The Colony Formation Assay (CFA) or Clonogenic Assay is often used to determine which effects a specific drug or irradiation has on cells. This method is frequently used in cancer biology to investigate whether a treatment might be beneficial for tumor cells that harbour a distinct mutation.

Would cells attach if they grow in suspension? I'm concerned most of my cells will die given the length of incubation time.
It depends on the cell line, check your cell line and the keyword clonogenic assay in the internet.. for many cell lines you would need to exchange medium in between more often!
