sample weight should be 4.7152. Because we tared (made zero) of the thimble weight on the banace machine before taking the sample in it. So, the thimble weight will not be considered.

      @@MicroChemsExperiments I'm using right now ethanol and water as solvent. Can I do with light weight oil and ethanol as solvent for extraction..I want oil soluble extracts or extraction in oil. If so which oil would be best. Kindly suggest. I'm mostly into native siddha ayurveda formulations.

      @@MicroChemsExperiments we generally soak the herbs for 12hrs in water alcohol mixture. And then next day make extract in soxhlet extractor. And the extraction is incorporated into base oils as per body type and heated on low temp until moisture evaporate. And the result is medicated oil. Where each herb has different weights.

      Before incorporating herbal liquid extract into oil , oil is heated separately until one froth. Then only we add liquid herbal extract. This step is very important. So I was thinking can I add oil and alcohol as solvent n do extraction in soxhlet

      @@theasoapsacademy Thank you for your query. If you want to go for oil extraction, you can use Organic Argon Oil (Light Weight Oil), if it does not distort your herbs or extracts. In this case, you have to extract using oil and ethanol separately because oil will not dissolve with ethanol and they both have different boiling point.

      This method is not applicable for liquid sample. If your sample is milk or milk product then follow this video: czcams.com/video/muswKsqvGnM/video.html.
