Yes, it makes sense and it would dramatically change the whole idea of the comic and course of Batman's action. Important to note: Joker was not planned/drawn/imagined as a criminal, at least the character evolved into trully iconic character and I do not think that criminal is the first association that readers and viewers have in mind when they encounter him. There are few tiny details that are important, but I will not mention them, just not to spoil the comment section, and to drag the discussion with irrelevant speculations.
If you can do a particular what if scenario based on this idea I say go for it. The closest one that actually fits the kind of narrative is Joker’s greatest fear during the events of Arkham knight that he was afraid of being forgotten and that he actually never existed. Now one of the other ideas behind this what if could be that Harley Quinn could end up still be what she is albeit more closely akin to her counterpart in the telltale Batman series. Having individuals like Two face, Riddler and penguin formed their own criminal underworld empire will be an interesting twist and even having Catwoman as their personal thief they could get past a lot of defenses.
I think it would Batman would just phase out. Joker puts everything in such a buzz that everyone puts on a costume to fight him and he maintains the chaotic energies. Without him, Batman isn't always on edge, Robins arent on edge, Costumed Villains aren't on edge. A lot of criminals don't put on the costumes because there's no point. They can do their criminal activities without them. Except for bursts of Clayface and Man-Bat, the mob takes control. They recruit most of the Rogue's gallery, and Batman would actually be more effective against them by hiring out. A lot of the less conventional villains exist either because of the high chaos levels or they moved in for the challenge. You really think Ras and Talia Al Ghul would really care about a happier, retired vigilante that decided he can fight crime with coin? Batman already fights crime with coin, lots of it, but as smart as he is, you really don't think without the Joker driving him past the edge all the time he wouldn't choose the more pragmatic path? Without Joker, there is simply no Batman. Just a happy rich guy with a troubled past and a house full of children.
Question: Without The Joker, who would Batman's arch nemesis be? In my opinion, we have 3 canidates: Riddler, Two-Face or Scarecrow. Riddler challenges Batman's intelligence and has outwitted him on several occassions. Two-Face is arguably Batman's most personal enemy besides The Joker due to their past friendship as Bruce Wayne and Harvey Dentz along with Batman and Harvey's partnership on taking down Carmine Falcone. Scarecrow is also someone we must consider due to him being just as evil and cruel as The Joker.
Batman could've just a device in joker's body to shock him into unconsciousness everytime he was acting up. That would've saved a lot of trouble.

It's also possible that Duke Thomas will never become a vigilante, he became Signal because his parents were poisoned by Joker gas laughing.
