  • Inoculation of 1.5% nutrient agar @ 2% gelling strength with a multispore solution using a disposable cell spreader.
    2 plates germinated while the others remained blank.
    When using a SAB or laminar flow, aseptic technique is extremely important, this is an example of that reality.
    Still air and laminar flow are advancements in the application of aseptic techniques that add layers of assurance, operating without them is possible but less reliable at scale due to variability in environmental factors that will affect the probability of success in an open environment.

    Very cool video. Thank you! I've never used one of the agar punches. Any particular type you might recommend?

      I made mine out of a stainless steel drinking straw. I made the handle by spraying the long end with plastidip, multiple coats makes a very nice handle.

      @StipeCap That's awesome. I'm definitely going to steal the label maker idea for my plates and tubs. My handwriting makes it look like a 5 year old is trying to grow mushrooms in my house.

    Like Mike458_ said love the label Maker and the transparent tape, can you give us a link for that?

      I used a label printer called D110 (Nimbot), it can be found on Amazon. Be sure to purchase the clear tape as well, it comes separately. 👌

    I'm still a beginner, If anyone has time to answer a question I would appreciate it: What are the main purposes of creating agar colonies? How long can an agar colony last? thank you

    • @StipeCap
      @StipeCap  Před 4 měsíci +2

      It's the fungal equivalent to sourdough starter, it's used to expand the colony into larger substrates or to store for in the fridge for later expansion.
      Plated cultures will last up to a year in the fridge before requiring fresh transfer to a clean plate to prevent drying out, other storage methods last for many years without requiring attention, if the storage temperature is stable.

    I've been wondering for a while, what percentage of the effectiveness of still air boxes is placebo?

    • @StipeCap
      @StipeCap  Před 4 měsíci +2

      Still air is not placebo, the SAB functions as described; however even when using a SAB or laminar flow, aseptic technique is still extremely important, this is an example of that reality.
      Still air and laminar flow are advancements in the application of aseptic techniques that add a layer of assurance, operating without them is possible but less reliable at scale due to variability in environmental factors that will affect the probability of success in an open environment.

    Hey stipe can you make agar to grain vids in an SAB? Like pleaaaaaaseeee

      I'll get some going in the near future, although I have other vids that already outline that technique.
