Instructional video of how to use the Olympus Fluoview FV10i confocal laser scanning microscope to image Arabidopsis thaliana roots. The protocol involves staining roots with wheat germ agglutinin and calcofluor white for imaging root endophytic fungi. Roots are mounted and visualized with the microscope. Filmed at Michigan State University by Julian Liber, on equipment from Dr. Hideki Takahashi's research group. Produced, edited, and narrated by Julian Liber.

You're welcome! I don't work much with roots anymore, but propidium iodide staining can be done with the same method. The staining occurs within 10 minutes of submerging in 10 ug/mL PI.
