- This video demonstrates the basic steps involved in sterilizing an explant and culturing the explant for callus growth. Make sure you follow your protocol while doing the experiment and add the required nutrients in media according to the defined composition along with the phytohormones required for callus growth.
This video is just for online teaching purposes and hence, time required for explant washing and callus growth is not indicative.

Tissue paper is clean?is there any chance of contamination during this soaking water process
I think her cultures will have a tremendously high chance of contamination! She's making a lot of mistakes!
1. Not wearing gloves
2. Not flame sterilizing her forcep and scalpel
3. Using unsterile paper towels
4. Using HgCl before ethanol instead of doing ethanol first and then HgCl
5. Not measuring the time that explants stay in HgCl and Ethanol
@@mahyarshokraeian Agree with most of what you said, but gloves are not used for in vitro culture, it is very dangerous to work with gloves close to flame, take care !
After placing the media in patri plate what to do and how can I place the media in culture tube please guide me madam.
Yes, you can place the media in culture tubes. You can make slants to culture your explant. For placing the media in the culture tubes, you can simply pour the sterilized media and keep the tubes in slant position over a surface and leave for solidification.

You can try combinations and permutations of high cytokinins: auxin.
Try to subculture it. It could be due to the depletion of media.

You don't. There are a number of much safer alternatives that can be used for plant explant sterilization.
