Sometimes, I get Lineweaver-Burk equation as y=ax-b. While 1/Km is possitive, 1/Vmax is negative. Is it problem? Why is it negative?
I found difficult when use this plot for bacteria fermentation kinetic, where the curve of 1/s and 1/u are not the same as the literature. the line of the curve is downward, Y = -ax + b, but the R square = 0,9 kind of. what if the result of this plot is downward curve, is it still can be use as a data to calculate Km?
