    This was great, to the point but with enough details to answer my questions. Do you know how expensive this diagnostic testing is? This is outside of my field but I'd assume the mRNA fluorescent probes would be more expensive than a primary+secondary antibody experiment? Thank you!

      @@animatedbiologywitharpan All due respect, but I will not invest money into something that is difficult to understand due to a language barrier. If you guys had subtitles/closed captions available on your videos, I would be more apt to offer financial support. There are sections that I cannot understand and have rewound to attempt to understand and still cannot; I haven't even finished the video because of it.

    My monkey brain "Fish", I don't understand fully: making fluorescent DNA, it was cuted with restrictases so I quess, it needed to be separeted through electrophoresis with big number of restrictases because these were leap endings of DNA. After isolation for what to put it in plasmid, no sense, we can already use polimerase RNA to make pre-mRNA or small mRNA. I understand hybrydyzation of RNA with RNA, I quess that we can also use retro-polymerase DNA to make single chain DNA from RNA wirh fluorescent nucleotydes and then we could somehow (how?) hybrydase it with chromosome, no we can hybrydase it during replication proces I quess.

    If DAPI is the counterstain used to stain the nucleus, which primary stain is used?
    Why do we need to use nuclear & cytoplasmic stains for detection and subcellular localisation of genes & mRNA in the nucleus and cytoplasm?

      both ISH and FISH are molecular biology techniques that involve hybridization of nucleic acid probes, FISH incorporates the additional use of fluorescent labels, allowing for visualization of the target sequences with fluorescence microscopy

    I have a question regarding dna sequence using FISH. "Is it possible to identify a particular gene sequence (for example - Amel Y) using FISH if there is lot of depurination (loss of adenine , guanine) in interested gene sequence ?"
