There is this moment when Cleese says” Well it’s a joke name sir” and then gives an ever so slight nod of the head to just slightly emphasize the point... absolute genius acting

The back story to this scene is that the extras were told that whatever happens they must not laugh or they won’t be getting paid for that days filming. That’s why they were trying so hard to hold it in and the guys laugh at the end just makes this one of the best and funniest scenes in the entire film😂😂😂😂

It is indeed wordplay. Pilate is saying, normally, the following:
    "The little rascal has spirit!"
    "Has what sir?"
    "Spirit."
"Yes he did sir." < That there is the first joke. That's an inappropriate response to what Pilate said. He should have said "Yes he does." Therefore the audience gets the impression that Centurion isn't fully understanding him.

"No no... Spirit, uh, Bravado, a touch of derring-do."
"Oh. Uh, about eleven sir." < The second part of the joke, Once again, Centurion hasn't a clue what Pilate has said, or has heard something entirely different to what we, the audience have heard. Centurion's response should have been something like, "Oh I see, sir." or you know something like that.
Totally destroyed the joke by explaining it, apologies folks. =P
    Hope that helps anyway.

That is exactly one joke i don't get. As I'm German, I'm used to the german version and Pilate's speech impediment there is a different one. So I don't get the joke in this scene. Is it a kind of wordplay as well? Would be very nice if somebody could help me :)

It's kind of just 2 jokes in one. The impediment doesn't strictly require the Biggus Dickus bit, but it's just a roman related joke. So you could have two scenes, one with a roman emperor with a speech impediment saying things like "thwow him to the fwoow" which is quite funny, and another scene of an emperor talking about his friend biggus, and they'd both be funny but they're funnier together, I think.
As for word play its just the simple stuff you probably noticed:
      Biggus dickus - big d**k
Naughtius maximus- Naughty (and then I think maximus was a common name so its a play on maximum I guess)
      Incontinentia Buttox - Incontinence butt
      Sillius soddus - silly sod
So nothing more fancy than that. I think that's part of why it's so funny - it very barely makes sense to begin with, and only gets more silly as it goes on. :)
