    In this video i demonstrate how to make a liquid culture of any mushroom species using Karo light corn syrup. This method is bare bones intro to liquid culture. Showing the most basic entry to grow mushrooms.
    EDIT:
    Use PAPER micropore tape, not plastic micro pore tape. You may use a steam canner (2 hours) to sterilize your culture media instead of a Pressure cooker. using a pressure cooker sometimes will caramelize the sugars (bad) due to the high temp.

      @@DAAI741 drill a separate hole for either poly fill (have to be careful not to get that wet) or look up hydrophobic syringe filters and google that with mushroom and you should find the size you need and videos on how to drill and attach to canning jars

    Thanx for this affordable L.C. video. Thank you for not including expensive ingredients. It's demoralizing to me when the video lecturers tell me to use costly ingedients or tools, like a Nasa space laser thermometer, or a calibrated P.H. meter that costs $200.00. You did great here.

My questions: 1) Where/under what temperature and light conditions should I store the jars? 2) For how long can the jars be stored before going bad? 3) Can you use liquid culture (mycelium) from a syringe as well as the spores within a syringe?       Thanks for watching! Store colonizing jars at 70-80 degrees light doesn’t matter but darkness is preferable , once full they will last for 6 months at room temp and can and should be refrigerated to force the mycelium to go dormant for longer than a year.

      @@jeff042854 Liquid culture is just spores that have been germinated by introduction to sterile nutrient media. Spore syringes are made with only water to prevent them from germinating, they can be store for years before use.

      @@jeff042854 Also a spore syringe contains many different strains of the species. A liquid culture syringe is one strain. So the genetic differences will be wide with spore syringe.

      @@SporenSprout I did broke boi Tek about 2 weeks ago. I have vermiculite ready for the monotub with all the alcohol to get it ready.
      I took 11 cc's and divided them between six bags. Some of the bags look like they're about done and some look like they're about halfway done. How do I know when to move the cake to the mono tub? Should I do each one as it's ready, or do I wait till they are all done? I'm going to be watching your videos because this is the first one that I've seen, but if you can answer my question I would be very much appreciative for it. They are already talking to me, I'm excited!

    NOTE: If there is more than 4% sugar to water ratio the liquid can be come toxic to the mycelium/spores. It's better to use less than more if you decide to guesstimate.

      @@contambrah If the maximum is answered above, isn't the minimum, by default understood to be ZERO - but with no food = no growth. Anything greater than ZERO will allow for reproduction.

      Also keep in mind that with less food available you will also be triggering rhizomorphic growth.

      Its best to use liquid culture as spores take up to a week or so to turn into mycelium and spores are also usually contaminated so its best to do a brf tek with the spores, get agar, take a piece of clean mycelium from either the middle of a cake or the middle of a mushroom or something and place it into the agar, once it turns to mycelium you can use that to create your own liquid culture, you have a much greater risk of contam and contam is very hard to see usually in liquid

    I will say that my current jar of liquid culture was made with no magnetic stirrer and no marble and the entire thing is evenly cloudy. I just spun the jar with my wrist at least 1x per day for the past 12 days.

      Where do you store the jars when doing this process for 2-3 weeks? In a cool room with indirect sunlight? Then fridge to store long-term?

      Yes! Best thing iv ever bought for this hobby! I think it was like $30 on amazon. As others have commented the stirrer actually brings fresh oxygen into the mixture and speeds healthy growth!

    Q1. Can sugar syrup made out of crystallized sugar be used instead of corn syrup?

      Yes you can use any sugar source you have available. You can just put a glass marble inside the jar instead of magnet.

    Super straightforward no fuss method. Thanks! Some newbie questions:
    1. How do you know when the mycelium has grown fully in the jar and ready for use/storage?
    2. Can you just throw them in the fridge after for storage? If not, what's the shelf life like before they go bad (and how do you know they've gone bad?)
    3. How does the Karo light compare with other mixes with malt extract / dextrose?

