    Hello Miss Tatum Sweet, I normally don't leave comments on CZcams videos. However I have watched many videos about Tissue Culture, and I have found your's to be the most useful and practical. I cannot help myself from saying thank you for your videos. The time spent in composing your TC videos was not in vain. You have completely persuaded me to get involved with TC. No other CZcams videos or CZcamsrs have ever done that. They have all made it seems very high and lofty, something the the average DYI type of person would not dare try. However you have succeed in persuading me that I can do it by myself. Thanks again! I cannot fully express the level of gratitude I have for your work. I have three questions.
    1)- If not moldy, Can the media be reused with different types of plant? And if yes, for how many times?
    2)- What is the difference between the multiplication media and the rooting media?
    3)- Is it toxic to dispose of the medias in the environment?

    love the videos! i’m just wondering do you ever tc nodes or pieces from existing/mature plants that weren’t plantlets from someone else previously tissue culturing them? if so, i’d love to see that as well!

    Great video! Just one question. When you’re separating them deciding what to keep in multiplication media vs which ones to plant, what do you look for? Do you try to keep a root on each piece that you are transferring?

    • @TatumSweet
      @TatumSweet  Před 3 lety +3

      If they have roots, it certainly helps to try and keep those, but roots aren’t necessary! I just look for plantlets that are about 1/2” or bigger.

    I love your videos! Thank you! Have you tried using rubbing alcohol or hydrogen peroxide instead of bleach? I know a lot of medical facilities rely on rubbing alcohol and hydrogen peroxide for disinfecting. I wonder if it would be an acceptable alternative to bleach.

    • @kend.3195
      @kend.3195 Před 2 lety +2

      I use alcohol for disinfecting and it's effective, but the fumes are pretty bad. I haven't tried bleach yet.

    Thanks so much for an in depth video! I have a couple questions...
    How do you know when the explant is mature enough to divide? How do you know when they're mature enough to pot up instead?
    Also a separate question...is the initiation phase necessary before moving to multiplication media, or is it fine to jump right to multiplication after you sterilize the explant?
    Thanks so much! Looking forward to the next video :)

    • @TatumSweet
      @TatumSweet  Před 3 lety +6

      These are great questions! I wish I would have been able to include the portion of the video that got deleted because I showed the plants up close to where you could see the size. I’ll be sure to get those in a future video though! I extract plantlets that are roughly 1/2” in size.
      Honestly, I typically skip the initiation phase but most of the plants I work with were already in TC to begin with. If you are taking an explant from a mother plant, then it kind of depends. Some plants need initiation and some are fine without it, so it’s kind of just trial and error for every plant!
      Thank you for following and I’m having so much fun creating this series❤️

    When you say, "10% bleach", do you mean 10% NaClO solution? or a 1:10 dilution of (fresh) grocery store bleach (which in my country is 3.5% NaClO, I believe a little more in yours)?

    • @TatumSweet

      1:10 dilution! I think mine is 7.5%
