  • Clozapine represents a breakthrough anti-psychotic medicine. Clozapine was introduced to the United States market in 1990 and is on the World Health Organization of essential medicines. Not only does Clozapine treat schizophrenia and related conditions, but it appears more effective than the standard anti-psychotic therapies. Clozapine also improves resistant schizophrenia and may additionally reduce the risk of suicide in these individuals.
    Due to safety concerns that originated in Finland, the use of Clozapine has decreased. Current practitioners lack familiarity with this drug, and as a result of some negative publicity seem disinclined to prescribe it.
    The two most worrisome issues involve a decline, sometimes serious, in the number of bacteria fighting neutrophils - a type of white blood cell. This occurs in about 3% of exposed individuals most commonly in the first 4 months of therapy. During this phase of Clozapine therapy, most recommend weekly laboratory tests.
    Perhaps even more troublesome is a slowing of movement of muscular activity of gastrointestinal tract. This may lead to constipation, fecal impaction and bowel obstruction. Pre-emptive therapy with laxatives minimizes this issue. Other adverse drug reactions may occur.
    In spite of the side effect profile, a recent study ranked Clozapine as the most effective drug in its class. As a generic, the financial burden it poses is minimal and more akin to the generic versions of Abilify, Zyprexa and Risperdal.

    Dont take clozapine for more than a year. You will experience severe withdrawal problems, in other words, coming off of this pill is almost impossible. I experienced Akathisia for 3 weeks, wanted to die every single second of it, and now im back on it again and there is no way to go back to normal for me. Antipsychotics are like a life-sentence in prison, except the fact that its your mind. God bless anyone struggling with mental health

      I've repeatedly gone on and off APs as add-on treatments for MDD and never had much of an issue.
      Maybe you're a fast metabolizer and hence Clozapine has a very short biological half life in your body (it's reported to be ranging from 4hrs to 26hrs depending on the patient) - this would explain the difficulties getting off of it, as your plasma levels would go down way too quickly, leading to intense and nasty withdrawl symptoms.
      When reducing the intake by 25-50% every two-three weeks isn't doable for you, you could switch to another AP, preferrably a long acting 2nd gen one that's metabolized via different enzymes than Clozapine (i.e. Olanzapine (30-51hrs), Risperidone(20hrs)) and then taper that one.

      I was on 600mg a day and I can tell you that I only had one side-effect but that it was horrible - Akathisia. I would pace around all day, couldn't sit still had this internal restlessness etc.

    Anyone notice this guy's continual sneering left hand side upper lip? Very disdainful, condescending attitude. And BTW clozapine is a horrible drug, severe life threatening side effects. Yes effective if being zombified is what you desire. See Alternative to Meds Centre for advice on alternatives.
