Controlling crystals for more reliable drugs | Headline Science

  • Sometimes, compounds crystallize in unexpected ways. This is a problem for drug manufacturers, who need to create consistent products. Scientists are working to understand why substances take on different crystal forms, called polymorphs, under different conditions. Recently, they’ve shown that impurities can affect the relative stability of those forms.

    Crystal growth is black magic. I swear, moving a crucible 2cm to the left: different crystal. Recipe works with alumina crucible for me, but lab down the hall can only get it to work with platinum crucibles, using the same batch of chemicals.

      It’s probably down to very slight differences in humidity, ambient airflow, solvent quality, reagent quality, or any one/combination of hundreds of variables

    They get approval for one polymorph.
    Is that for patent reasons or does the drug work differently in a different shape?

      Mostly for patent reasons, but also the polymorphic form can affect properties like solubility.
      If you look up "disappearing polymorphs," you'll find examples where drug makers had to change their formulations when they kept getting the wrong crystal form.

    This is true. LOL. In the world of methamphetamine, you could tell when it was of lesser quality by the crystalline structure sometimes.
