    Hi Gary, really love the technical content, please keep up the great work! I have always grown from multispore inoculation and was wondering if monokaryonic mycelium looks the same as dikaryonic mycelium? Is there a way to tell (other than trying to fruit it) if a culture is a mated pair?

      genetic testing can help but yes it looks the same except it will stall out relatively quickly if expanding onto more media. Best way is to fruit

    Hi Gary, thank you so mush for sharing these videos. Your mushroom knowledge has been an amazing source of education for me in my pursuit to cultivate mushrooms. I am wondering if it is possible to breed two strains using complete (not from spore-just two clean and colonized agar wedsges) mycelial cultures, rather than by using 2 haploid cultures from spores to create a diploid? Just curious, as I am inspired to breed some of my strains after seeing your videos, but haven't grown from spore yet. Thanks again for your time. Love what you do man.

      Thank you again Gary. I admit that I watched Di-Mon mating video last week and didn't pay enough attention on the first run through.
      Just watched again and totally got the answer I was looking for. Just found a couple papers on Di-Mon mating with P. ostreatus and P. eryngii, so I'm excited to learn more and give it a try in the future. Your sharing of knowledge has been crucial in my mushroom cultivation education, so just had to let you know how much it is appreciated. Mush Love

      it’s from Air Science Global in Florida - they dont make this model anymore but there is one on my amazon link that is similar. They have a larger one now for around 4k shipped.

    Just to be clear..you don't have to do what your doing right now to get fruiting bodies correct? Those single isolates that have germinated the plates could go to spawn? I am excited about what your doing now dont get me wrong ☺️🍄❤️

      it’s assumed these ones were single spores - go back and watch this video - these are from single colonies from a dilution - some may in fact be mated already but this explains why czcams.com/video/1oqchhQbxBA/video.html (12:30 into the video)

    Why are you taking from the outer edge of the growth? (VS middle..)

      the middle can hide potential contamination so I am taking from the leading edge - I am 99% positive they are clean but this is best practice (there are some exceptions like cordyceps and nameko which can spontaneously mutate at the edges of the colony)

    Can you cross a pink oyster with a blue oyster? Is that possible

      I believe anything is possible and worth trying - it may require more genetic manipulation then just isolating haploids and crossing but they are both pleurotus sp.

    Gary didn't you guesstimate your original spore solution in your last videos? Did you verify with a microscope you had single spore isolates?

      no it’s not guaranteed you can’t really verify with a microscope anyway it’s pretty subjective. The best way is to grow them out

    Can you cross 2 isolates (from sectoring to a monoculture and cross those 2 monocultures)?

      Im not sure what you mean by sectoring but if you take 2 monocultures and pair them yes - but if you mean that you take sectors from an area where spore germination was heavy, then selected different regions that emerge there is a high probability that they are already mated and would not be compatible with another monoculture.

      @@FreshfromtheFarmFungi sorry, yeah I meant taking sections of rhizomorphic growth until you get to a monoculture and then cross that with another monoculture. I don’t think they would be a haploid tho
