How to Use a Centrifuge

  • Think you know how to use a centrifuge? Sure it's pretty straight forward, but a mistake can easily be made, resulting in serious damage or even injury.
    So just remember these 4 points:
    1) Use the right tubes
    2) Balance the tubes
    3) Secure the rotor lid
    4) Check the settings

    @Amorquies When you centrifuge your sample, the pellet will form on the side of the tube that is facing outwards. So, if you place the hinge on the outside, your pellet will form on the hinge side. This makes it easier to pipette out the supernatant without disturbing the pellet, as your pipette tip can rest against the other side of the tube (without being blocked by the tube lid) as you remove the liquid.
    Try it out both ways and you'll see! =)

    @luciferin1 Not balancing the tubes puts extra strain on the spindle. Depending on how imbalanced the tubes are, and how fast the rotor is spinning, you can badly damage the centrifuge....and even other objects (and people) around it. So always balance your tubes!
    For many tabletop centrifuges, you can simply look at the tubes to see if they balanced. But for certain high speed centrifuges, you need to check their mass to make sure they are balanced.

    @8Avenger If you are worried that your protein may be coming out in the flow through, keep the flow through, and run a sample of it on an SDS-PAGE gel. If you see a band, then some of your protein leaked out through the membrane.
    Also, just make sure that your filter's MW cut off isn't too close to your protein's molecular weight or else you run the risk of your protein going through.

    @allroundgirl123 When referring to centrifuges, usually the term centrifugal force is used. However, it is not a "real" force. Centripetal force is the "real" force which acts inwards, while centrifugal force appears to act outwards. I'm not an expert in physics, so if you want more details about this, check out your inbox as I just sent you some links (I can't insert them here) - the links also provide some good examples to help understand the concept :)

    First thing you need to know is that it's not a way to wash clothes faster. It... doesn't work out well. And the tube slots aren't for laundry detergent, like I thought....

    How does a centrifuge motor stop? what is the force that stops the motor spinning?

      Michael Salsaa In my experience, when the centrifuge is unbalanced or improperly placed, the whole unit starts to shake violently. This can ultimately break the centrifuge.

    Thanks! Why do they make the centrifuges able to go too fast? For quick bursts of special tubes? In nuclear engineering labs a hacker slipped a thumb drive into an Iraqui computer control center and caused all their centrifuges to spin too fast and burn up the motors.

    No they don't. As Dan mentions at 2:05 if you only have one tube, fill the other tube with water to the same volume.

      Shafriena Farhana In most cases, centrifugation is used for the separation of particles from a solution. For example; blood specimens are centrifuged to obtain serum/plasma. The serum/plasma can then be further analyzed in chemistry, coagulation, therapeutics, genetics, or blood banking.

    Can anyone here educate me how to prepare prf( protein rich fibrin) to use in infected wounds as I m working at periphery n not much facilities r available here , we try to heal n help patients on our own within our resources...
    I ve got a centrifuge old machine China made 800 centrifuge wth rpm:4000.n rcf:1790×g.capacity 20cc×5.
    I also wanna know test tube's length its material n time to spin to retrieve max benefit from my produce.though I have general info frm watching diff u tube video but wanna get know frm experts as reading comments here I noticed that quite an experts in centrifuge technique are commenting here.
    I wd really appreciate if anyone is to extend his knowledge to where it's really needed n be used productively..
