Healing factors only work on injuries you get after having it activated but wade had cancer before his healing factor was active so they merged.
His body kept healing his cancer at the same speed it spred because his healing factor only remembers wade as having cancer be part of his dna

So while it isn’t really discussed in the first film apart from a throwaway line in the bar there is that thing about X-men origins wolverine Wade is fast and agile enough to deflect a whole bunch of bullets been shot at him and fast enough to turn and cut a bullet in half so it goes around him ending two guys behind him (they make a joke of this scene in deadpool 2 when fighting Cable) that’s not something a human can do so the film version had powers/skill before he got cancer and unlocked his healing factor.
There is also the question of if his 4th wall braking is part of his mutant powers or something else? In the film he dosnt talk to the camera until after surviving the fire so maybe it also unlocked that or been burned to death and suffocating to death but not actually been able to die broke his mind to the point that he could see higher reality’s?
