Between Arkham City and Arkham Knight, Batman destroyed the tainted blood at the hospital but four people were infected by it. In Arkham Knight, Batman gathered up the four infected and determined that eventually the poisoned blood would drive them all insane and make everyone infected think like Joker. Batman was still infected from Arkham City, so he is considered the '5th' person who could become the new Joker. Each infected person believed they would be the best Joker and wanted to kill all the other infected - that's why the man with the gun shot all the other people. But when he sees that Batman is infected, he believes Batman would become the greatest Joker of all ('you're going to be... spectacular') so he destroyed himself.

The thing about this that is truly scary is that despite how strong Batman is, he is still fundamentally holding back. But Joker doesn't have those limitations. And if the Batman can offer him anything, it is his almost superhuman strength and arsenal. And if anything is to be considered it is the fact that their union would create something that would be utterly monstrous to Gotham, a perfect Joker.
After replaying the game a dozen times it did start to annoy me that Batman never even speaks to him until the end. It would have sold him losing his grip a lot better.
On the ending of the game you get a glimpse of that, joker pretty much kills every single one of the major villains, the commissioner and probably is coming for Alfred while Gotham burns to the ground, he also gets a jokermobile
Only in the challenge mode. And rather than fighting other crime bosses, you fight other heroes and bats instead. What's the point in being the bad guy if you're going to fight other bad guys?
The Crime bosses have always fought eachother though. Sometimes they team up but other times they have their own internal war going on as well as trying to kill Bat-Man

After putting my mind to it I just realized all these Joker hallucinations started way back in Arkham City. Remember the Mr. Freeze boss fight when you kept punching Freeze in the face for a very brief moment you could see Joker's head laughing instead of Freeze's. It all makes sense since he was injected with Joker's infected blood back then. The connection between the games is just magnificent.

When Joker takes over we see how easily Batman could take out he’s enemies if he weren’t holding back. Even the mercenaries become afraid and plead him to stay away. I hope we get to see an animated adaptation of The Batman Who Laughs some day

Maybe Batman Who Laughs was inspired by this game plot from Batman: Arkham Knight? I hope in Suicide Squad: Kill The Justice League if Rocksteady will add Batman Who Laughs as a secret boss battle.

After this game (Batman: Arkham Knight), there is a hybrid between Batman and Joker was called Batman Who Laughs which came out on 2017 just 2 years after Batman: Arkham Knight was released. Maybe Batman Who Laughs was inspired by this game plot from Batman: Arkham Knight? I hope in Suicide Squad: Kill The Justice League if Rocksteady will add Batman Who Laughs as a secret boss battle.