      1. It is not definitive, but most aggressive species will completely fill the liquid culture jar with mycelium. Once the jar is full of thick mycelium it should be stored in a cold room or in refrigerator. Some species like lions mane and shiitake will usually only grow half way in the liquid culture jar and are not as aggressive.
      2. Storage at room temp is fine for up to 6 months, refrigerated or cold room shelf life is over 12 months. It’s hard to tell when it has gone bad, but a way to test it is to put a drop on agar plates. If it grows it is still viable. If the culture is very old it will be noticeably slower or yields will be smaller and smaller over time.
      3. Honey is way better than karo in my experience. Karo produces more thin, weaker mycelium and honey produces very thick, aggressive mycelium. It is good to switch between honey and LME once every 1-2 years to change up food source.

    If you invert your lid and put it in a hot oven it will melt stuck to the ring. That makes it easy to remove when you inject spore solution.

      If your talking about making your own spore syringe then no it's not the same. You will need a spore print, and you would not add the sugar to the water. There's videos here on CZcams that you can follow.

      "NEED to buy, all the 'special stuff'- affiliated w/their 'operations'."
      There are MANY, many diff ways to go about "growing your OWN mushrooms"- whether for medicinal/food/fun purposes.
      A good place to start is the Reddit group! "r/mycology"- I believe its called.

    Can you do a video showing how you would extract it into a syringe that you could sell or give to a friend?

      Yes I can do that. I’ll start working on it ASAP. In the mean time, quick sneak preview would be to tilt the jar of colonized liquid culture and inject empty syringe and suck up the culture that way. The only problem with that is, if your mycelium isn’t very dense and full in the jar it will be hard to reach. If that happens, try using LME recipe l (I will be making a vid on that soon too). Or you can super sanitize your work area and also use a UV-C light to sterilize everything and then simply open the jar lid and suck the culture up and close the lid right after. Hope this helps.

      @@SporenSprout yes this helps but I’m also trying to figure out what syringes you purchase for this exactly like you order the syringes, extract the liquid culture and the replace the syringe with a stopper under a flow hood and then send a sterile packaged syringe to a buyer along with the stoppered liquid culture correct? Also when is your LC considered colonized enough to start extracting it? I have a weeks worth of lions mane LC that I stir everyday. When can I start extracting it? Should I wait a few more weeks of mycelium accumulation? And also how do I prevent it from forming hyphal knots?

      @@HoneyandMyco order empty sterile 10 ml syringes from Medvet international (they come with needle attached) and a separate order of 18 gauge sterile needles, and syringe caps. Open the syringe package near flow hood or in SAB and suck the culture up, take needle off and replace with syringe cap. Now the syringe is ready to be labeled and packaged along with one sterile needle. That is all the friend or customer would need. One capped syringe of culture along with one sterile packaged needle. There is no set maturity to the Liquid Cultures. As soon as there is enough to fill the syringe with a decent amount it is good to go. Usually takes me about 1-2 weeks for adequate growth . Use a magnetic stirrer to insure they mycelium can be broken up and distributed for faster growth and also fresh air sucked in my vortex it creates.

    Hi thanks for the video. I couldn't find my question in all the other comments, here it is: when storing the jars in the fridge do you put the jars in something like a sealable plastic bag to protect them from any nasties in the fridge, or do you just put the jars in the fridge as they are?       In a perfect world, you would have a dedicated mini fridge just for culture but to answer your question, I don’t use a plastic bag and I don’t know that anyone else does. If the liquid culture is super thick and full like it’s supposed to be, there is less room for contam also.

    Hi my friend I was just watching you on CZcams and I will say I was very impressed by your knowledge and willing to share with us. My son has been growing mushrooms for me to eat because of the research he has been doing about the health benefits of a few different types.... I have a question. We are in the process of making the jar myzilyume. We are in Utah so it's extremely cold. For two to three months his been trying to get some to grow. My question is how long will the myzilyume last in the jar before it's not good to grow anything. We want to be ready for when the weather changes and it's warm enough to grow them. We were trying for a long time and finally I told him to go ahead and keep the room really warm and let's see what happens. Two weeks later we were enjoying some kind mushrooms lol thank you

    Hi thanks for this video! You said we could use a marble too. Does that mean we need the stirrer machine or would we just shake the jar with the marble in it?

      You wouldn’t need the stirring machine if your using a marble. Just tilting the jar back and forth is good enough for the marble to break up the mycelium that will clump at the bottom.

      Got a stir machine for less than $30. Its worth it. Make sure to put the stir bar in before sterilizing, but sanitize the bars first. I think they were the source of contam the first time.

