  • BAFF is a soluble factor that activates the noncanonical NF-κB pathway, which is a key arm of NF-κB signaling. BAFF provides essential survival signals during B cell maturation.
    The NF-κB pathway is a prototypical proinflammatory signaling pathway that plays a critical role in the development, survival, and activation of B lymphocytes. NF-κB is a family of transcription factors that bind the enhancer of the immunoglobulin κ light-chain gene.
    The noncanonical NF-κB pathway primarily targets the activation of the p52/RelB NF-κB complex. This pathway depends on the inducible processing of p100, which is both the precursor of p52 and a RelB-specific inhibitor.
    BAFF is a critical cytokine that supports the survival of mature naïve B cells. Recent studies show that BAFF and BAFFR are also required for the survival of memory B cells, autoimmune B cells, and malignant chronic lymphocytic leukemia (CLL) cells.

    I have a homozygous tnfrsf13b deficiency and my child has a heterozygous version -- this is over my head really - and I am just wondering if you could possibly do a video (explaining things in a way) about how having a deficiency of TACI can cause health problems .... I won't get into detail about what I have going on...but just might ask for you to do a video explaining the pathogenic nature of TACI variants...
