NEMATODE EXTRACTION: BAERMANN FUNNEL METHOD

This video demonstrates extraction of motile nematodes using Baermann funnel method. This extraction technique is based on the motility of nematodes. Sedentary nematodes, nematode cyst etc. can be extracted using other technique.

In my opinion, the green label should be applied to the funnel BEFORE it is placed in the stand. This risks releasing small parts of the soil into suspension by careless movement, making it more difficult to examine.

Why 24hours wait is must for the observation? Why not we extract water before this time?

it's shouldn't be 24 hours exactly, overnight is sufficient too ... we should wait for some hours to allow larvae to sink

leaving the set up for atleast 24 hours gives sufficient time to let most of the nematodes move to the water. It can be just overnight or even more than 24h!
