My take for what it's worth (probably not much): Watch the footage again. Will laughed at the joke, which wasn't even that harsh compared to Rock's other material. Will laughed until he saw his wife's death stare. He didn't act out of anger or outrage, he acted out of fear of his wife's reaction. Not a healthy relationship.

I watched a video on the three people's body language (very interesting!) and slapping C R was W S's way of "defending" his wife. He laughed at the joke, then saw she looked uncomfortable and angry, then he slapped him. She's probably heard it all before, and it probably hurts her. Will has seen his wife get so emotionally hurt by the fact that she's losing her hair, becoming less of a woman, so he stood up for her via violence, which is wrong.
