The conversation of Autoclave during the morning coffee between my boyfriend and I birthed the ‘could we dyi autoclave’ question 😊 -
Thank you for your great video.
You are so good at explaining your process. Keep this up. You're killing it and you look so comfortable on cam. Great, great video
Thank you so much! That is such an amazing compliment :)
I don't like the instant pot for autoclaving. It only goes to 12PSI, and they are excruciatingly tiny.
Electric stove is fine, just use "medium high." The warning in the manual refers to the propane "jet burners" as opposed to household cooktops.
My favorite is the Presto "induction compatible" model, gets to pressure super fast on a 3500W induction burner.
I've never found any benefit to venting off steam for ten minutes. I believe that tidbit in the manual (and from the USDA apparently) is based on flawed folk lore. With or without venting, I read identical temperatures at 15PSI.
Just my two cents from the field ;)
I originally purchased my current multicooker to use as an autoclave like 11 years ago. It was used to do some citizen science with animal cell cultures, which require significantly more sterile conditions than plants do. I didn't even have a laminar flow hood or a CO2 capable incubator and still got virtually no contamination.
I currently cultivate mushrooms and am fascinated by PTC. I had heard of it before but never figured to look it up until I saw one of your videos. Just wanted to add a quick note about pressure cookers; I have the same one but made for induction heating and I use it on a standalone electric stove. I start it at 1600w and wait until the safety plug (also check if the two halves are properly screwed together before running the cooker) pops up and closes on it's own, by that time you will have a steady stream of steam coming from the rocker spout, then I place the rocker on top and monitor it until the pressure builds up to 15psi. Then I lower the power down to 1200w and leave it there for however long I am running the cooker. No further adjustments needed. (on my specific stove, your mileage may vary) Always remember to keep an eye on it though as you stated. Your videos are super clear and nice to follow :)
This is awesome. I'm binging all your videos. Thanks for putting all of this info up!!
thank you!!
Thank you for the tutorials, I am surprised at everything that can be done in vitro. And very interested. great job. Greetings from Mallorca.
Thanks, been needing to get instruments done.
keep it up! i love your videos
Thanks for the tip
I'm Going to try this on Thanksgiving cactus.
Does this sterilise through the aluminium foil to inside?
What if the thing that you need to autoclave is a solution/liquid. How will that work?
Great video!! I am interested in your growing bananas protocol video please. How do I get it? Thank you Blessings
I have a question my vessels have a filter batch on the lid do u think I should cover them with foil to autoclave so filter patch don’t get wet ?
Mam, as you said not to open the lid even after the gauge hits zero... So , when should we open it mam...
Can i sterilize food using this autoclave? is that possible?
I literally have no use for this information but great video
An electric range is perfectly fine to use with the pressure cooker
Could I potentially use water bathing like you do for canning instead of pressure cooking? Or would that not have the same effect?
No. Water bath canning does not eliminate all pathogens.
Really good explanation! As someone who is interested into getting in plant tissue culture just for the fun of learning, may I ask, what container materials would you suggest kind of to compare the pros and cons. I know glass is 100% autoclavable but some people use plastic containers (I guess it would be cheaper) and I'm wondering what type of polymers are they made from? Also, can you use tight-seal mason glass jars in autoclaving or would the rubber melt?
Hi! You can use deli containers instead of glass jars if both the plastic container and the plastic lid are autoclavable (aka can withstand 121 degrees Celcius). I have never used a mason jar for tissue culture, but if you buy mason jars that are designed for pressure canning, they should be able to withstand the pressure cooker. If you're going to try tissue culture with mason jars, I would probably opt for mason jars that don't contain a rubber seal, like Ball mason jars
@@plantsinjars thanks a lot!
@@plantsinjarsHi, would this autoclaving method sufficiently disinfect human hair to be safely used for stiching? Outer scars rather than bloodvessels.
Thank you.
Great channel. I have always been interested in tissue culture, but all the resources seem "lab grade" Makes me think I might be able to do this
Thanks! I have a video coming out in the next few months about how to get started with tissue culture for under $200 using a still air box and deli containers that you might be interested in
@@plantsinjars That would be great!
1:54 Yeah only the Europians use celcius, right. It's not that only Americans using fahrenheit instead celcius in the whole world.
Btw really nice video thanks.
So much natural beauty and charisma. Don’t waste it all here discussing autoclaves and plants!
Ok boomer.
Lost me. I don't get the reference. @@js12352

youre not being sarcastic are you?
