Great video! I am actually the export manager for Yuantide Peptone and your video helps us decide to release the consumer version so thank you for that! Peptone is commonly used in fermentation industry acting as a source for protein. Based on our experience from that practice, you can also try to add yeast extract (not yeast) into the mix and see if there's any benefit. Peptone from different material sources can also make a difference (think of it like eating different types of meat), and our client often sees greater benefits from mixing both plant-based and animal-based peptone together (so soy peptone and bone/fish/casein peptone).
I was thinking about buying your product online. I was wondering do you know if yeast extract would be faster for mycelium? I see you have both products and was wondering.
@@andrewbarnett2761 Actually, our yeast extract product is still in transit, but it should be ready in the next few weeks. Meanwhile, you can purchase yeast extract from other companies. Yeast extract is essentially yeast without its outer cell layer, which leads to a higher concentration of nitrogen or protein. We haven't tried it on mycelium, so the main question would be whether the fungi prefer a lower concentration of sugar and a higher concentration of protein from yeast or not. Most bio-pharma plants here have chosen a combination of yeast extract and peptone (because the combination provides a better profile of amino acids), so I hope this provides you with some direction :)
Do all of the peptones smell like rot? I’ve used hiveg in my LC but it makes it cloudy and stinky.
Hi, in terms of smell yes. Peptone is basically hydrolyzed protein, and those smaller molecule tends to smell rot as you said. However, it shouldn't be cloudy at the concentration this video is using. I assume hiveg is a product by HiMedia so I can't vouch for their products, but our peptone don't typically appear cloudy at such low concentration. @@TheOffCycle

I read about peptone in Radical Mycology in one of the recipes, and have also found it to be AMAZING. I find adding peptone to Lion's Mane liquid culture is especially a game changer as Lion's Mane in my experience has a harder time growing in LC than other species. Thanks for the video! I will have to try out that laser tip.

Bro, You have hit the pandora box on LC. I did some Isolate and since using Peptone it has only taken 2 days (unheard of for me/lol) for the Mycelium to grow.. Thank you, thank you, thank you, you have made my life so much more complete!! Mush Luv 🍄💙

I just realized that you can use air pressure to dispense directly from your LC jars. Connect an air hose to the air exhaust port filter, and "run it in reverse" to push out the liquid...

Unfortunately, when you haven't been around long enough, it's hard to tell who to listen to and who not to. Adding motorbike sounds to your video doesn't make it less bullshit. Guy shows no proof, and makes bullshit claims about using a Lazer pointer to look for contams is the dead give away, the guy is living in his own privates confirmation bias lala land. Only way to safety test LC is on agar.

@@dracolusus Lol, never said it shows contamination. I said I use it to help look at the mycelium, if the LC is cloudy, that is usually a dead give away that it's contaminated.

So funny I just ordered peptone today before you posted this. Heard in a video about organic cordyceps growing that they would have used peptones but could not because of the organic status. I figured since I am not growing organic I could benefit from it. Plan on using in my multipurpose LC, and in nutrient mix/brown rice for cordyceps.
There are soy based peptones, too. In fact the NIH study found plant based peptone to be superior growth.

Idk for sure, just know it works, I guess it's just easier for mycelium to catch on to agar than just water, but the other good side to it is it makes pooling less of an issue when using LC in a grain jar

used LME and peptone in some LC and let it set for a few days after PC. when I went to inoculate I noticed a lot of stuff floating around. Is this normal? Doesn't look cloudy, but definitely a lot of stuff in there. I figured the nutrients would remain as sediment.

I bought peptone and have been using it for the last couple months. It's funny, you talked about myc forming a mat on the surface on the LC and I almost never had a problem with that just stirring the jars by hand. I started using peptone, and my jars became cloudy, started to look way different to the point I thought they had contams. Also started seeing myc grow vigorous mats on the surface - eventually I realized this was all because of the peptone. I have noticed way faster colonization times from the jars that have peptone and stir bars compared to without them. It's pretty insane.

@@JayJay-bd6ti I've been experimenting a lot.Traditionally I've used molasses, now I add a small amount of peptone to that. Visually, I add enough to darken the water but keep from making it cloudy. I feel like adding too much sugar can be bad but I don't know about peptone.

I know this video is old, but I really appreciate the tips! I just started collecting strains and making LCs. But I have a question, how do you dispose of your contaminated LC’s?? Do you toss the whole jar? Save the jar?
Without opening, run the contaminated jar through the pressure cooker to kill the contaminant, then you can open the jar and dispose of the contents and wash the jar normally. The whole culture is definitely lost though, if that's what you meant, barring re-isolating the sample using agar techniques.
Hi Andrew! Love your videos. I was reading to stay around 4% sugars, but see that with your premix it's a little higher. Have you run into any troubles with a sweeter mix?
I have to reset my cultures more often when running this rich, but that's it really.
You also have to be careful when autoclaving to high or too long, as you can caramelize the sugar. Then it's no good.

Man you know some stuff.
I've gotta say, you should be careful with which lasers you use to check LC!!! A lot of internet green lasers are insanely overpowered and often outright lie on the sticker. A $10 green laser could be easily be 80mW+ which are *powerful enough to vaporize your retina faster than you can blink* should an errant or reflected beam strike your eye in the wrong way.
Just make sure you use with extreme caution. Green lasers are actually made by shining IR laser diodes through a crystal that modifies the beam's wavelength. These are not efficient either, so even laser safety goggles rated for just green light could let IR right through, which is invisible but just as dangerous as visible light.
That being said if you are careful and only shine it directly into a body like in the video it gets dispersed rapidly. This reduces intensity of the light but is not guaranteed to be eye safe either.
What is the difference between mycological peptone and bacteriological peptone? Why do you use bacteriological?

Hey guys can someone help me out. I bought golden light dry malt extract and also peptone. So to keep it at 4% do I add 1/2 tsp of LME and 1/2 of peptone? Or do I keep 4% LME And then add an additional 1 gram of peptone? How does this work? Doesn’t matter I just don’t want to add to much nutrients to my pint size jar and make the mycelium lazy.
4% sugar to water... the peptone is additional and not factored in to the %
