    (Twitter: @DrWhitneyHolden) The Acid-Fast stain is an important differential stain used to distinguish between Acid-Fast bacteria and Non-Acid-Fast Bacteria, based on the presence or absence of mycolic acid in their cell walls. In this video, Biology Professor will cover the Acid-Fast stain procedure and discuss how it is applied to determine the presence of Mycobacteria, which include the bacteria that cause tuberculosis and leprosy. Great for MCAT Biology Review! For information on the Gram Stain, another important differential stain, see Biology Professor's video: • Gram Stain Procedure

    leprae is a gram positive so how can you use acid-fast stain to detect it?

    • @BiologyProfessor
      @BiologyProfessor  Před 7 lety

      Based on its cell wall structure, Mycobacterium leprae is a gram-positive, rod-shaped bacteria with a thick waxy coating. Due to this coating with mycolic acid, it cannot be stained very well with the regular Gram stain procedure. That is why we use the acid fast stain on it instead.

    hello prof. if the bacteria is non acid fast then it is gram negative?

    • @BiologyProfessor
      @BiologyProfessor  Před 7 lety

      Not necessarily. You would need to actually do a Gram stain. Non acid fast bacteria can be Gram positive or Gram negative. The acid fast stain just shows that non acid fast bacteria don't have mycolic acids, but it cannot assign their Gram type.

  • @virtuality400

    What outcome (color) is expected for a Gram positive, non-acid fast bacterium like Staphylococcus aureus if alcohol is mistakenly used instead of acid alcohol?

    • @BiologyProfessor
      @BiologyProfessor  Před 10 lety +2

      Alcohol is not good enough to remove the carbolfuchsin dye from non-acid fast bacteria because carbolfuchsin isn't very soluble in it. So using alcohol instead of acid alcohol will not decolorize properly and more non-acid fast bacteria will appear red. Hope this helps!

    Why is ZN stain used instead of Gram stain in spite of the organism being a bacteria?

    • @BiologyProfessor
      @BiologyProfessor  Před 3 lety

      Acid-fast bacteria don’t stain well with Gram stain because of the waxy mycolic acids in their cell wall. So another stain is necessary.

    Why used acid fast staining for actinomycetes

    • @BiologyProfessor
      @BiologyProfessor  Před 2 lety

      To distinguish it from Nocardia. Actinomyces and Nocardia can be difficult to distinguish without staining. Acid-fast Nocardia can be distinguished from from non-acid-fast Actinomyces with an acid fast stain.

    • @BiologyProfessor
