    thank you for the video , i usually never autoclave the media , specially not the selective. i test a small quantity for contamination and if it is okay i do not autoclave the , i just heat it for 10 muntes after the agar has totally melted and it does the job.

    Why did you put the hose inside the autoclave into Erlen?

      Thank you for your question. The hose is actually a temperature probe. As the video explains the probe can be placed directly into the media (which is in the Erlenmeyer flask aka “Erlen”). As the directions state: “Place the flask of boiling media into an autoclave to sterilize. If your autoclave is equipped with a temperature probe, the temperature probe can be placed directly into the media and the cycle parameters will be based on the readings of the probe.”

    Why does boiling for too long damage the media, but autoclaving at 120 degrees doesn't?

      You can boil it longer than one minute, we do it all the time with no issues. However bubble formation should be avoided

      You can also try misting the inside of your empty flask with 75% ethanol, see if this helps!
