  • Lesson on the Non-Canonical NF-kB Signaling pathway.
    The Noncanonical NFkB signaling pathway is primarily involved in B cell maturation, lymphoid organogenesis and immune system functioning. This signaling pathway differs from the canonical pathway in terms of cell receptors and cytosolic adaptor proteins. In a constitutional, inactivated state, cell levels of NIK are kept low due to proteasomal degradation. However, when this pathway becomes activated, NIK levels rise, leading to activation of IKKalpha and subsequent formation of an active protein dimer. This active protein dimer translocates into the nucleus and leads to the induction of non-canonical NF-kB signaling genes, which increase B cell survival. Unsurprisingly, mutations in this pathway can lead to the onset of specific immune cancers including Hodgkin’s lymphoma and diffuse large B cell lymphoma.

    Very interesting. So why would a drug like Aubagio, with NF-kB inhibiting properties, cause lymphoma?
