    I just put a 2 year old blue oyster culture to agar a couple weeks ago when I was home and that sucker took right off

      I recently purchased a small 100 gram millet grain spawn of blue pearl oysters and put some to my LC mix, some to agar and some to chopped pasteurised straw in a bag. Very aggressive and fast, the LC jar is so full of spawn in only 5 days and everything else is colonising rapidly. I got black king oyster ( a king oyster/oyster mushroom cross) from the same seller on eBay Australia. Just as aggressive. It would be great if all our mycelium was this fast and contam hardy.

    I am no expert, however prior to your video, through lack of experience more than anything. I carefully removed just a few spawned grains form one of my jars with sterile tweezers and dumped them in a fresh LC mix. After 5 days lots of spawn was growing and I used a sterilized glass pipette to suck up the grains and remove them from the jar to prevent them going bad in the future. Thus far the mycelium is going great guns and multiplying. Just experimenting through trial and error.

      Thanks. I am trying to be as hygenic as i can. I made pretty good enclosed box witha clear top for light and visibility. I put 4 computer fans with a variable speed controller and a hepa filter with a good seal in front of the fans. I spray it all down with Isopropyl before I turn the fans on. It has good positive airflow out front opening of the box.

    Great video, thank you! You are showing the cans with visible mycelium clouds at the end. How long time did it take to grow? And is that the point when you put them into fridge?

      The incubation usually takes 2 - 3 weeks, sometimes a little longer. Once they look nice and chunky like that, I store them in a storage tote in the fridge. That goes for all cold tolerant species. Fridge temps will kill some tropical strains like pink oyster and almond agaricus. Those strains can just be stored at room temperature.

    About the enzyme blindness, I'm thinking of adding wheat starch to my LC, as some of my older jars are taking much line to start than the others. I use wheat grain almost exclusively, so why not add the wheat starch to my sugar base LC to keep those enzymes in play. I saved the water after draining some pasta and I'll give it a shot.

      Cool idea, let me know how it works out for you. Soy peptone is pretty amazing stuff to if you've never added it to your LC.

      I'm loving the soy peptone. My Lion's Mane LC went from mycelium flakes to mycelium which looks like cotton threads and REALLY hard to bust up with a stir bar. Quicker and stronger. @@RenegadeMushrooms

      You can definitely use them, but you may not be able to get away with just doing pasteurization. You may have to sterilize them. I would say if they are readily available to you, just start small and see how they work.

      Ah ok. I have a pressure cooker. Just done 3 jars in it. But I have bags for chippings etc. But would that replace the pellets in weight. Would have to experiment really. I have pellets also. Ready for bran Tek mix.

    Can injection needles and syringes be re-used by autoclaving? If so, any pointers for best practices? I'm trying to limit single-use supplies. Thanks so very much for your great videos!!

      Yes, just wash them out with soapy water and let dry. Then wrap them tightly in aluminum foil or put them in a canning jar and autoclave.

      Takes the soot off the needle and rapidly cools it so you can work faster. Other than that, no effect.

    Why do you wipe the needle after sterilizing it with flame? Wouldnt it be better not to touch it with anythiny once its sterilized ? Or is it just preference?

      It drives some people crazy that I do that, but it really doesn't matter. I've been doing it that way for 20 years and never get any contams in my LC. It's not necessary, but it rapidly cools and removes soot from the needle allowing me to work faster and keep my injection ports clean. Also, the injection port the needle goes through is wiped with alcohol, so it's really a wash.

    Not sure how good my LC mix will be long term. I just a add 1/4 tsp of powdered dextrose in 200ml boiled tap water. All my LCs and spores to this LC mix are growing very fast on this simple sugar water mix. I keep them warm around 25 degrees C, 77F until well established and refrigerate for longer storage. I tried light malt, but it seemed to contaminate and sometimes went cloudy.

      I try to never open my jars, unless I'm dropping in agar wedges in front of the flowhood. We've all fumbled through things at the beginning and there are a lot of ways to make LC. Just make sure to test your LC on a small jar of sterilized grain before doing any large batches. The catch with LC is contam molds can grow right along side your mycelium and you won't know until you test it. Cloudiness can indicate bacterial contamination, but powdered malt extracts don't always fully dissolve and can look cloudy even when they're good.

    Using that master jars do you prepare another jar of lc to inoculate grain spawn or you directly use master lc jars to make spawn?

    is the best way to get a culture sample to go on a hike looking for them?

      That is not the only way, but it is a really fun way to do it. Established cultures from reputable culture suppliers are well worth it if you're growing commercially because commercial grade cultures simply produce way better than 99% of random strains. Random strains are fun to play with though and you never know when you will discover the next awesome strain.

    I have a strange problem with LC
    (White oyster mushroom)
    When inoculted to agar ,it grows normally but when inoculated to sterilized grains it forms some dot like whitish structure but not mycelium. Same thing happened to one of the jar a year ago but it started to grow mycelium later and produced some good mushrooms. It happens to only some syring from a jar of lc .

      Sounds like you have some yeast mixed in with your mycelium. It's a common contaminant and forms white dots like you are describing.

    How long can culture live in the jar with the liquid? Is there a way to tell that it is starting to fail?

      Should easily last over a year depending on how aggressive the culture is. Always test on agar before doing a grain run for quality control 👌

      I refresh mine once a year, but strong cultures will live longer as mentioned. I've had some of my favorite cultures going for 12+ years using the yearly refresh method I show in the video.

      Not sure, I've never tried that. I have always used a pressure cooker. I believe there are some videos on CZcams showing you how to do it in a microwave, but I haven't tried that method yet myself.

      Why spend 4 dollars each on a piece of silicone, when a 3 cent piece of tinfoil works just fine? if you do a lot of jars you could be spending 100's on some caps that people have been using tinfoil forever on. They do look clean though, would be good esthetic for videos, however I think the mushroom community appreciates simplicity and budget solutions.

      I use the silicone lids now on all my grain jars in the PC, but then I pull them off for colonization. I still prefer the aluminum foil lids though for LC because I leave them on and I think they do a better job preventing any ambient airflow to the top of the jar during storage and handling. I see this as an advantage because more airflow to the lid increases the chance that a contam could make it through one of the microfissures on a well used injection port or just settle on the lid. The silicone lids have a small hole in the center to prevent them from popping off during the heat build in the PC, so they wouldn't serve that purpose as well.

      Jars that size don't need an air filter, but you don need to manually equalize the pressure in the jar after the PC run. I show that process in this video:

      @@RenegadeMushrooms Nice video, thank you! I think I will put a sterile syringe filter between the needle and the syringe. For the moment I've tried only to introduce sterile air into the growing mushrooms bags without laminar flow and no syringes. Maybe I will make a video showing that because I didn't find this idea on the internet. The air in the grow bags is useful for mixing the grain spawn into the substrate.