    I appreciate if you answer my questions?
    1. So after you do this how long do you need to let them sit and in what conditions until you can draw up the culture into new syringes?
    2. How long can you keep the culture syringes and do they need to be in the fridge?
    3. In order to grow fruit, will you then inject the same way you'd use spore syringes?

      1. 1-2 weeks growth in dark 75-80 degrees.
      2. 6 months room temp, much longer if refrigerated.
      3.Yes, same as spores

    Huge fan and customer. One thing... you are using gas heat and alot of people have electric. Gas heats up faster. What would be the time in the pc for electric?

      The time starts after it’s heated up to 15 psi so it would be the same time. I usually do 25-30 minutes at 15 psi.

    I have a wild idea and I'm looking for some mushroom fanatics to tell me if (and how) it can be done. I really want to have lots of fly agaric in my garden. I'm looking for a way to make this happen. Should I try it with the method of this video? And what kinds of substrate should I use? 🍄 they are soooo gorgeous and magical!       Amanita muscaria forms symbiotic relationships with many trees, including pine, oak, spruce, fir, birch, and cedar. You would have to plant the mycelium in a forest not in a garden.

    I have had two starter kits for over four months and they both would not sprout. They are both very well covered with mycelium

      After that long I would assume they are dried out. You should submerge the blocks in water over night and then set the hydrated blocks inside a shotgun fruiting chamber or mushroom grow tent to fruit.

      Rust is not harmful to the mycelium but It is very ugly, you can switch to plastic lids instead of metal.

    One question: after all that, what's the best way to "duplicate" the LC? Should I take a few CC from the jar and pour them in other LC jars? How many times could I do that?

      Yes you can put a few ml in more LC media or you can put a few drops on a bunch of agar plates to multiply that way. There is no definitive answer to your question, it can be done many times until it stops performing well from old age, some species degrade quicker like cordyceps.

    You will have mushrooms forever but you need to start your own generation(s)

    The reason why you magnetic stir is there is no oxygen in the sterile liquid culture so stirring right after inoculation introduces oxygen to the culture as well as breaking up the mycelium to encourage growth. Great video!

      @@georgekatsinis5224 I would assume that wake, water falls, cascading rivers all help to provide oxygen as well as aquatic plant species, occasional erosion from cliffs or banks calves and this would also introduce oxygen.

      @@SporenSprout I was skeptical about there being less oxygen in boiled water till I googled it and read up on tea related articles. It would make sense that when the magnetic stir bar is spinning the vortex it’s also pulling fresh air in the Jar introducing oxygen bubbles just like blowing throw a straw in a cup of water. I know when I spoke of it to other growers we all kinda had our hands in there air 😂 I’ve seen people not use any fresh air exchange and there LC was fine so adding the filter patch for fresh air exchange could benefit the final results. I know people who do use fresh air exchange for LC and people who don’t do fresh air exchange. It’s really up to the grower and what works best.

      Only if you are inoculating some wood chips outside in the forest or something. Look up the term “slurry” for growing mushrooms.

    So why do i need an agar plate?
    I could simply buy a LC syringe from the lab and make unlimitided amounts of LC. And if my LC contaminates i could rebuy a new syringe for 15$

    Most liquid cultures are made from mycelium inoculated liquids such as your water and syrup solution , but spores from mushrooms I believe take much longer to develop into growing mycelium in a culture if I’m correct or put onto Petri dishes to grow first , how effective is spores alone from the mushroom on its own being injected into a grain to grow I have not seen anyone do this .

    Amazing tutorial, man. I just tried this for the first time using my pressure canner and it ended up slightly yellow (caramelized) but not too much. Should i even bother inoculating it or just try again with a shorter cook time?

      I think you should start over. I prefer using a steam canner for 2.5 hours. But you can do 15 psi for 15 minutes in your pressure cooker and it should be ok

I think the two jars i overcooked cost about 14 cents in ingredients so its really not much of a loss. I appreciate the info and thanks again.

    Hey brother thanks again for the content. I have an extra LC jar of Cordyceps but I’m not ready to use it right now. I’ve been told cordy LC doesn’t store well but I was hoping to hear your thoughts on how I can make this LC last a few more weeks.

    In the end, aren't you just diluting the one syringe? Thereby potentially getting low results from your diluted batches or needing to spend more time building up the culture from one?
