Zola said he got his terminal diagnosis in 1972, and supposedly the year that Tony and Steve travelled to was 1970, 2 years before Zola got his terminal diagnosis, so he should still be physically alive at this point, for 2 more years.
Could be that it was in development in 1970, and then two years after zola had his terminal diagnosis and volunteered to be the test subject. Possibly, probably.

I would say it wasn't, as Dr. Zola isn't physically found in the scene and it's clear Howard was looking for a physical person. If he wasn't, he would have been far more suspicious of Tony. Besides, Howard was still considered a threat to the deep cover Hydra agents within SHIELD, to the point they used the Winter Soldier to kill him decades later.
