You are so good at making science understandable and attractive. Once upon a time, this was also my business - molecular immunology. But I've always dreamed of cultivating plant tissues. I will definitely try, especially since I now have a greenhouse of tropical species, some rare, and it will be a great experience. Besides, let me tell you that you are a very beautiful, funny, real little witch, only kind.
We use filtered containers and have great results. There is no build up of gas. It's the same filters Francisco uses on the bio couplers.

You are getting good at this video thing, very entertaining.
Still can't get any live tissue to be sterile. Done everything to the T in all the videos, but everytime it's instantly contaminated. I premake the vials, so the media/container is sterile but the plant tissue always has something on it rotting or something from the cleaning that it brings in.

I have seen your videos also how you mentioned before about callusing from leaf explant.
I'm working on a papaya plant and using it's immature and mature leaves for callusing. I have used all reported PGRs on different concentrations but no PGR is helping in callus induction. Its been 4 weeks and still there is no callus.
What could be the reason and how to induce callus in this situation? 😢

Potatoes are totally possible to do from tc! A lot of the early research in tc was done with potatoes (as well as things like carrots and tobacco..) because they respond well and grow fast in vitro

Depends on whatever protocol you’re following
@thatboyzerk1680 I've spent hours trying to find a protocol for a particular species, with no success. I guess my next best option is to find a protocol for the same Genus and experiment based on that.

Hey! Love your content. I am brand new and will be mixing up my first batch of PTC for philodendrons today. I have a question about this video. at 3:19 you show that you are doing your final tissue transfer to new media so the plants can get bigger before you acclimate them to soil, but you didn't mention the final media recipe. was it different than the last 2? More of the rooting hormone maybe?
It was the same as the previous 2. Begonias grow easily in TC, sometimes even with no PGRs. For rooting philodendrons I'd just use 0.5mg/L BAP and 1.0mg/L IBA
@plantsinjars ahh OK I see. And for the vegetative stage (by that I mean the first culture) for philodendrons would it be 1 ml BAP, and 1 ml NAA? (I don't have IBA)
@@buckbeaksgarden761 1mg/L BAP, 0.5mg/L IBA. NAA doesn't work well for philodendrons bc it causes a lot of callus growth, which is not what you're looking for

Amazon IS slave labour! And they sell the exact same thing as Amazon and everyone else. One is no better than the other, if you want to take a moral stance, only buy LOCALLY MADE.

How did u cut the begonia on the paper towel? How did you get it sterile.
wrapped paper towels in aluminum foil and pressure cooked it first
I really want to pick up this skill but I have never had indoor plants so it's highly likely that I'd murder an army plants for sure plus I don't even know if I'd able to maintain a sterile space to do this

Have you ever just kept your plants fed with worm castings. Then… Just pulled off a part of the plant and just stick it into a small jar of worm castings? I like how you science the shit out of it. I bet your plants wouldn’t be allowed to hang out with my plants. My plants would be too dirty, unvaccinated and well hardy and wild.
